    Features of Forming a Cubic Li7La3Zr2O12 Film by Tape Casting

    Thin films of Li7La3Zr2O12 solid electrolyte were obtained by tape casting.The optimal heat treatment mode without films’ deformation was proposed.The highest conductivity values were observed for the Li7La3Zr2O12 film annealed at 800 °С – 1.5·10–5 S·cm–1 at 215 °С.Received: 13.10.23. Revised: 07.11.23. Accepted: 17.11.23. Available online: 21.11.23.Currently, interest to lithium and lithium-ion all-solid-state power sources is rapidly growing all over the world. However, several issues should be addressed before all-solid-state batteries production: high resistance values of the solid electrolyte membrane and poor contact between electrolyte and electrode materials. The transition to thin-film technologies is one of the promising ways to solve these problems. Tape casting can be proposed to obtain thin-film solid electrolytes. In this research, the features of the structure formation, morphology and lithium-ion conductivity of Li7La3Zr2O12 films were investigated. Li7La3Zr2O12 films with the thickness of 35 μm were obtained by tape casting on Ni substrate. The influence of organic components’ content on homogeneous coatings formation was established. Heat treatment conditions for dried films were chosen based on differential scanning calorimetry and optical dilatometry. Phase change from tetragonal to cubic modification occurs after annealing the Li7La3Zr2O12 films at 700 °C and higher. The annealed Li7La3Zr2O12 films have developed surface, which can lead to improved contact between the solid electrolyte and an electrode in an electrochemical cell. Li7La3Zr2O12 films annealed at 800 °C have the highest lithium-ion conductivity values (2.5·10–7 and 1.5·10–5 S·cm–1 at 90 and 215 °С, respectively).     Wheat-barley hybridization – the last forty years

    Abstract Several useful alien gene transfers have been reported from related species into wheat (Triticum aestivum), but very few publications have dealt with the development of wheat/barley (Hordeum vulgare) introgression lines. An overview is given here of wheat 9 barley hybridization over the last forty years, including the development of wheat 9 barley hybrids, and of addition and translocation lines with various barley cultivars. A short summary is also given of the wheat 9 barley hybrids produced with other Hordeum species. The meiotic pairing behaviour of wheat 9 barley hybrids is presented, with special regard to the detection of wheat– barley homoeologous pairing using the molecular cytogenetic technique GISH. The effect of in vitro multiplication on the genome composition of intergeneric hybrids is discussed, and the production and characterization of the latest wheat/barley translocation lines are presented. An overview of the agronomical traits (b-glucan content, earliness, salt tolerance, sprouting resistance, etc.) of the newly developed introgression lines is given. The exploitation and possible use of wheat/barley introgression lines for the most up-to-date molecular genetic studies (transcriptome analysis, sequencing of flow-sorted chromosomes) are also discussed

    Особенности патологической анатомии легких при COVID-19

    The research aim is to study the morphological features of COVID-19 in the lungs of patients who died in Moscow from March 20 to June 6, 2020. Methods. Autopsy material of the lungs from 123 deceased (54 women, 69 men) with COVID-19 coronavirus infection (confirmed by PCR) was analyzed, the median age was 71 (30 – 94) years, and the duration of the disease was 14 (3 – 65) days. In all cases, the patient’s medical records and autopsy reports were analyzed. Macro- and microscopic changes in the lungs were evaluated in all the observations. Results. The pathology of the lungs in COVID-19 corresponds to various phases of diffuse alveolar damage (DAD). The exudative phase of DAD was detected in 54 (43.9%), the proliferative phase – in 21 (14.63%), and their combination – in 51 (41.46%) of the deceased. Histological features of different phases of DAD are described. Conclusion. An analysis of autopsy material revealed a mismatch between the duration of the course of the disease and the phase of diffuse alveolar damage. A significant portion of the dead found a combination of exudative and proliferative phases of the disease. Histological signs that indirectly indicate a violation of the coagulation system during COVID-19 are described.Целью статьи явилось изучение особенностей морфологических изменений в легких у умерших от COVID-19 в Москве за период 20.03.20–06.06.20. Материалы и методы. Проанализирован аутопсийный материал легких умерших от коронавирусной инфекции COVID-19 больных (n = 123: 54 женщины, 69 мужчин; средний возраст – 71 (30–94) год; продолжительность заболевания – 14 (3–65) суток), подтвержденной методом полимеразной цепной реакции. Проанализированы медицинские карты всех стационарных больных и все протоколы вскрытий. По данным всех наблюдений оценены макро- и микроскопические изменения в легких. Результаты. Патоморфологические изменения в легких соответствовали различным фазам диффузного альвеолярного повреждения (ДАП). Экссудативная фаза ДАП выявлена у 54 (43,9 %), пролиферативная – у 21 (14,63 %), их сочетание – у 51 (41,46 %) умершего. Описаны патогистологические особенности изменений в разные фазы заболевания. Заключение. При анализе аутопсийного материала установлено несоответствие между продолжительностью течения заболевания и фазой ДАП. У значительной части умерших обнаружено сочетание экссудативной и пролиферативной фазы заболевания. Описаны гистологические признаки, косвенно указывающие на нарушение системы коагуляции в течении COVID-19

    Supporting data and methods for the characterization of iron oxide nanoparticles conjugated with pH-(low)-insertion peptide, testing their cytotoxicity and analyses of biodistribution in SCID mice bearing MDA-MB231 tumor

    The method of Fe3O4 magnetic nanoparticle synthesis by co-precipitation, modification by 3-aminopropylsilane and conjugation with pH-(low)-insertion peptide (pHLIP) is reported. The characterization of nanoparticles by scanning electron microscopy, transmission electron microscopy, Fourier-transform infrared spectroscopy, elemental and thermogravimetric analyses as well as dynamic light scattering and z-potential measurements is provided. The effect of nanoparticles on the viability of mouse and human peripheral blood mononuclear cells is tested by flow cytometry. The experimental details of nanoparticle administration to tumor-bearing mice, magnetic resonance imaging scanning as well as subsequent tumor sample collection and their processing for transmission electron microscopy, inductively coupled plasma atomic emission spectroscopy, histological and immunohistochemical analyses are described. Biodistribution of the nanoparticles in mice and blood serum analysis data for experimental animals are given.     Оптимизация метода исследования маммосферообразования для оценки il-6 индуцированной стволовой пластичности дифференцированных клеток рака молочной железы

    The aim of the study was mammosphere assay optimization for quantifcation of IL6-induced stemness in differentiated (СD44– ) T47D breast cancer cells.Material and Methods. The effect of three commonly used cell-detaching methods (TrypLE, accutase, cell scrapper) at various confuence (40–50 % and 70–80 %) on cell viability, phenotypic profle and mammosphere formation was tested. The cell viability was examined using AnnexinV/propidium iodide assay. The phenotypic profle was analyzed by fow cytometry with fuorescent markers CD24 and CD44.Results. Detachment of the cells using scrapper led to substantial increase in early apoptotic and late apoptotic cells in comparison with TrypLE and accutase. Dissociation with TrypLE reduced the percentage of detected CD44+ positive cells, whereas accutase saved the surface marker. The number of mammosphere and their diameter did not differ between groups. Incubation of differentiated (CD44– CD24+) T47D cells with IL-6 for 24 hours resulted in an appearance of CD44+CD24+ and CD44+CD24–/low subpopulation. Furthermore, the differentiated cells after 24 hours of IL6 exposure formed 3 times more mammospheres compared to the control.Conclusion. Usage of cells with confuence of no more than 80 % and accutase for detachment of cells is recommended for mammosphere assay. Incubation of CD44– CD24+ T47D cells with IL6 for 24 hours is suffcient for stimulation of stemness plasticity. Цель исследования – оптимизация метода оценки образования маммосфер после индуцированной интерлейкином 6 (IL6) стволовой пластичности у дифференцированных (CD44– ) опухолевых клеток линии рака молочной железы человека T47D.Материал и методы. Проведена оценка жизнеспособности, цитофлуориметрического профиля клеток и их способности к маммосферообразованию при различной исходной конфлюентности (40–50 % и 70–80 %) и методах снятия с поверхности пластика (использование TrypLE, аккутазы, скребка для клеток). Жизнеспособность клеток оценивали методом проточной цитометрии с использованием флуоресцентно-меченного АннексинV и йодистого пропидия. Динамику изменения экспрессии поверхностных маркеров клеток оценивали методом проточной цитометрии с флуоресцентно-меченными антителами к CD24 и CD44.Результаты. Применение скребка для снятия клеток приводило к существенному повышению количества клеток с цитофлуориметрическими признаками раннего и позднего апоптоза по сравнению с использованием TrypLE и аккутазы. Использование TrypLE приводило к снижению количества CD44+ клеток, тогда как аккутаза не влияла на представленность данного маркера. Количество маммосфер и их средний диаметр не отличались между исследуемыми группами. Стимуляция дифференцированных (CD44– CD24+) клеток линии T47D IL6 уже через 24 ч приводила к появлению популяций CD44+CD24+ прогениторных и CD44+CD24–/low опухолевых стволовых клеток. Дифференцированные клетки после стимуляции IL6 в течение 24 ч образовывали в 3 раза больше маммосфер по сравнению с контролем.Заключение. Для оценки маммосферообразования целесообразно использовать клетки с конфлюентностью до 80 % и применять аккутазу для снятия с поверхности пластика. Для стимуляции стволовой пластичности у дифференцированных клеток линии T47D достаточной является их инкубация с IL6 в течение 24 ч.
