    HfB2-SiC-MoSi2 oxidation resistance coating fabricated through in-situ synthesis for SiC coated C/C composites

    A brand new HfB2-SiC-MoSi2 coating was fabricated to protect carbon/carbon (C/C) composites with inner SiC coating from oxidation, which was prepared by in-situ synthesis. In this paper, the C/C substrate with the protection of the HfB2-SiC-MoSi2/SiC coating could resist oxidation in 1773 K air for 408 h. The double coating also presented expected oxidation protection performance at dynamic oxidation environment. In the test process, the surface coating was oxidized to form a self-sealing silicate glass layer containing HfO2 and HfSiO4, which could hinder crack propagation in coating

    Text-Driven Foley Sound Generation With Latent Diffusion Model

    Foley sound generation aims to synthesise the background sound for multimedia content. Previous models usually employ a large development set with labels as input (e.g., single numbers or one-hot vector). In this work, we propose a diffusion model based system for Foley sound generation with text conditions. To alleviate the data scarcity issue, our model is initially pre-trained with large-scale datasets and fine-tuned to this task via transfer learning using the contrastive language-audio pertaining (CLAP) technique. We have observed that the feature embedding extracted by the text encoder can significantly affect the performance of the generation model. Hence, we introduce a trainable layer after the encoder to improve the text embedding produced by the encoder. In addition, we further refine the generated waveform by generating multiple candidate audio clips simultaneously and selecting the best one, which is determined in terms of the similarity score between the embedding of the candidate clips and the embedding of the target text label. Using the proposed method, our system ranks 1st{1}^{st} among the systems submitted to DCASE Challenge 2023 Task 7. The results of the ablation studies illustrate that the proposed techniques significantly improve sound generation performance.     Revisiting Copenhagen climate mitigation targets

    Many economies set climate mitigation targets for 2020 at the 2009 15th Conference of the Parties conference of the United Nations Framework Convention on Climate Change in Copenhagen. Yet no retrospective review of the implementation and actual mitigation associated with these targets has materialized. Here we track the national CO2 emissions from both territory and consumption (trade adjusted) perspectives to assess socioeconomic factors affecting changes in emissions. Among the 34 countries analysed, 12 failed to meet their targets (among them Portugal, Spain and Japan) and 7 achieved the target for territorial emissions, albeit with carbon leakage through international trade to meet domestic demand while increasing emissions in other countries. Key factors in meeting targets were intensity reduction of energy and the improvement of the energy mix. However, many countries efforts fell short of their latest nationally determined contributions. Timely tracking and review of mitigation efforts are critical for meeting the Paris Agreement targets

    Study on different potato continuous cropping ways on rhizosphere soil nutrients and enzyme activities

    To address the problem of food security, China produced potatoes as a staple food in 2015.However, there are increasing problems with continuous cropping production methods, potato continuous cropping has been inevitable.So it is necessary to research under the different potato continuous cropping ways, potato rhizosphere soil nutrients and enzyme activities which can direct potato fertilizer and ease potato continuous cropping obstacle. A two-growing season investigation was carried out during the spring and autumn of 2014 and 2015 to determine the different ways of potato continuous cropping on the overall growth of potatoes, soil nutrients, and enzyme activities. During continuous cropping nitrogen (N) content of rhizosphere soil was reduced; available potassium (Kav) was significantly reduced(p≤5%), especially in spring and autumn continuous cropping; and total phosphorus (Ptot) was reduced during the growth stage. However, the total potassium (Ktot), available phosphorus(Pav), and organic carbon (Ctot) increased before they decreased. For rhizosphere soil enzyme activities, urease initially increased and then decreased, and was lower in continuous cropping than multiple continuous cropping; in spring of 2015, invertase was the highest with continuous cropping. Catalase and polyphenol oxidase decreased initially before increasing. Continuous cropping in spring and autumn consumed more nutrients, especially potassium (K) than in spring. Therefore, potatoes planted in both spring and autumn enhanced the problems of continuous cropping. However, multiple continuous cropping that eased rhizosphere soil nutrient absorption and effectively improves soil nutrients and enzyme activities could provide an effective method for managing the negative impacts associated with continuous cropping

    CRISPR/Cas9-mediated gene mutation of EcIAG leads to sex reversal in the male ridgetail white prawn Exopalaemon carinicauda

    In the culture of crustaceans, most species show sexual dimorphism. Monosex culture is an effective approach to achieve high yield and economic value, especially for decapods of high value. Previous studies have developed some sex control strategies such as manual segregation, manipulation of male androgenic gland and knockdown of the male sexual differentiation switch gene encoding insulin-like androgenic gland hormone (IAG) in decapods. However, these methods could not generate hereditable changes. Genetic manipulation to achieve sex reversal individuals is absent up to now. In the present study, the gene encoding IAG (EcIAG) was identified in the ridgetail white prawn Exopalaemon carinicauda. Sequence analysis showed that EcIAG encoded conserved amino acid structure like IAGs in other decapod species. CRISPR/Cas9-mediated genome editing technology was used to knock out EcIAG. Two sgRNAs targeting the second exon of EcIAG were designed and microinjected into the prawn zygotes or the embryos at the first cleavage with commercial Cas9 protein. EcIAG in three genetic males was knocked out in both chromosome sets, which successfully generated sex reversal and phenotypic female characters. The results suggest that CRISPR/Cas9-mediated genome editing technology is an effective way to develop sex manipulation technology and contribute to monosex aquaculture in crustaceans

    Discovery and Characterization of a High-Affinity Small Peptide Ligand, H1, Targeting FGFR2IIIc for Skin Wound Healing

    Background/Aims: How to aid recovery from severe skin injuries, such as burns, chronic or radiation ulcers, and trauma, is a critical clinical problem. Current treatment methods remain limited, and the discovery of ideal wound-healing therapeutics has been a focus of research. Functional recombinant proteins such as basic fibroblast growth factor (bFGF) and epidermal growth factor (EGF) have been developed for skin repair, however, some disadvantages in their use remain. This study reports the discovery of a novel small peptide targeting fibroblast growth factor receptor 2 IIIc (FGFR2IIIc) as a potential candidate for skin wound healing. Methods: A phage-displayed peptide library was used for biopanning FGFR2IIIc-targeting small peptides. The selected small peptides binding to FGFR2IIIc were qualitatively evaluated by an enzyme-linked immunosorbent assay. Their biological function was detected by a cell proliferation assay. Among them, an optimized small peptide named H1 was selected for further study. The affinity of the H1 peptide and FGFR2IIIc was determined by an isothermal titration calorimetry device. The ability of theH1 peptide to promote skin wound repair was investigated using an endothelial cell tube formation assay and wound healing scratch assay in vitro. Subsequently, the H1 peptide was assessed using a rat skin full-thickness wound model and chorioallantoic membrane (CAM) assays in vivo. To explore its molecular mechanisms, RNA-Seq, quantitative real-time PCR, and western blot assays were performed. Computer molecular simulations were also conducted to analyze the binding model. Results: We identified a novel FGFR2IIIc-targeting small peptide, called H1, with 7 amino acid residues using phage display. H1 had high binding affinity with FGFR2IIIc. The H1 peptide promoted the proliferation and motility of fibroblasts and vascular endothelial cells in vitro. In addition, the H1 peptide enhanced angiogenesis in the chick chorioallantoic membrane and accelerated wound healing in a rat full-thickness wound model in vivo. The H1 peptide activated both the PI3K-AKT and MAPK-ERK1/2 pathways and simultaneously increased the secretion of vascular endothelial growth factor. Computer analysis demonstrated that the model of H1 peptide binding to FGFR2IIIc was similar to that of FGF2 and FGFR2IIIc. Conclusion: The H1 peptide has a high affinity for FGFR2IIIc and shows potential as a wound healing agent. As a substitute for bFGF, it could be developed into a novel therapeutic candidate for skin wound repair in the future
