    Strategies for Using Spiritual Gifts in Korean Church Growth

    This writer believes that one of the key ingredients to building an effective growing Korean church is spiritual gifts. If every Korean Christian were involved according to his spiritual gifts, he would be used in ministering where he is most usable. This writer finds that growing churches cannot be dissociated from growing Christians, when spiritual gifts are well used. Therefore, when Christians have found their gifts, knows the significance of their gifts and properly exercise them, the Korean church will grow. The purpose of this dissertation is to introduce readers to the best use of spiritual gifts in the contemporary Korean church, and to build up an important contribution to Korean church growth. One more purpose of this dissertation project is to understand what the biblical principles for the Church Growth Paradigm are, and to evaluate the problems hindering it in the Korean Church Growth

    Rim 2/Hipa CACTA transposon display ; A new genetic marker technique in Oryza species

    BACKGROUND: Transposons constitute the major fractions of repetitive sequences in eukaryotes, and have been crucial in the shaping of current genomes. Transposons are generally divided into two classes according to the mechanism underlying their transposition: RNA intermediate class 1 and DNA intermediate class 2. CACTA is a class 2 transposon superfamily, which is found exclusively in plants. As some transposons, including the CACTA superfamily, are highly abundant in plant species, and their nucleotide sequences are highly conserved within a family, they can be utilized as genetic markers, using a slightly modified version of the conventional AFLP protocol. Rim2 /Hipa is a CACTA transposon family having 16 bp consensus TIR sequences to be present in high copy numbers in rice genome. This research was carried out in order to develop a Rim2/Hipa CACTA-AFLP or Rim2/Hipa CACTA-TD (transposon display, hereafter Rim2/Hipa-TD) protocol for the study of genetic markers in map construction and the study of genetic diversity in rice. RESULTS: Rim2/Hipa-TD generated ample polymorphic profiles among the different rice accessions, and the amplification profiles were highly reproducible between different thermocyclers and Taq polymerases. These amplification profiles allowed for clear distinction between two different ecotypes, Japonica and Indica, of Oryza sativa. In the analysis of RIL populations, the Rim2/Hipa-TD markers were found to be segregated largely in a dominant manner, although in a few cases, non-parental bands were observed in the segregating populations. Upon linkage analysis, the Rim2/Hipa-TD markers were found to be distributed in the regions proximal to the centromeres of the chromosomes. The distribution of the Rim2/Hipa CACTA elements was surveyed in 15 different Oryza species via Rim2/Hipa-TD. While Rim2/Hipa-TD yielded ample amplification profiles between 100 to 700 bp in the AA diploid Oryza species, other species having BB, CC, EE, BBCC and CCDD, profiles demonstrated that most of the amplified fragments were larger than 400 bp, and that our methods were insufficient to clearly distinguish between these fragments. However, the overall amplification profiles between species in the Oryza genus were fully distinct. Phenetic relationships among the AA diploid Oryza species, as evidenced by the Rim2/Hipa-TD markers, were matched with their geographical distributions. CONCLUSION: The abundance of the Rim2/Hipa TIR sequences is very informative since the Rim2/Hipa-TD produced high polymorphic profiles with ample reproducibility within a species as well as between species in the Oryza genus. Therefore, Rim2/Hipa-TD markers can be useful in the development of high-density of genetic map around the centromeric regions. Rim2/Hipa-TD may also prove useful in evaluations of genetic variation and species relationships in the Oryza species

    Perception of precocious puberty among school-aged children in South Korea with the experience of treatment for precocious puberty: a Q methodological approach

    Purpose This study aimed to investigate the perceptions of precocious puberty and elucidate the distinct characteristics of each type of perception related to precocious puberty among school-aged children who had undergone treatment for the condition. Methods This study applied the Q methodology to identify and classify the perceptions of precocious puberty among school-aged children who had undergone treatment for the condition. The analysis involved 34 questions from the Q sample and data from 35 individuals in the P sample, using the PC-QUANL Program for analysis. Results The perceptions of precocious puberty among school-aged children who had undergone precocious puberty treatment were classified into the following four types: "shyness - passive self-management," "resentment - suppression," "anxiety - fear," and "adaptation - acceptance." Conclusion This study investigated the experiences and perceptions of children who have undergone treatment for precocious puberty. Through the identification of four types of perceptions, we can see that there is a need to develop an intervention program for nursing that is tailored to the specific type of precocious puberty

    Elevated intracellular cAMP exacerbates vulnerability to oxidative stress in optic nerve head astrocytes.

    Glaucoma is characterized by a progressive loss of retinal ganglion cells and their axons, but the underlying biological basis for the accompanying neurodegeneration is not known. Accumulating evidence indicates that structural and functional abnormalities of astrocytes within the optic nerve head (ONH) have a role. However, whether the activation of cyclic adenosine 3',5'-monophosphate (cAMP) signaling pathway is associated with astrocyte dysfunction in the ONH remains unknown. We report here that the cAMP/protein kinase A (PKA) pathway is critical to ONH astrocyte dysfunction, leading to caspase-3 activation and cell death via the AKT/Bim/Bax signaling pathway. Furthermore, elevated intracellular cAMP exacerbates vulnerability to oxidative stress in ONH astrocytes, and this may contribute to axonal damage in glaucomatous neurodegeneration. Inhibition of intracellular cAMP/PKA signaling activation protects ONH astrocytes by increasing AKT phosphorylation against oxidative stress. These results strongly indicate that activation of cAMP/PKA pathway has an important role in astrocyte dysfunction, and suggest that modulating cAMP/PKA pathway has therapeutic potential for glaucomatous ONH degeneration

    M3FPolypSegNet: Segmentation Network with Multi-frequency Feature Fusion for Polyp Localization in Colonoscopy Images

    Polyp segmentation is crucial for preventing colorectal cancer a common type of cancer. Deep learning has been used to segment polyps automatically, which reduces the risk of misdiagnosis. Localizing small polyps in colonoscopy images is challenging because of its complex characteristics, such as color, occlusion, and various shapes of polyps. To address this challenge, a novel frequency-based fully convolutional neural network, Multi-Frequency Feature Fusion Polyp Segmentation Network (M3FPolypSegNet) was proposed to decompose the input image into low/high/full-frequency components to use the characteristics of each component. We used three independent multi-frequency encoders to map multiple input images into a high-dimensional feature space. In the Frequency-ASPP Scalable Attention Module (F-ASPP SAM), ASPP was applied between each frequency component to preserve scale information. Subsequently, scalable attention was applied to emphasize polyp regions in a high-dimensional feature space. Finally, we designed three multi-task learning (i.e., region, edge, and distance) in four decoder blocks to learn the structural characteristics of the region. The proposed model outperformed various segmentation models with performance gains of 6.92% and 7.52% on average for all metrics on CVC-ClinicDB and BKAI-IGH-NeoPolyp, respectively.Comment: 5pages. 2023 IEEE International Conference on Image Processing (ICIP).     Therapeutic Co-targeting of WEE1 and ATM Downregulates PD-L1 Expression in Pancreatic Cancer

    Purpose Pancreatic cancer (PC) is one of the most lethal cancers worldwide, but there are currently no effective treatments. The DNA damage response (DDR) is under investigation for the development of novel anti-cancer drugs. Since DNA repair pathway alterations have been found frequently in PC, the purpose of this study was to test the DDR-targeting strategy in PC using WEE1 and ATM inhibitors. Materials and Methods We performed in vitro experiments using a total of ten human PC cell lines to evaluate antitumor effect of AZD1775 (WEE1 inhibitor) alone or combination with AZD0156 (ATM inhibitor). We established Capan-1-mouse model for in vivo experiments to confirm our findings. Results In our research, we found that WEE1 inhibitor (AZD1775) as single agent showed anti-tumor effects in PC cells, however, targeting WEE1 upregulated p-ATM level. Here, we observed that co-targeting of WEE1 and ATM acted synergistically to reduce cell proliferation and migration, and to induce DNA damage in vitro. Notably, inhibition of WEE1 or WEE1/ATM downregulated programmed cell death ligand 1 expression by blocking glycogen synthase kinase-3 beta serine 9 phosphorylation and decrease of CMTM6 expression. In Capan-1 mouse xenograft model, AZD1775 plus AZD0156 (ATM inhibitor) treatment reduced tumor growth and downregulated tumor expression of programmed cell death ligand 1, CMTM6, CD163, and CXCR2, all of which contribute to tumor immune evasion. Conclusion Dual blockade of WEE1 and ATM might be a potential therapeutic strategy for PC. Taken together, our results support further clinical development of DDR-targeting strategies for PC.

    Estimating remineralized phosphate and its remineralization rate in the northern East China Sea during Summer 1997 : a snapshot study before Three-Gorges Dam construction

As the northern East China Sea region is vulnerable to climate forcing and anthropogenic impacts, it is important to investigate how the remineralization rate in the northern East China Sea has changed in response to such external forcing. We used an historical hydrographic dataset from August 1997 to obtain a baseline for future comparison. We estimate the amount of remineralized phosphate by decomposing the physical mixing and biogeochemical process effect using water column measurements (temperature, salinity, and phosphate). The estimated remineralized phosphate column inventory ranged from 0.8 to 42.4 mmol P m-2 (mean value of 15.2 ± 12.0 mmol P m-2). Our results suggest that the Tsushima Warm Current was a strong contributor to primary production during the summer of 1997 in the study area. The estimated summer (June - August) remineralization rate in the region before Three-Gorges Dam construction was 18 ± 14 mmol C m-2 d-1.T.
