Characteristics of anvil-top associated with the Poplar Bluff tornado of 7 May 1973
Investigation of potential tornado-producing thunderstorms was performed during part of the 1972 and 1973 tornado seasons. On May 7, 1973 twenty-one tornadoes were confirmed over southern Missouri, northern Arkansas, and southwestern Illinois. The region was surveyed by high altitude photography performed on a Learjet over the region of reported tornadoes. Two storms were chosen from aircraft observation with the guidance of ground and radar reports, and a series of photographs were taken of a tornado producing cloud. An analysis of the activity before and during the tornado is made, and most noteworthy were changes detected in the growth and collapse of overshooting domes above the anvil. Suggestions are included for a comprehensive study
Satellite-tracked cumulus velocities
The research indicates that extreme caution must be exercised in converting cloud velocities into winds. The motion of fair-weather cumuli obtained by tracking their shadows over Springfield, Missouri revealed that the standard deviation in the individual cloud motion is several times the tracking error. The motion of over-ocean cumuli near Barbados clearly indicated the complicated nature of cumulus velocities. Analysis of whole-sky images obtained near Tampa, Florida failed to show significant continuity and stability of cumulus plumes, less than 0.3 mile in diameter. Cumulus turrets with 0.3 to 2 mile in size appear to be the best target to infer the mean wind within the subcloud layers. Cumulus or stratocumulus cells consisting of x number of turrets do not always move with wind. The addition and deletion of turrets belonging to a specific cell appear to be the cause of the erratic motion of a tracer cell. It may by concluded that the accuracy of wind estimates is unlikely to be better than 2m/sec unless the physical and dynamical characteristics of cumulus motion is futher investigated
Specific recognition of a multiply phosphorylated motif in the DNA repair scaffold XRCC1 by the FHA domain of human PNK.
Short-patch repair of DNA single-strand breaks and gaps (SSB) is coordinated by XRCC1, a scaffold protein that recruits the DNA polymerase and DNA ligase required for filling and sealing the damaged strand. XRCC1 can also recruit end-processing enzymes, such as PNK (polynucleotide kinase 3'-phosphatase), Aprataxin and APLF (aprataxin/PNK-like factor), which ensure the availability of a free 3'-hydroxyl on one side of the gap, and a 5'-phosphate group on the other, for the polymerase and ligase reactions respectively. PNK binds to a phosphorylated segment of XRCC1 (between its two C-terminal BRCT domains) via its Forkhead-associated (FHA) domain. We show here, contrary to previous studies, that the FHA domain of PNK binds specifically, and with high affinity to a multiply phosphorylated motif in XRCC1 containing a pSer-pThr dipeptide, and forms a 2:1 PNK:XRCC1 complex. The high-resolution crystal structure of a PNK-FHA-XRCC1 phosphopeptide complex reveals the basis for this unusual bis-phosphopeptide recognition, which is probably a common feature of the known XRCC1-associating end-processing enzymes
The Value Of Extra Curricula Activities In Rural Schools
Within the past ten years, the educational program has undergone many changes. The school has come to realize the importance of the distinction between the mastery of school tasks and the learning that takes place outside the school, and the wise schoolmaster has come to see that both he and his teachers are not fulfilling their true function as instructors, guides and councilors of the youth unless they all help to organize and direct the many leisure activities of the pupil. The result of this new vision has been that the function of the teacher is changed, her responsibility has been broadened, to include the whole child. Extra curricula activities have gained recognition among educators as a vital part of every school program intended to train the pupils to take their places in the democracy of the social order. Their place In the program is the result of a changed viewpoint as to what the aim of the school should be. An entirely new interest in the extra-curricula activities of the youth has been taken over by the school, and the attention of the world is directed a new interest to the youth, as the hope of civilization. There has been a fortunate change in attitudes, and all new tendencies of action have arisen, new emotions begin to sway youth, new ideas as to life begin to be formulated, and tends to become fixed, and serious thought is given to the conduct and qualities of teachers. Social attitudes and tendencies of importance in later life are inclined to become fixed through these activities. The rule of the group tends to become the rewards of regular school credit. Within the past eight years, the general social condition of America has increased the importance of extra-curricula! activities to the extent that the schools of the country have been forced to a greater degree to place them in the program of the schools. The purpose of this paper is to show that the extracurricular activities are developing to the extent of showing increasing importance to the rural as well as the urban school children, in helping them to live a less burdensome rural life
Making Accident Statistics More Meaningful
With the improvement in driver licensing and highway policing systems throughout the country there has arisen the need for better reporting and analysis of accidents and accident trends. Many states have inaugurated standard IBM equipment and follow a more or less uniform system of recording and punching accident data on cards for machine analysis. In most respects the trends in this development have been very desirable but standardization is sometimes a virtue which may occasionally boomerang. After a system is once established it takes a great deal of effort to make a change and changes are frequently desirable. Standardization may thus become a hindrance to progress. In other areas this may be readily illustrated by the English and American system of mensuration as well as by our clumsy 12-month calendar. However, the purpose of this paper is not to propose a new system, but to add some needed extensions and improvements which make any system now in common usage much more effective. An illustration using actual data will be set forth to illustrate one type of problem in the field of accident prevention
Genetic analysis of safflower domestication.
BackgroundSafflower (Carthamus tinctorius L.) is an oilseed crop in the Compositae (a.k.a. Asteraceae) that is valued for its oils rich in unsaturated fatty acids. Here, we present an analysis of the genetic architecture of safflower domestication and compare our findings to those from sunflower (Helianthus annuus L.), an independently domesticated oilseed crop within the same family.We mapped quantitative trait loci (QTL) underlying 24 domestication-related traits in progeny from a cross between safflower and its wild progenitor, Carthamus palaestinus Eig. Also, we compared QTL positions in safflower against those that have been previously identified in cultivated x wild sunflower crosses to identify instances of colocalization.ResultsWe mapped 61 QTL, the vast majority of which (59) exhibited minor or moderate phenotypic effects. The two large-effect QTL corresponded to one each for flower color and leaf spininess. A total of 14 safflower QTL colocalized with previously reported sunflower QTL for the same traits. Of these, QTL for three traits (days to flower, achene length, and number of selfed seed) had cultivar alleles that conferred effects in the same direction in both species.ConclusionsAs has been observed in sunflower, and unlike many other crops, our results suggest that the genetics of safflower domestication is quite complex. Moreover, our comparative mapping results indicate that safflower and sunflower exhibit numerous instances of QTL colocalization, suggesting that parallel trait transitions during domestication may have been driven, at least in part, by parallel genotypic evolution at some of the same underlying genes
RankPL: A Qualitative Probabilistic Programming Language
In this paper we introduce RankPL, a modeling language that can be thought of
as a qualitative variant of a probabilistic programming language with a
semantics based on Spohn's ranking theory. Broadly speaking, RankPL can be used
to represent and reason about processes that exhibit uncertainty expressible by
distinguishing "normal" from" surprising" events. RankPL allows (iterated)
revision of rankings over alternative program states and supports various types
of reasoning, including abduction and causal inference. We present the
language, its denotational semantics, and a number of practical examples. We
also discuss an implementation of RankPL that is available for download
Expressed in the yeast Saccharomyces cerevisiae, human ERK5 is a client of the Hsp90 chaperone that complements loss of the Slt2p (Mpk1p) cell integrity stress-activated protein kinase
ERK5 is a mitogen-activated protein (MAP) kinase regulated in human cells by diverse mitogens and stresses but also suspected of mediating the effects of a number of oncogenes. Its expression in the slt2Delta Saccharomyces cerevisiae mutant rescued several of the phenotypes caused by the lack of Slt2p (Mpk1p) cell integrity MAP kinase. ERK5 is able to provide this cell integrity MAP kinase function in yeast, as it is activated by the cell integrity signaling cascade that normally activates Slt2p and, in its active form, able to stimulate at least one key Slt2p target (Rlm1p, the major transcriptional regulator of cell wall genes). In vitro ERK5 kinase activity was abolished by Hsp90 inhibition. ERK5 activity in vivo was also lost in a strain that expresses a mutant Hsp90 chaperone. Therefore, human ERK5 expressed in yeast is an Hsp90 client, despite the widely held belief that the protein kinases of the MAP kinase class are non-Hsp90-dependent activities. Two-hybrid and protein binding studies revealed that strong association of Hsp90 with ERK5 requires the dual phosphorylation of the TEY motif in the MAP kinase activation loop. These phosphorylations, at positions adjacent to the Hsp90-binding surface recently identified for a number of protein kinases, may cause a localized rearrangement of this MAP kinase region that leads to creation of the Hsp90-binding surface. Complementation of the slt2Delta yeast defect by ERK5 expression establishes a new tool with which to screen for novel agonists and antagonists of ERK5 signaling as well as for isolating mutant forms of ERK5
Crystal structure of the catalytic fragment of murine poly(ADP-ribose) polymerase-2.
Poly(ADP-ribose) polymerase-1 (PARP-1) has become an important pharmacological target in the treatment of cancer due to its cellular role as a 'DNA-strand break sensor', which leads in part to resistance to some existing chemo- and radiological treatments. Inhibitors have now been developed which prevent PARP-1 from synthesizing poly(ADP-ribose) in response to DNA-breaks and potentiate the cytotoxicity of DNA damaging agents. However, with the recent discoveries of PARP-2, which has a similar DNA-damage dependent catalytic activity, and additional members containing the 'PARP catalytic' signature, the isoform selectivity and resultant pharmacological effects of existing inhibitors are brought into question. We present here the crystal structure of the catalytic fragment of murine PARP-2, at 2.8 A resolution, and compare this to the catalytic fragment of PARP-1, with an emphasis on providing a possible framework for rational drug design in order to develop future isoform-specific inhibitors
