Demonstration of Adiabatic Variational Quantum Computing with a Superconducting Quantum Coprocessor
Adiabatic quantum computing enables the preparation of many-body ground
states. This is key for applications in chemistry, materials science, and
beyond. Realisation poses major experimental challenges: Direct analog
implementation requires complex Hamiltonian engineering, while the digitised
version needs deep quantum gate circuits. To bypass these obstacles, we suggest
an adiabatic variational hybrid algorithm, which employs short quantum circuits
and provides a systematic quantum adiabatic optimisation of the circuit
parameters. The quantum adiabatic theorem promises not only the ground state
but also that the excited eigenstates can be found. We report the first
experimental demonstration that many-body eigenstates can be efficiently
prepared by an adiabatic variational algorithm assisted with a multi-qubit
superconducting coprocessor. We track the real-time evolution of the ground and
exited states of transverse-field Ising spins with a fidelity up that can reach
about 99%.Comment: 12 pages, 4 figure
Comparing health workforce forecasting approaches for healthcare planning: The case for ophthalmologists
Health workforce planning is essential in the provision of quality healthcare. Several approaches to planning are customarily used
and advocated, each with unique underlying assumptions. Thus, a thorough understanding of each assumption is required in
order to make an informed decision on the choice of forecasting approach to be used. For illustration, we compare results for
eye care requirements in Singapore using three established workforce forecasting approaches β workforce-to-population-ratio,
needs based approach, utilization based approach β and a proposed robust integrated approach to discuss the appropriateness of
each approach under various scenarios. Four simulation models using the systems modeling methodology of system dynamics
were developed for use in each approach. These models were initialized and simulated using the example of eye care workforce
planning in Singapore, to project the number of ophthalmologists required up to the year 2040 under the four different approaches.
We found that each approach projects a different number of ophthalmologists required over time. The needs based approach tends
to project the largest number of required ophthalmologists, followed by integrated, utilization based and workforce-to-population
ratio approaches in descending order. The four different approaches vary widely in their forecasted workforce requirements and
reinforce the need to be discerning of the fundamental differences of each approach in order to choose the most appropriate
one. Further, health workforce planning should also be approached in a comprehensive and integrated manner that accounts for
developments in demographic and healthcare systems
Future requirements for and supply of ophthalmologists for an aging population in Singapore
#### Background
Singaporeβs population, as that of many other countries, is aging; this is likely to lead to an increase in eye diseases and the demand for eye care. Since ophthalmologist training is long and expensive, early planning is essential. This paper forecasts workforce and training requirements for Singapore up to the year 2040 under several plausible future scenarios.
#### Methods
The Singapore Eye Care Workforce Model was created as a continuous time compartment model with explicit workforce stocks using system dynamics. The model has three modules: prevalence of eye disease, demand, and workforce requirements. The model is used to simulate the prevalence of eye diseases, patient visits, and workforce requirements for the public sector under different scenarios in order to determine training requirements.
#### Results
Four scenarios were constructed. Under the baseline business-as-usual scenario, the required number of ophthalmologists is projected to increase by 117% from 2015 to 2040.
Under the current policy scenario (assuming an increase of service uptake due to increased awareness, availability, and accessibility of eye care services), the increase will be 175%, while under the new model of care scenario (considering the additional effect of providing some services by non-ophthalmologists) the increase will only be 150%. The moderated workload scenario (assuming in addition a reduction of the clinical workload) projects an increase in the required number of ophthalmologists of 192% by 2040.
Considering the uncertainties in the projected demand for eye care services, under the business-as-usual scenario, a residency intake of 8β22 residents per year is required, 17β21 under the current policy scenario, 14β18 under the new model of care scenario, and, under the moderated workload scenario, an intake of 18β23 residents per year is required.
#### Conclusions
The results show that under all scenarios considered, Singaporeβs aging and growing population will result in an almost doubling of the number of Singaporeans with eye conditions, a significant increase in public sector eye care demand and, consequently, a greater requirement for ophthalmologists
Free flux flow resistivity in strongly overdoped high-T_c cuprate; purely viscous motion of the vortices in semiclassical d-wave superconductor
We report the free flux flow (FFF) resistivity associated with a purely
viscous motion of the vortices in moderately clean d-wave superconductor
Bi:2201 in the strongly overdoped regime (T_c=16K) for a wide range of the
magnetic field in the vortex state. The FFF resistivity is obtained by
measuring the microwave surface impedance at different microwave frequencies.
It is found that the FFF resistivity is remarkably different from that of
conventional s-wave superconductors. At low fields (H<0.2H_c2) the FFF
resistivity increases linearly with H with a coefficient which is far larger
than that found in conventional s-wave superconductors. At higher fields, the
FFF resistivity increases in proportion to \sqrt H up to H_c2. Based on these
results, the energy dissipation mechanism associated with the viscous vortex
motion in "semiclassical" d-wave superconductors with gap nodes is discussed.
Two possible scenarios are put forth for these field dependence; the
enhancement of the quasiparticle relaxation rate and the reduction of the
number of the quasiparticles participating the energy dissipation in d-wave
vortex state.Comment: 9 pages 7 figures, to appear in Phys. Rev.
Clofazimine Inhibits Human Kv1.3 Potassium Channel by Perturbing Calcium Oscillation in T Lymphocytes
The Kv1.3 potassium channel plays an essential role in effector memory T cells and has been implicated in several important autoimmune diseases including multiple sclerosis, psoriasis and type 1 diabetes. A number of potent small molecule inhibitors of Kv1.3 channel have been reported, some of which were found to be effective in various animal models of autoimmune diseases. We report herein the identification of clofazimine, a known anti-mycobacterial drug, as a novel inhibitor of human Kv1.3. Clofazimine was initially identified as an inhibitor of intracellular T cell receptor-mediated signaling leading to the transcriptional activation of human interleukin-2 gene in T cells from a screen of the Johns Hopkins Drug Library. A systematic mechanistic deconvolution revealed that clofazimine selectively blocked the Kv1.3 channel activity, perturbing the oscillation frequency of the calcium-release activated calcium channel, which in turn led to the inhibition of the calcineurin-NFAT signaling pathway. These effects of clofazimine provide the first line of experimental evidence in support of a causal relationship between Kv1.3 and calcium oscillation in human T cells. Furthermore, clofazimine was found to be effective in blocking human T cell-mediated skin graft rejection in an animal model in vivo. Together, these results suggest that clofazimine is a promising immunomodulatory drug candidate for treating a variety of autoimmune disorders

Transgenic Expression of P1A Induced Thymic Tumor: A Role for Onco-Fetal Antigens in Tumorigenesis
P1A is the first known tumor rejection antigen. It is expressed in embryonic stem cells and multiple tumors but is silent in adult tissues except for the testis and placenta. Therefore, P1A represents a prototype for onco-fetal antigens. To test the potential function of P1A in tumorigenesis, we used a transgenic mouse expressing P1A in lymphoid cells. We observed that immunodeficient host P1A transgenic mice developed thymic tumors after 7 months of age and had shorter survival rates compared to control groups. Most of the 7 examined tumors displayed B cell lineage markers. The P1A transgenic bone marrow cells had higher proliferation ability and more potential progenitors compared to control bone marrow cells. To our knowledge, our data provided the first example that onco-fetal antigen can promote tumorigenesis
Identification of a Common Lupus Disease-Associated microRNA Expression Pattern in Three Different Murine Models of Lupus
Recent reports have shown that microRNAs (miRNAs) regulate vital immunological processes and have emerged as key regulators of immune system development and function. Therefore, it is important to determine miRNA dysregulation and its pathogenic contribution in autoimmune diseases, an aspect not adequately addressed thus far.In this study, we profiled miRNA expressions in splenic lymphocytes from three murine lupus models (MRL-lpr, B6-lpr and NZB/W(Fβ)) with different genetic background by miRNA microarray assays and Real-time RT-PCR. Despite the genetic differences among these three lupus stains, a common set of dysregulated miRNAs (miR-182-96-183 cluster, miR-31, and miR-155) was identified in splenocytes when compared with age-matched control mice. The association of these miRNAs with the disease was highlighted by our observation that this miRNA expression pattern was evident in NZB/W mice only at an age when lupus disease is manifested. Further, we have shown that the miRNA dysregulation in MRL-lpr mice was not simply due to the activation of splenocytes. By Real-time RT-PCR, we confirmed that these miRNAs were upregulated in both purified splenic B and T cells from MRL-lpr mice. miR-127 and miR-379, which were greatly upregulated in splenocytes from lpr mice, were moderately increased in diseased NZB/W mice. In addition, Real-time RT-PCR revealed that miR-146a, miR-101a, and miR-17-92 were also markedly upregulated in splenic T, but not B cells from MRL-lpr mice. The identification of common lupus disease-associated miRNAs now forms the basis for the further investigation of the pathogenic contribution of these miRNAs in autoimmune lupus, which will advance our knowledge of the role of miRNAs in autoimmunity. Given that miRNAs are conserved, with regard to both evolution and function, our observation of a common lupus disease-associated miRNA expression pattern in murine lupus models is likely to have significant pathogenic, diagnostic, and/or therapeutic implications in human lupus
In-situ yeast fermentation to enhance bioconversion of coconut endosperm waste into larval biomass of hermetia illucens: statistical augmentation of larval lipid content
The aim of this study was to spur the lipid accumulation by larvae of Hermetia illucens or black soldier fly (BSFL) via feeding with yeast fermented medium. The Saccharomyces cerevisiae, a single cell yeast, was introduced at different concentrations (0.02, 0.1, 0.5, 1.0, 2.5 wt %) to execute an in-situ fermentation on coconut endosperm waste. The rearing of BSFL was started simultaneously and the rearing was stopped once the BSFL reached the fifth instar. With the increasing of yeast concentration, the rearing duration of BSFL was shortened from 15.5 to 13.5 days. Moreover, it was found that at 0.5 to 1.0 wt % yeast concentration, the lipid yield and lipid productivity of BSFL were statistically enhanced to their highest peaks, namely, at 49.4% and 0.53 g/day, respectively. With regard to biodiesel composition, BSFL-derived biodiesel contained mainly C12:0, C14:0, C16:0 and C18:1. The higher amount of saturated fatty acids could strengthen the oxidative stability biodiesel produced as compared with non-edible oils or microalgal lipid. At last, the addition of yeast was also found to improve the waste reduction index of coconut endosperm waste (CEW) from 0.31 to 0.40 g/day, heralding the capability of BSFL to valorize organic waste via bioconversion into its biomass to serve as a feedstock for biodiesel production
