Abstract: The effect of drying method on the preservation of aloe's functional substances, as well as total antioxidants and glass transition temperature (T g ) of dried aloe were investigated. The differentiations of drying upon the polysaccharides of the gel were studied using 1 H NMR and FT-IR. The concentration of minerals in aloe vera gels were determined by ICP-AES before and after drying. Total antioxidants were measured using DPPH enzyme. T g , measured for various water activities using DSC, decreased with water activity increment. Concentration of polysaccharides was affected by the drying process, while minerals concentration remained practically constant
AF-MSCs fate can be regulated by culture conditions
Human mesenchymal stem cells (hMSCs) represent a population of multipotent adherent cells able to differentiate into many lineages. In our previous studies, we isolated and expanded fetal MSCs from second-trimester amniotic fluid (AF) and characterized them based on their phenotype, pluripotency and proteomic profile. In the present study, we investigated the plasticity of these cells based on their differentiation, dedifferentiation and transdifferentiation potential in vitro. To this end, adipocyte-like cells (AL cells) derived from AF-MSCs can regain, under certain culture conditions, a more primitive phenotype through the process of dedifferentiation. Dedifferentiated AL cells derived from AF-MSCs (DAF-MSCs), gradually lost the expression of adipogenic markers and obtained similar morphology and differentiation potential to AF-MSCs, together with regaining the pluripotency marker expression. Moreover, a comparative proteomic analysis of AF-MSCs, AL cells and DAF-MSCs revealed 31 differentially expressed proteins among the three cell populations. Proteins, such as vimentin, galectin-1 and prohibitin that have a significant role in stem cell regulatory mechanisms, were expressed in higher levels in AF-MSCs and DAF-MSCs compared with AL cells. We next investigated whether AL cells could transdifferentiate into hepatocyte-like cells (HL cells) directly or through a dedifferentiation step. AL cells were cultured in hepatogenic medium and 4 days later they obtained a phenotype similar to AF-MSCs, and were termed as transdifferentiated AF-MSCs (TRAF-MSCs). This finding, together with the increase in pluripotency marker expression, indicated the adaption of a more primitive phenotype before transdifferentiation. Additionally, we observed that AF-, DAF- and TRAF-MSCs displayed similar clonogenic potential, secretome and proteome profile. Considering the easy access to this fetal cell source, the plasticity of AF-MSCs and their potential to dedifferentiate and transdifferentiate, AF may provide a valuable tool for cell therapy and tissue engineering applications
Secure certification of mixed quantum states with application to two-party randomness generation
We investigate sampling procedures that certify that an arbitrary quantum
state on subsystems is close to an ideal mixed state
for a given reference state , up to errors on a few positions. This
task makes no sense classically: it would correspond to certifying that a given
bitstring was generated according to some desired probability distribution.
However, in the quantum case, this is possible if one has access to a prover
who can supply a purification of the mixed state.
In this work, we introduce the concept of mixed-state certification, and we
show that a natural sampling protocol offers secure certification in the
presence of a possibly dishonest prover: if the verifier accepts then he can be
almost certain that the state in question has been correctly prepared, up to a
small number of errors.
We then apply this result to two-party quantum coin-tossing. Given that
strong coin tossing is impossible, it is natural to ask "how close can we get".
This question has been well studied and is nowadays well understood from the
perspective of the bias of individual coin tosses. We approach and answer this
question from a different---and somewhat orthogonal---perspective, where we do
not look at individual coin tosses but at the global entropy instead. We show
how two distrusting parties can produce a common high-entropy source, where the
entropy is an arbitrarily small fraction below the maximum (except with
negligible probability)
Tailoring the Surface Chemistry of PEDOT:PSS to Promote Supported Lipid Bilayer Formation
This communication reports on a versatile and substrate-agnostic method to tune the surface chemistry of conducting polymers with the aim of bridging the chemical mismatch between bioelectronic devices and biological systems. As a proof of concept, the surface of poly(3,4-ethylenedioxythiophene) polystyrene sulfonate (PEDOT:PSS) is grafted with a short-chain oligoethylene glycol monolayer to favor the formation of cell-derived supported lipid bilayers (SLBs). This method is tuned to optimize the affinity between the supported lipid bilayer and the conducting polymer, leading to significant improvements in bilayer quality and therefore electronic readouts. To validate the impact of surface functionalization on the system's ability to transduce biological phenomena into quantifiable electronic signals, the activity of a virus commonly used as a surrogate for SARS-CoV-2 (mouse hepatitis virus) is monitored with and without surface treatment. The functionalized devices exhibit significant improvements in electronic output, stemming from the improved SLB quality, therefore strengthening the case for the use of such an approach in membrane-on-a-chip systems
Cosmological Evolution of a Brane Universe in a Type 0 String Background
We study the cosmological evolution of a D3-brane Universe in a type 0 string
background. We follow the brane-universe along the radial coordinate of the
background and we calculate the energy density which is induced on the brane
because of its motion in the bulk. We find that for some typical values of the
parameters and for a particular range of values of the scale factor of the
brane-universe, the effective energy density is dominated by a term
proportional to indicating a slow varying inflationary
phase. For larger values of the scale factor the effective energy density takes

Assessing cognitive dysfunction in Parkinson’s: An online tool to detect visuo-perceptual deficits
BACKGROUND: People with Parkinson’s disease
(PD) who develop visuo-perceptual deficits are at higher
risk of dementia, but we lack tests that detect subtle visuoperceptual
deficits and can be performed by untrained personnel.
Hallucinations are associated with cognitive impairment
and typically involve perception of complex objects.
Changes in object perception may therefore be a sensitive
marker of visuo-perceptual deficits in PD.
Objective: We developed an online platform to test
visuo-perceptual function. We hypothesised that (1)
visuo-perceptual deficits in PD could be detected using
online tests, (2) object perception would be preferentially
affected, and (3) these deficits would be caused
by changes in perception rather than response bias.
METHODS: We assessed 91 people with PD and 275 controls.
Performance was compared using classical frequentist
statistics. We then fitted a hierarchical Bayesian signal
detection theory model to a subset of tasks.
RESULTS: People with PD were worse than controls at
object recognition, showing no deficits in other visuoperceptual
tests. Specifically, they were worse at identifying
skewed images (P <.0001); at detecting hidden
objects (P 5.0039); at identifying objects in peripheral
vision (P <.0001); and at detecting biological motion
(P 5.0065). In contrast, people with PD were not worse
at mental rotation or subjective size perception. Using
signal detection modelling, we found this effect was
driven by change in perceptual sensitivity rather than
response bias.
CONCLUSIONS: Online tests can detect visuo-perceptual defi-
cits in people with PD, with object recognition particularly
affected. Ultimately, visuo-perceptual tests may be developed
to identify at-risk patients for clinical trials to slow PD dementia

Određivanje donepezil hidroklorida u humanoj plazmi i ljekovitim oblicima pomoću HPLC s detekcijom fluorescencije
A sensitive, isocratic reversed-phase high performance liquid chromatographic method involving fluorescence detection was developed for the determination of donepezil hydrochloride in tablets and in human plasma. Pindolol was successfully used as an internal standard. Good chromatographic separation was achieved by using analytical column C18. The system operated at room temperature using a mobile phase consisting of methanol, phosphate buffer (0.02 mol L1) and triethyl amine (pH 3.5) (55: 45: 0.5 V/V/V) at a flow rate 0.9 mL min1. The analyte and internal standard were extracted from human plasma via liquid-liquid extraction. The proposed method was validated for selectivity, linearity, accuracy and precision. The calibration curve was linear over the range of 5-2000 ng mL1 of donepezil with detection limit of 1.5 ng mL1. Intra- and inter-day relative standard deviations were less than 2.5 %. The method was found to be suitable for the quality control of donepezil hydrochloride in bulk drug as well as in human plasma.Ovaj rad opisuje HPLC metodu određivanja donepezil hidroklorida (DP) u tabletama i u ljudskoj plazmi u nano području. Postavljena je osjetljiva metoda izokratične HPLC s fluorescencijskom detekcijom. Kao unutarnji standard upotrebljen je pindolol. Dobro kromatografsko odjeljivanje postignuto je primjenom analitičke kolone C18. Radna temperatura bila je sobna, a kao mobilna faza upotrebljena je smjesa metanola, fosfatnog pufera (0,02 mol L1) i trietilamina (pH 3,5) (55:45:0.5 V/V/V). Analit i unutarnji standard su ekstrahirani iz ljudske plazme ekstrakcijom tekuće-tekuće. Predložena metoda je validirana s obzirom na selektivnost, područje linearnosti, ispravnost i preciznost. Kalibracijska funkcija bila je linearna u području od 5-2000 ng mL1 donepezila, a granica detekcije iznosila je 2 ng mL1. Relativna standardna devijacija za repetabilnost i intermedijarnu preciznost bila je manja od 2,5 %. Metoda je primjenljliva u kontroli kvalitete ljekovitih formulacija s DP-om i u praćenju DP-a u ljudskoj plazmi
Effect of intravenous clarithromycin in patients with sepsis, respiratory and multiple organ dysfunction syndrome: a randomized clinical trial.
Clarithromycin may act as immune-regulating treatment in sepsis and acute respiratory dysfunction syndrome. However, clinical evidence remains inconclusive. We aimed to evaluate whether clarithromycin improves 28-day mortality among patients with sepsis, respiratory and multiple organ dysfunction syndrome.
We conducted a multicenter, randomized, clinical trial in patients with sepsis. Participants with ratio of partial oxygen pressure to fraction of inspired oxygen less than 200 and more than 3 SOFA points from systems other than the respiratory function were enrolled between December 2017 and September 2019. Patients were randomized to receive 1 gr of clarithromycin or placebo intravenously once daily for 4 consecutive days. The primary endpoint was 28-day all-cause mortality. Secondary outcomes were 90-day mortality; sepsis response (defined as at least 25% decrease in SOFA score by day 7); sepsis recurrence; and differences in peripheral blood cell populations and leukocyte transcriptomics.
Fifty-five patients were allocated to each arm. By day 28, 27 (49.1%) patients in the clarithromycin and 25 (45.5%) in the placebo group died (risk difference 3.6% [95% confidence interval (CI) - 15.7 to 22.7]; P = 0.703, adjusted OR 1.03 [95%CI 0.35-3.06]; P = 0.959). There were no statistical differences in 90-day mortality and sepsis response. Clarithromycin was associated with lower incidence of sepsis recurrence (OR 0.21 [95%CI 0.06-0.68]; P = 0.012); significant increase in monocyte HLA-DR expression; expansion of non-classical monocytes; and upregulation of genes involved in cholesterol homeostasis. Serious and non-serious adverse events were equally distributed.
Clarithromycin did not reduce mortality among patients with sepsis with respiratory and multiple organ dysfunction. Clarithromycin was associated with lower sepsis recurrence, possibly through a mechanism of immune restoration. Mammography screening: views from women and primary care physicians in Crete
Background: Breast cancer is the most commonly diagnosed cancer among women and a leading cause of death from cancer in women in Europe. Although breast cancer incidence is on the rise worldwide, breast cancer mortality over the past 25 years has been stable or decreasing in some countries and a fall in breast cancer mortality rates in most European countries in the 1990s was reported by several studies, in contrast, in Greece have not reported these favourable trends. In Greece, the age-standardised incidence and mortality rate for breast cancer per 100.000 in 2006 was 81,8 and 21,7 and although it is lower than most other countries in Europe, the fall in breast cancer mortality that observed has not been as great as in other European countries. There is no national strategy for screening in this country. This study reports on the use of mammography among middleaged women in rural Crete and investigates barriers to mammography screening encountered by women and their primary care physicians.
Methods: Design: Semi-structured individual interviews. Setting and participants: Thirty women between 45–65
years of age, with a mean age of 54,6 years, and standard deviation 6,8 from rural areas of Crete and 28 qualified
primary care physicians, with a mean age of 44,7 years and standard deviation 7,0 serving this rural population.
Main outcome measure: Qualitative thematic analysis.
Results: Most women identified several reasons for not using mammography. These included poor knowledge
of the benefits and indications for mammography screening, fear of pain during the procedure, fear of a serious
diagnosis, embarrassment, stress while anticipating the results, cost and lack of physician recommendation.
Physicians identified difficulties in scheduling an appointment as one reason women did not use mammography
and both women and physicians identified distance from the screening site, transportation problems and the
absence of symptoms as reasons for non-use.
Conclusion: Women are inhibited from participating in mammography screening in rural Crete. The provision
of more accessible screening services may improve this. However physician recommendation is important in
overcoming women's inhibitions. Primary care physicians serving rural areas need to be aware of barriers
preventing women from attending mammography screening and provide women with information and advice in a sensitive way so women can make informed decisions regarding breast caner screening
