No more time to stay ‘single’ in the detection of Anisakis pegreffii, A. simplex (s. s.) and hybridization events between them: a multi-marker nuclear genotyping approach
A multi-marker nuclear genotyping approach was performed on larval and adult specimens of Anisakis spp. (N = 689) collected from fish and cetaceans in allopatric and sympatric areas of the two species Anisakis pegreffii and Anisakis simplex
(s. s.), in order to: (1) identify specimens belonging to the parental taxa by using nuclear markers (allozymes loci) and sequence analysis of a new diagnostic nuclear DNA locus (i.e. partial sequence of the EF1 α−1 nDNA region) and (2) recognize hybrid categories. According to the Bayesian clustering algorithms, based on those markers, most of the individuals
(N = 678) were identified as the parental species [i.e. A. pegreffii or A. simplex (s. s.)], whereas a smaller portion (N = 11)
were recognized as F1 hybrids. Discordant results were obtained when using the polymerase chain reaction–restriction
fragment length polymorphisms (PCR–RFLPs) of the internal transcribed spacer (ITS) ribosomal DNA (rDNA) on
the same specimens, which indicated the occurrence of a large number of ‘hybrids’ both in sympatry and allopatry.
These findings raise the question of possible misidentification of specimens belonging to the two parental Anisakis and
their hybrid categories derived from the application of that single marker (i.e. PCR–RFLPs analysis of the ITS of
rDNA). Finally, Bayesian clustering, using allozymes and EF1 α−1 nDNA markers, has demonstrated that hybridization
between A. pegreffii and A. simplex (s. s.) is a contemporary phenomenon in sympatric areas, while no introgressive hybridization takes place between the two species
Infection levels and species diversity of ascaridoid nematodes in Atlantic cod, Gadus morhua, are correlated with geographic area and fish size
Atlantic cod (Gadus morhua) is among the most important commercial fish species on the world market. Its
infection by ascaridoid nematodes has long been known, Pseudoterranova even being named cod worm. In the
present study, 755 individuals were sampled in the Barents, Baltic and North Seas during 2012–2014.
Prevalences for Anisakis in whole fish and in fillets in the different fishing areas varied from 16 to 100% and
from 12 to 90% respectively. Abundance was also greatly influenced by the sampling area. Generalized additive
model results indicate higher numbers of Anisakis in the North Sea, even after the larger body size was accounted
for. Numbers and prevalence of Anisakis were positively related to fish length or weight. The prevalence of
parasites in whole fish and in fillets was also influenced by the season, with the spring displaying a peak for the
prevalence in whole fish and, at the same time, a drop for the prevalence in fillets. Whereas 46% of cod had
Anisakis larvae in their fillets, the majority (39%) had parasites mainly in the ventral part of the fillet and only
12% had parasites in their dorsal part. This observation is of importance for the processing of the fish. Indeed,
the trimming of the ventral part of the cod fillet would allow the almost total elimination of ascaridoids except
for cod from the Baltic Sea where there was no difference between the dorsal and the ventral part.
The presence of other ascaridoid genera was also noticeable in some areas. For Pseudoterranova, the highest
prevalence (45%) in whole fish was observed in the Northern North Sea, whereas the other areas had prevalences between 3 and 16%. Contracaecum was present in every commercial size cod sampled in the Baltic Sea
with an intensity of up to 96 worms but no Contracaecum was isolated from the Central North Sea. Non-zoonotic
Hysterothylacium was absent from the Baltic Sea but with a prevalence of 83% in the Barents and the Northern
North Sea.
A subsample of worms was identified with genetic-molecular tools and assigned to the species A. simplex (s.s.),
A. pegreffii, P. decipiens (s.s.), P. krabbei, C. osculatum and H. aduncum. In addition to high prevalence and
abundance values, the cod sampled in this study presented a diversity of ascaridoid nematodes with a majority of
fish displaying a co-infection. Out of 295 whole infected fish, 269 were co-infected by at least 2 genera
SiPM and front-end electronics development for Cherenkov light detection
The Italian Institute of Nuclear Physics (INFN) is involved in the
development of a demonstrator for a SiPM-based camera for the Cherenkov
Telescope Array (CTA) experiment, with a pixel size of 66 mm. The
camera houses about two thousands electronics channels and is both light and
compact. In this framework, a R&D program for the development of SiPMs suitable
for Cherenkov light detection (so called NUV SiPMs) is ongoing. Different
photosensors have been produced at Fondazione Bruno Kessler (FBK), with
different micro-cell dimensions and fill factors, in different geometrical
arrangements. At the same time, INFN is developing front-end electronics based
on the waveform sampling technique optimized for the new NUV SiPM. Measurements

Cognitive modulation of psychophysical, respiratory and autonomic responses to cold pressor test
In healthy subjects with high hypnotisability (highs) under hypnosis, subjectively effective suggestions for analgesia abolish the increases in blood pressure associated with cold pressor test (cpt) by reducing the peripheral vascular resistance. The aim of the present study was to investigate the effects of the suggestions of analgesia on the responses to cpt in healthy highs (n = 22) and in low hypnotisable participants (lows, n = 22) out of hypnosis. Cpt was administered without (CPT) and
with suggestions for analgesia (CPT+AN). Psychophysical (pain intensity, pain threshold, cpt duration (time of immersion) and pain tolerance, defined as the difference between cpt duration and pain threshold), respiratory (amplitude and frequency) and autonomic variables (tonic skin conductance, mean RR interval (RR = 1/heart rate), blood pressure, skin blood flow) were studied. The suggestions for analgesia increased cpt duration and RR in both groups, but decreased pain intensity and enhanced pain threshold only in highs; in both groups they did not modulate systolic blood pressure, tonic skin conductance and skin blood flow; thus, increased parasympathetic activity appears responsible for the heart rate reduction induced by suggestions in both groups. In conclusion, our findings show that suggestions modulate pain experience differentially in highs and lows, and are partially effective also in lows. We hypothesize that the mechanisms responsible for the efficacy of suggestions in healthy lows may be involved also in their efficacy in chronic pain patients with
low hypnotisability
MicroRNA-Mediated Direct Reprogramming of Human Adult Fibroblasts Toward Cardiac Phenotype
open6Modulation of microRNA expression holds the promise to achieve direct reprogramming of fibroblasts into cardiomyocyte-like cells as a new strategy for myocardial regeneration after ischemic heart disease. Previous reports have shown that murine fibroblasts can be directly reprogrammed into induced cardiomyocytes (iCMs) by transient transfection with four microRNA mimics (miR-1, 133, 208, and 499, termed "miRcombo"). Hence, study on the effect of miRcombo transfection on adult human cardiac fibroblasts (AHCFs) deserves attention in the perspective of a future clinical translation of the approach. In this brief report, we studied for the first time whether miRcombo transient transfection of AHCFs by non-viral vectors might trigger direct reprogramming of AHCFs into cardiomyocyte-like cells. Initially, efficient miRNA delivery to cells was demonstrated through the use of a commercially available transfection agent (DharmaFECT1). Transient transfection of AHCFs with miRcombo was found to upregulate early cardiac transcription factors after 7 days post-transfection and cardiomyocyte specific marker cTnT after 15 days post-transfection, and to downregulate the expression of fibroblast markers at 15 days post-transfection. The percentage of cTnT-positive cells after 15 days from miRcombo transfection was ∼11%, as evaluated by flow cytometry. Furthermore, a relevant percentage of miRcombo-transfected AHCFs (∼38%) displayed spontaneous calcium transients at 30 days post-transfection. VanA type enterococci from humans, animals and food: species distribution, population structure, Tn1546-typing and location, and virulence determinants
VanA-type human (n = 69), animal (n = 49), and food (n =36) glycopeptide-resistant enterococci (GRE)
from different geographic areas were investigated to study their possible reservoirs and transmission routes.
Pulsed-field gel electrophoresis (PFGE) revealed two small genetically related clusters, M39 (n = 4) and M49
(n = 13), representing Enterococcus faecium isolates from animal and human feces and from clinical and fecal
human samples. Multilocus sequence typing showed that both belonged to the epidemic lineage of CC17. purK
allele analysis of 28 selected isolates revealed that type 1 was prevalent in human strains (8/11) and types 6 and
3 (14/15) were prevalent in poultry (animals and meat). One hundred and five of the 154 VanA GRE isolates,
encompassing different species, origins, and PFGE types, were examined for Tn1546 type and location (plasmid
or chromosome) and the incidence of virulence determinants. Hybridization of S1- and I-CeuI-digested total
DNA revealed a plasmid location in 98% of the isolates. Human intestinal and animal E. faecium isolates bore
large (>150 kb) vanA plasmids. Results of PCR-restriction fragment length polymorphism and sequencing
showed the presence of prototype Tn1546 in 80% of strains and the G-to-T mutation at position 8234 in three
human intestinal and two pork E. faecium isolates. There were no significant associations (P > 0.5) between
Tn1546 type and GRE source or enterococcal species. Virulence determinants were detected in all reservoirs
but were significantly more frequent (P < 0.02) among clinical strains. Multiple determinants were found in
clinical and meat Enterococcus faecalis isolates. The presence of indistinguishable vanA elements (mostly
plasmid borne) and virulence determinants in different species and PFGE-diverse populations in the presence
of host-specific purK housekeeping genes suggested that all GRE might be potential reservoirs of resistance
determinants and virulence traits transferable to human-adapted clusters
The impact of COVID-19 lockdown on admission to gynecological emergency departments: Results from a multicenter Italian study
Objective: To evaluate the impact of the COVID-19 lockdown on admissions to gynecological emergency departments (ED) of three Italian university hospitals with different rates of COVID-19 incidence. Methods: A retrospective study was conducted in the gynecological EDs of Modena (Emilia-Romagna), Sassari and Cagliari (Sardinia) regarding all admissions to gynecological EDs during November 1 to 30, 2019, and March 11 to April 9, 2020 (lockdown period). Results: A total of 691 women (mean age 38.3 \ub1 14.3 years) who were admitted to the gynecological EDs were included. The relative decrease in women evaluated from March 11 to April 9, 2020, was 1256.6% (95% confidence interval [CI] 52.2\u201361.1). Time spent in the ED was also significantly shorter during this period (P=0.02) in comparison to November 1 to 30, 2019. The most evident decrease was observed for pelvic pain ( 1268.9% [95% CI 60.3\u201376.7]; 1291 cases). The management of women suggests a more effective use of the ED, with higher rates of hospitalization (P=0.001) and recourse to emergent surgeries (P=0.005) and lower rates of discharge to home (P=0.03). Conclusion: The COVID-19 lockdown greatly reduced the rate of admission to gynecological EDs, but the real emergencies were filtered from the more deferrable ones
