Role and Status of Biomarkers in Technostress Research: A Systematic Review
Pawan Kumar Mishra, Martina RaÅ¡ticov󡾬ulty of Business and Economics, Mendel University in Brno, Brno, CzechiaCorrespondence: Pawan Kumar Mishra, Email [email protected]: The revolution in technology has impacted the work and personal lives of human beings greatly. While it has introduced the mankind to a more comfortable life, it has brought in the stress too in the form of technostress, the situation where a person fails to cope up with the ever-advancing technology and experiences stress symptoms. The increasing intensity of technostress calls for more research on technostress diving deeper into the causes and coping mechanisms. However, technostress research requires successful and reliable assessment of stress. It has been observed in recent years that biomarkers such as cortisol and salivary alpha amylase are reliable indicators of stress. There are several reports where the researchers have used questionnaires and surveys to assess the technostress, but the number of studies using biomarkers for technostress assessment is limited. It has been established that biomarker assessment is an important complement to the surveys to study the technostress. Here, we summarize the important studies done on technostress using the biomarkers along with the rationale of using these biomarkers. Keywords: technostress, biomarkers, cortisol, skin conductance, salivary alpha amylase, heart rate variabilit
Genomic DNA isolation from Artemisia species grown in cold desert high altitude of India
Artemisia which produces a large number of secondary metabolites is naturally found in cold desert high altitude environment of India. Secondary metabolites such as alkaloids, flavonoids, phenols, polysaccharides and terpenes represent a significant barrier to the extraction of pure genomic DNA. Thus, in this study, the DNA extraction protocol to extract pure genomic DNA from different Artemisia species was tailored. The protocol was based on the CTAB method with slight modifications. In the study, 1.6 M NaCl, 2% cetyltrimethyl ammonium bromide (CTAB), 3% polyvinylpyrrolidone (PVP) and 0.5% β-mercaptoethanol was used in the extraction buffer. The incubation period was kept for 1 h at 65°C with one-tenth of the volume of warm (55°C) 10% CTAB solution during the extraction process. This study described a reliable protocol for extracting good quality and optimum amount of DNA from Artemisia species suitable for PCR analysis.Key words: Artemisia, genomic DNA isolation, PCR amplification, secondary metabolites

Bio-Inspired Preparation of Clay–Hexacyanoferrate Composite Hydrogels as Super Adsorbents for Cs+
A facile and low-cost fabrication route, inspired by the adhesive proteins secreted by mussels, has been developed to prepare a clay-based composite hydrogel (DHG(Cu)) containing hexacyanoferrate (HCF) nanoparticles for the selective removal of Cs+ from contaminated water. Initially, montmorillonite was exfoliated prior to coating with a thin layer of polydopamine (PDOPA) via the self-polymerization of dopamine. Mixing the composite (D-clay) with the HCF precursor, followed by the addition of copper ions, led to the self-assembly of the polymer-coated exfoliated clay nanosheets into a three-dimensional network and in situ growth of KCuHCF nanoparticles embedded within the gel structure. Analytical characterization verified the fabrication route and KCuHCF immobilization by a copper–ligand complexation. Rheology testing revealed the composite hydrogel to be elastic under low strain and exhibited reversible, self-healing behavior following high strain deformation, providing a good retention of KCuHCF nanoparticles in the membrane. The adsorbent DHG(Cu) showed a superior Cs+ adsorption capacity (∼173 mg/g), with the performance maintained over a wide pH range, and an excellent selectivity for Cs+ when dispersed in seawater at low concentrations of 0.2 ppm. On the basis of its excellent mechanico-chemical properties, the fabricated hydrogel was tested as a membrane in column filtration, showing excellent removal of Cs+ from Milli-Q water and seawater, with the performance only limited by the fluid residence time. For comparison, the study also considered other composite hydrogels, which were fabricated as intermediates of DHG(Cu) or fabricated with Fe3+ as the cross-linker and reactant for HCF nanoparticle synthesis
PSO-GWO Optimized Fractional Order PID Based Hybrid Shunt Active Power Filter for Power Quality Improvements
This paper presents a Hybrid Shunt Active Power Filter (HSAPF) optimized by hybrid Particle Swarm Optimization-Grey Wolf Optimization (PSO-GWO) and Fractional Order Proportional-Integral-Derivative Controller (FOPIDC) for reactive power and harmonic compensation under balance and unbalance loading conditions. Here, the parameters of FOPID controller are tuned by PSO-GWO technique to mitigate the harmonics. Comparing Passive with Active Filters, the former is tested to be bulky and design is complex and the later is not cost effective for high rating. Hence, a hybrid structure of shunt active and passive filter is designed using MATLAB/Simulink and in real time experimental set up. The compensation process for shunt active filter is different from predictable methods such as (p-q) or (id-i ) theory, in which only the source current is to be sensed. The performance of the proposed controller is tested under different operating conditions such as steady and transient states and indices like Total Harmonic Distortion (THD), Input Power Factor (IPF), Real Power (P) and Reactive Power (Q) are estimated and compared with that of other controllers. The parameters of FOPIDC and Conventional PID Controller (CPIDC) are optimized by the techniques such as PSO, GWO and hybrid PSO-GWO. The comparative simulation/experiment results reflect the better performance of PSO-GWO optimized FOPIDC based HSAPF with respect to PSO/GWO optimized FOPIDC/CPIDC based HSAPF under different operating conditions.
Spectroscopic investigation of quantum confinement effects in ion implanted silicon-on-sapphire films
Crystalline Silicon-on-Sapphire (SOS) films were implanted with boron (B)
and phosphorous (P) ions. Different samples, prepared by varying the ion
dose in the range to 5 x and ion energy in the range
150-350 keV, were investigated by the Raman spectroscopy, photoluminescence
(PL) spectroscopy and glancing angle x-ray diffraction (GAXRD). The Raman
results from dose dependent B implanted samples show red-shifted and
asymmetrically broadened Raman line-shape for B dose greater than
ions cm. The asymmetry and red shift in the Raman line-shape is
explained in terms of quantum confinement of phonons in silicon nanostructures
formed as a result of ion implantation. PL spectra shows size dependent visible
luminescence at 1.9 eV at room temperature, which confirms the presence
of silicon nanostructures. Raman studies on P implanted samples were also
done as a function of ion energy. The Raman results show an amorphous top SOS
surface for sample implanted with 150 keV P ions of dose 5 x ions
cm. The nanostructures are formed when the P energy is increased to
350 keV by keeping the ion dose fixed. The GAXRD results show consistency with
the Raman results.Comment: 9 Pages, 6 Figures and 1 Table, \LaTex format To appear in

Misregulation of cell cycle-dependent methylation of budding yeast CENP-A contributes to chromosomal instability.
Centromere (CEN) identity is specified epigenetically by specialized nucleosomes containing evolutionarily conserved CEN-specific histone H3 variant CENP-A (Cse4 in Saccharomyces cerevisiae, CENP-A in humans), which is essential for faithful chromosome segregation. However, the epigenetic mechanisms that regulate Cse4 function have not been fully defined. In this study, we show that cell cycle-dependent methylation of Cse4-R37 regulates kinetochore function and high-fidelity chromosome segregation. We generated a custom antibody that specifically recognizes methylated Cse4-R37 and showed that methylation of Cse4 is cell cycle regulated with maximum levels of methylated Cse4-R37 and its enrichment at the CEN chromatin occur in the mitotic cells. Methyl-mimic cse4-R37F mutant exhibits synthetic lethality with kinetochore mutants, reduced levels of CEN-associated kinetochore proteins and chromosome instability (CIN), suggesting that mimicking the methylation of Cse4-R37 throughout the cell cycle is detrimental to faithful chromosome segregation. Our results showed that SPOUT methyltransferase Upa1 contributes to methylation of Cse4-R37 and overexpression of UPA1 leads to CIN phenotype. In summary, our studies have defined a role for cell cycle-regulated methylation of Cse4 in high-fidelity chromosome segregation and highlight an important role of epigenetic modifications such as methylation of kinetochore proteins in preventing CIN, an important hallmark of human cancers
