    Static & Thermal Analysis of Piston for Single Cylinder Diesel Engine by Ansys : A Review

    The main objective of this present research work is to investigate and analyze the stresses which are acting on the piston. The parameter which is used for analysis is temperature, operating pressure and properties of materials. Piston in internal combustion engine play most important role because it transfers the force which is produced during the combustion of fuel to crankshaft due to which it converts the pressure energy into mechanical energy. Piston fails due to thermal and mechanical stresses which are produced under the running condition of engine. The boundary conditions are applied for static and thermal analysis which includes pressure on piston head during working period and uneven temperature distributions occurs from head of piston to skirt of piston. The 3D-model will be created using SOLIDWORKS2018.the 3D model will be imported in the Ansys software for geometry and meshing purposes then the FEM Analysis will be done on ANSYS software. Analysis will help to change the existing design for reduce the weight. Finite Element Analysis is the best method for analyzing the stresses in piston

    DEVELOPMENT OPTIMIZATION AND EVALUATION OF EFFERVESCENT TABLETS OF CHLORPHENIRAMINE MALEATE USING BOX BEHNKEN DESIGN

    Objective: The objective of present study was to develop effervescent tablets of Chlorpheniramine maleate (CPM) for the treatment of dysphasia.Methods: Effervescent tablets were prepared by direct compression method and were optimized using box behnken design. Amount to sodium bicarbonate (X1), amount of tartaric acid (X2) and amount of fumaric acid (X3) were selected as independent variables, whereas disintegration time (Y1), amount of carbon dioxide (Y2) and drug release in 5 minutes (Y3) were selected as dependent variables. All the batches were also evaluated for general post compression evaluation of tablet such as-weight variation, thickness, friability and hardness. From the results of design batches, best batch was selected and evaluated for in vivo pharmacokinetic study in rabbit model.Results: The disintegration time ranged from 103.33 ± 0.24 sec to 157.00 ± 0.75 sec while amount of carbon dioxide ranged from 0.26±0.014 g to 2.03±0.056 g in all the design batches. From the results of design batches, batch B4 was selected as optimized batch due to higher amount of released carbon dioxide and faster drug release as compared to other batches. Batch B4 was showing higher AUC and Cmax while lower tmax as compared to drug suspension while performing in vivo study of optimized batch in rabbit model.Conclusion: The study concluded that the combination of sodium bicarbonate, tartaric acid and fumaric acid approach for development of effervescent tablet aids to achieve faster disintegration and faster drug release property for CPM.Â

    Spectrum of clinical and biochemical profile and its importance in diabetic ketoacidosis: a tertiary care hospital experience in Gujarat

    Background: Diabetes mellitus (DM) is a disorder of multiple etiologies, and diabetic ketoacidosis (DKA) is one of the life-threatening complications of DM. This study was aimed to study the clinical and biochemical profile of DKA patients. Methods: We conducted this retrospective study at a tertiary care hospital in Gujarat. We included total 100 patients above the age of 18 years having DM, presented with DKA. A detailed history, clinical examination and biochemical tests were carried out: random blood sugar, urinalysis, arterial blood gas analysis, serum ketone, HbA1c, serum creatinine, serum electrolytes- potassium and sodium and serum osmolality. The results were analysed using Microsoft excel. Results: Out of total 100 patients, 76 (76%) patients had type 1 DM and 24 (24%) had type 2 DM, 42 (42%) were in the age group of 18-30 years. The mean age was 39.75±12.14 years. There were 56 (56%) males and 44 (44%) females with male: female ratio of 1.3:1. Most common clinical features of DKA were nausea/vomiting (88%), breathlessness (43%) and fever (35%). The main biochemical parameters altered were blood sugar (>400 mg/dl in 41%) and serum ketones (≥5 in 84%) as well as majority of patients (64%) had their HbA1c level between 11-15% with the mean HbA1c value of 12.25±2.43%. Conclusions: DKA is a frequently observed emergency with high mortality rate. Education regarding symptoms of ketoacidosis, not missing insulin doses especially during illness, strict adherence to treatment and lifestyle modifications can greatly reduce DKA occurrence

    Changes in WBC and platelet count in patients with malaria: a hospital based comparative study

    Introduction: Malaria is one of the most common infectious diseases of tropics. It presents with varied clinicopathological manifestations. Most of the complication in malaria occurs due to various hematological abnormalities. Present study was aimed to find out abnormalities in WBC and platelet counts in patients with malaria. Methods: A total 135 patients either hospitalized or treated on an outpatient basis were included in the study after positive identification for malarial parasites on Giemsa stained PSMP smears. WBC and platelet count was carried out on 3 part hematology analyzer (Sysmax KX 21). WBC count less than 4000/cumm was considered as leucopenia and platelet count less than 150000/cumm was considered as thrombocytopenia. Results: The present study includes 135 patients with malaria from which 72.59% of subjects were male and 27.41% of subjects were female. P. falciparum was present in 68.89% of cases, P. vivax in 28.15% of cases. Majority of patients had normal leucocyte count (97.03%). Neutrophilia with lymphopenia was observed in both species of malaria in our study. Thrombocytopenia was observed in89.62% of cases in malaria. Thrombocytopenia in P. falciparum was found in 92.48% of cases and in P. vivax it was 81.57% of cases. Conclusion: Present study did not show any significant change in WBC count. Present study showed neutrophilia with relative lymphopenia in both group of malaria. Incidence of thrombocytopenia was observed in both species of malaria without any statistical significance.

    Determination of Mesalamine Related Impurities from Drug Product by Reversed Phase Validated UPLC Method

    In the present study gradient reversed-phase UPLC method was developed for simultaneous determination and separation of impurities and degradation products from drug product. The chromatographic separation was performed on acquity UPLC BEH C18 column (50 mm×2.1 mm, 1.7 µm) using gradient elution. Other UPLC parameters which were optimised are flow rate, 0.7 mL/min; detection wavelength, 220 nm; column oven temperature, 40 °C and injection volume 7 µL. Stability indicating capability was established by forced degradation experiments and separation of known degradation products. The method was validated as per International Conference on Harmonization (ICH) guideline. For all impurities and mesalamine, LOQ (limit of quantification) value was found precise with RSD (related standard daviation) of less than 2.0%. In essence, the present study provides an improved low detection limit and lower run time for evaluation of pharmaceutical quality of mesalamine delayed-release formulation. Moreover, the developed method was successfully applied for quantification of impurities and degradation products in mesalamine delayed-release formulation. The same method can also be used for determination of related substances from mesalamine drug substance
