    Invasive micropapillary carcinoma of the breast overexpresses MUC4 and is associated with poor outcome to adjuvant trastuzumab in HER2-positive breast cancer

    Invasive micropapillary carcinoma of the breast (IMPC) is a histological tumor variant that occurs with low frequency characterized by an inside-out formation of tumor clusters with a pseudopapillary arrangement. IMPC is an aggressive tumor with poor clinical outcome. In addition, this histological subtype usually expresses human epidermal growth factor receptor 2 (HER2) which also correlates with a more aggressive tumor. In this work we studied the clinical significance of IMPC in HER2-positive breast cancer patients treated with adjuvant trastuzumab.     Comparison of the productive performance in broilers supplemented with probiotic or antibiotic

    El estudio comparó parámetros productivos de pollos de carne suplementados con un probiótico Biomin® Poultry 5 Star (Enterococcus faecium, Pediococcus acidilactici, Bifidobacterium animalis, Lactobacillus salivarius y Lactobacillus reuteri) versus un antibiótico (Zinc Bacitracina). Se usaron 333 aves divididas en tres tratamientos de 111 animales con 3 repeticiones por tratamiento: 1, control; 2, antibiótico; y 3, probiótico. En las seis semanas de crianza no se encontró diferencia significativa entre tratamientos para peso corporal, consumo de alimento, conversión alimenticia (ICA), porcentaje de mortalidad y el índice de eficiencia productiva (IEP).The present study compared productive parameters in broilers supplemented with a probiotic Biomin® Poultry 5 Star (Enterococcus faecium, Pediococcus acidilactici, Bifidobacterium animalis, Lactobacillus salivarius y Lactobacillus reuteri) versus an antibiotic (Bacitracin). A total of 333 chicks were divided into three treatments of 111 animals with 3 repetitions for group: 1 control; 2 antibiotic; 3 probiotic.     Biología reproductiva del pepino de mar Holothuria sanctori (Echinodermata: Holothuroidea)

    The reproductive biology of the sea cucumber Holothuria sanctori was studied over 24 months (February 2009 to January 2011) at Gran Canaria through the gonad index and a combination of macro- and microscopic analysis of the gonads. Holothuria sanctori showed a 1:1 sex ratio and a seasonal reproductive cycle with a summer spawning: the mean gonad index showed a maximum (3.99±0.02) in summer (June-July) and a minimum (0.05±0.04) between late autumn (November) and early spring (March). Females had significantly wider gonad tubules than males. First maturity occurred at a size of 201 to 210 mm, a gutted body weight of 101 to 110 g and a total weight of 176 to 200 g. Holothuria sanctori shows a typical temperate species reproduction pattern. These results could be useful for managing current extractions of H. sanctori in the Mediterranean and in case a specific fishery is started in the eastern Atlantic region.Se estudió la biología reproductiva del pepino de mar Holothuria sanctori durante 24 meses (Febrero de 2009 a Enero de 2010) en la isla de Gran Canaria, mediante el índice gonadal y una combinación de análisis macro y microscópicos de sus gónadas. Holothuria sanctori presentó una relación de sexos de 1:1 y un solo ciclo reproductivo anual con desove en los meses estivales: el índice gonadal medio presentó un máximo (3.99±0.02) en verano (Junio-Julio) y un mínimo (0.05±0.04) entre finales de otoño (Noviembre) y primeros de primavera (Marzo). Las hembras presentaron túbulos gonadales significativamente más anchos que los de los machos. La primera madurez tuvo lugar con una talla de 201-210 mm, un peso eviscerado de 101-110 g y un peso total de 176-200 g. Holothuria sanctori presenta un patrón de reproducción típico en especies de zonas templadas. Estos resultados pueden ser útiles para gestionar las capturas actuales de H. sanctori en el Mediterráneo y en el caso de que una pesquería específica empezase en la región oriental del Atlántico

    Seasonal analysis of the atmosphere during five years by using microwave radiometry over a mid-latitude site

    This work focuses on the analysis of the seasonal cycle of temperature and relative humidity (RH) profiles and integrated water vapor (IWV) obtained from microwave radiometer (MWR) measurements over the mid-latitude city of Granada, southern Spain. For completeness the study, the maximum atmospheric boundary layer height (ABLHmax) is also included. To this end, we have firstly characterized the HATPRO-RPG MWR errors using 55 co-located radiosondes (RS) by means of the mean-bias (biasbar) profile and the standard deviation (SDbias) profile classified under all-weather conditions and cloud-free conditions. This characterization pointed out that temperature from HATPRO-MWR presents a very low biasbar respects RS mostly below 2.0 km agl, ranging from positive to negative values under all-weather conditions (from 1.7 to -0.4 K with SDbias up to 3.0 K). Under cloud-free conditions, the bias was very similar to that found under all-weather conditions (1.8 to -0.4 K) but with smaller SDbias (up to 1.1 K). The same behavior is also seen in this lower part (ground to 2.0 km agl) for RH. Under all-weather conditions, the mean RH bias ranged from 3.0 to -4.0% with SDbias between 10 and 16.3% while under cloud-free conditions the bias ranged from 2.0 to -0.4% with SDbias from 0.5 to 13.3%. Above 2.0 km agl, the SDbias error increases considerably up to 4 km agl (up to -20%), and then decreases slightly above 7.0 km agl (up to -5%). In addition, IWV values from MWR were also compared with the values obtained from the integration of RS profiles, showing a better linear fit under cloud-free conditions (R2 = 0.96) than under all-weather conditions (R2 = 0.82). The mean bias under cloud-free conditions was -0.80 kg/m2 while for all-weather conditions it was -1.25 kg/m2. Thus, the SDbiasfor all the statistics (temperature, RH and IWV) of the comparison between MWR and RS presented higher values for all-weather conditions than for cloud-free conditions ones. It points out that the presence of clouds is a key factor to take into account when MWR products are used. The second part of this work is devoted to a seasonal variability analysis over five years, leading us to characterize thermodynamically the troposphere over our site. This city atmosphere presents a clear seasonal cycle where temperature, ABLHmax and IWV increase from winter to summer and decrease in autumn, meanwhile RH decreases along the warmer seasons. This city presents cold winters (mean daily maximum temperature: 10.6 ± 1.1 °C) and dry/hot summers (mean daily maximum temperature of 28.8 ± 0.9 °C and mean daily maximum of surface RH up to 55.0 ± 6.0%) at surface (680 m asl). Moreover, considering temporal trends, our study pointed out that only temperature and RH showed a linear increase in winters with a mean-rate of (0.5 ± 0.1) °C/year and (3.4 ± 1.7) %/year, respectively, from ground to 2.0 km agl, meanwhile IWV presented a linear increase of 1.0 kg·m-2/year in winters, 0.78 kg·m-2/year in summers and a linear decrease in autumns of -0.75 kg·m-2/year.Andalusia Regional Government through project P12-RNM-2409Spanish Ministry of Economy and Competitiveness through projects CGL2013-45410-R, CGL2015-73250-JIN and CGL2016-81092-RJuan de la Cierva grant IJCI-2016-3000

    Piloting the informed health choices resources in Barcelona primary schools: A mixed methods study

    IntroductionThe main objective of the Informed Health Choices (IHC) project is to teach people to assess treatment claims and make informed health choices. For this purpose, the IHC learning resources were developed for primary school children. The aim of this study is to explore students' and teachers' experience when using the IHC resources in primary schools in Barcelona (Spain). MethodsWe conducted a mixed methods study for piloting the IHC resources in a convenience sample of primary schools in Barcelona. The intervention included a workshop with teachers, and nine lessons with students. We collected data using multiple approaches. We performed quantitative and qualitative analyses, and integrated the findings in a joint display. Finally, we formulated recommendations for using the IHC resources in this setting. ResultsTwo schools, with a total of 143 students in 4th and 5th grade and six teachers, participated in the study. One school followed the suggested IHC teaching plan and competed all the lessons; the other school modified the plan substantially and did not complete all the lessons. Overall, students and teachers from both schools understood, were interested in, and were able to apply the content of the lessons. During the lessons, the textbook was useful for students; nevertheless, for the teachers, the usefulness of the IHC resources was variable. Teachers adapted the IHC resources to increase student participation and used Information and Communications Technologies tools. We observed more facilitators than barriers to teach the lessons. The teachers suggested some ideas to improve the lessons based on activities they developed and implemented. The integration analysis showed great convergence of the quantitative and qualitative findings. We propose seven recommendations for using the IHC resources in this setting. ConclusionsStudents and teachers from primary schools in Barcelona showed a positive experience when using IHC resources; however, these resources should be adapted to promote classroom participation

    EFFECT OF THE SUPPLEMENTATION OF ORGANIC ACIDS ON PRODUCTIVE PARAMETERS IN BROILERS

    Se evaluó el efecto de la suplementación de una mezcla de ácidos orgánicos y sus sales sobre los parámetros productivos en pollos de engorde. Se utilizaron 333 pollos machos de un día de edad de la línea Cobb-Vantress 500, divididos en tres tratamientos con tres repeticiones por tratamiento: T1, dieta con antibiótico Zinc Bacitracina; T2, dieta con ácidos orgánicos, y T3, tratamiento control, dieta sin promotor de crecimiento. A los 42 días de edad, la conversión alimenticia de T2 fue 5.2% menor que T3 (p<0.05); sin embargo, no se observaron diferencias estadísticas entre tratamientos por efecto del peso corporal, ganancia de peso, consumo de alimento, porcentaje de mortalidad e índice de eficiencia productiva. Los resultados permiten concluir que los ácidos orgánicos pueden remplazar eficientemente a los promotores de crecimiento tipo antibióticos en la alimentación de las aves.The objective of the study was to evaluate the effect of a mixture of organic acids and their salts on the productive parameters in broilers. A total of 133 1-day-old male chicks of the Cobb Vantress 500 Line were distributed in three treatments with three repetitions per treatment: T1, diet with Zinc-Bacitracin; T2, diet containing organic acids; and T3, control treatment, diet without growth promoter. At 42 days of age, feed conversion in T2 was 5.2% less than in T3 (p<0.05); however, no statistical differences were observed between treatments regarding body weight, body weight gain, feed consumption, mortality rate and productive efficiency index.     Nuclear PDCD4 Expression Predicts Good Clinical Outcome in Luminal A-Like and Luminal B-Like Breast Cancer Subtypes

    Hormone receptor-positive (HR+, estrogen and/or progesterone receptor-positive) and HER2-negative breast cancer (BC) subtype is a biologically heterogeneous entity that comprises 70% of BCs. This subtype includes both luminal (Lum) A- and B-like subtypes, which have differences in prognosis and sensitivity to endocrine therapies. The development of biomarkers guiding treatment decisions in these settings is required. Tumor suppressor PDCD4 (programmed cell death 4), which can be found both in the nucleus (NPDCD4) or the cytoplasm (CPDCD4), inhibits tumor growth and metastasis, and its loss is associated with poor prognosis in solid tumors. To explore the clinical relevance of PDCD4 in BC, we analyzed its expression by immunohistochemistry in a cohort of 619 patients with primary invasive BC. We found that 34.7% of patients showed NPDCD4 and 21.3% showed CPDCD4. NPDCD4 positivity, but not CPDCD4, was associated with lower clinical stage (P = 0.0003), with presence of more differentiated tumors (P = 6.4x10-6), and with estrogen and progesterone receptor (PR) expression (P = 9.2x10-9 and P = 2.8x10-9, respectively). Kaplan-Meier analysis revealed that NPDCD4 expression was associated with a longer overall survival (OS) and disease-free survival (DFS) in LumA-like (P = 0.008 and P = 0.028, respectively) and LumB-like (P = 0.004 and P = 0.012, respectively) subtypes. Interestingly, patients with LumB-like tumors displaying NPDCD4 presented estimated OS and DFS rates similar to the ones observed in patients with LumA-like tumors also expressing NPDCD4, indicating that its presence improves the clinical outcome of LumB-like patients. Multivariate Cox regression analysis identified NPDCD4 as an independent predictor of good clinical outcome in both LumA-like (HR: 0.45, 95% CI 0.22-0.96, P = 0.038) and LumB-like (HR: 0.28, 95% CI 0.10-0.80, P = 0.018) subtypes. We validated our results by in silico analysis using expression data from the METABRIC cohort. Bioinformatics analysis of BC cells from the Cancer Cell Line Encyclopedia revealed a positive correlation between PDCD4 and PR expression (P = 0.015). Since LumB-like tumors present a higher risk of resistance to endocrine therapy and both PR and PDCD4 levels in this subtype are lower than in the LumA-like one, we postulated that the presence of PR may modulate PDCD4 expression. Silencing of PR expression in HR+ cells decreased PDCD4 protein levels while reconstitution of PR in a PR-null cell line increased them, confirming PR requirement for PDCD4 modulation. In line with PDCD4 physiological function, its knockdown increased cell migration capability of HR+ BC cells, whereas its restoration led to a decrease in cell migration of HR-negative BC models. Our findings identified NPDCD4 positivity as a novel biomarker of clinical outcome in LumA- and B-like subtypes and revealed PDCD4 reconstitution as a novel therapeutic strategy in BC.Fil: Madera, Santiago.
