    Vacuum Alignment in SUSY A4 Models

    In this note we discuss the vacuum alignment in supersymmetric models with spontaneously broken flavour symmetries in the presence of soft supersymmetry (SUSY) breaking terms. We show that the inclusion of soft SUSY breaking terms can give rise to non-vanishing vacuum expectation values (VEVs) for the auxiliary components of the flavon fields. These non-zero VEVs can have an important impact on the phenomenology of this class of models, since they can induce an additional flavour violating contribution to the sfermion soft mass matrix of right-left (RL) type. We carry out an explicit computation in a class of SUSY A4 models predicting tri-bimaximal mixing in the lepton sector. The flavour symmetry breaking sector is described in terms of flavon and driving supermultiplets. We find non-vanishing VEVs for the auxiliary components of the flavon fields and for the scalar components of the driving fields which are of order m_{SUSY} x and m_{SUSY}, respectively. Thereby, m_{SUSY} is the generic soft SUSY breaking scale which is expected to be around 1 TeV and is the VEV of scalar components of the flavon fields.     Multigrid algorithms for hp-version interior penalty discontinuous Galerkin methods on polygonal and polyhedral meshes

    In this paper we analyze the convergence properties of two-level and W-cycle multigrid solvers for the numerical solution of the linear system of equations arising from hp-version symmetric interior penalty discontinuous Galerkin discretizations of second-order elliptic partial differential equations on polygonal/polyhedral meshes. We prove that the two-level method converges uniformly with respect to the granularity of the grid and the polynomial approximation degree p, provided that the number of smoothing steps, which depends on p, is chosen sufficiently large. An analogous result is obtained for the W-cycle multigrid algorithm, which is proved to be uniformly convergent with respect to the mesh size, the polynomial approximation degree, and the number of levels, provided the latter remains bounded and the number of smoothing steps is chosen sufficiently large. Numerical experiments are presented which underpin the theoretical predictions; moreover, the proposed multilevel solvers are shown to be convergent in practice, even when some of the theoretical assumptions are not fully satisfied

Automating this task is an important step in reducing the financial cost associated with moderation, but the majority of automated approaches strictly based on message content are highly vulnerable to intentional obfuscation. In this paper, we discuss methods for extracting conversational networks based on raw multi-participant chat logs, and we study the contribution of graph features to a classification system that aims to determine if a given message is abusive. The conversational graph-based system yields unexpectedly high performance , with results comparable to those previously obtained with a content-based approach

    Intramuscular Immunisation with Chlamydial Proteins Induces Chlamydia trachomatis Specific Ocular Antibodies.

    BACKGROUND: Ocular infection with Chlamydia trachomatis can cause trachoma, which is the leading cause of blindness due to infection worldwide. Despite the large-scale implementation of trachoma control programmes in the majority of countries where trachoma is endemic, there remains a need for a vaccine. Since C. trachomatis infects the conjunctival epithelium and stimulates an immune response in the associated lymphoid tissue, vaccine regimens that enhance local antibody responses could be advantageous. In experimental infections of non-human primates (NHPs), antibody specificity to C. trachomatis antigens was found to change over the course of ocular infection. The appearance of major outer membrane protein (MOMP) specific antibodies correlated with a reduction in ocular chlamydial burden, while subsequent generation of antibodies specific for PmpD and Pgp3 correlated with C. trachomatis eradication. METHODS: We used a range of heterologous prime-boost vaccinations with DNA, Adenovirus, modified vaccinia Ankara (MVA) and protein vaccines based on the major outer membrane protein (MOMP) as an antigen, and investigated the effect of vaccine route, antigen and regimen on the induction of anti-chlamydial antibodies detectable in the ocular lavage fluid of mice. RESULTS: Three intramuscular vaccinations with recombinant protein adjuvanted with MF59 induced significantly greater levels of anti-MOMP ocular antibodies than the other regimens tested. Intranasal delivery of vaccines induced less IgG antibody in the eye than intramuscular delivery. The inclusion of the antigens PmpD and Pgp3, singly or in combination, induced ocular antigen-specific IgG antibodies, although the anti-PmpD antibody response was consistently lower and attenuated by combination with other antigens. CONCLUSIONS: If translatable to NHPs and/or humans, this investigation of the murine C. trachomatis specific ocular antibody response following vaccination provides a potential mouse model for the rapid and high throughput evaluation of future trachoma vaccines

    Scalable Full Flow with Learned Binary Descriptors

    We propose a method for large displacement optical flow in which local matching costs are learned by a convolutional neural network (CNN) and a smoothness prior is imposed by a conditional random field (CRF). We tackle the computation- and memory-intensive operations on the 4D cost volume by a min-projection which reduces memory complexity from quadratic to linear and binary descriptors for efficient matching. This enables evaluation of the cost on the fly and allows to perform learning and CRF inference on high resolution images without ever storing the 4D cost volume. To address the problem of learning binary descriptors we propose a new hybrid learning scheme. In contrast to current state of the art approaches for learning binary CNNs we can compute the exact non-zero gradient within our model. We compare several methods for training binary descriptors and show results on public available benchmarks.Comment: GCPR 201

    Exploration of a potent PI3 kinase/mTOR inhibitor as a novel anti-fibrotic agent in IPF

    © 2016 BMJ Publishing Group Ltd & British Thoracic Society.Rationale Idiopathic pulmonary fibrosis (IPF) is the most rapidly progressive and fatal of all fibrotic conditions with no curative therapies. Common pathomechanisms between IPF and cancer are increasingly recognised, including dysfunctional pan-PI3 kinase (PI3K) signalling as a driver of aberrant proliferative responses. GSK2126458 is a novel, potent, PI3K/mammalian target of rapamycin (mTOR) inhibitor which has recently completed phase I trials in the oncology setting. Our aim was to establish a scientific and dosing framework for PI3K inhibition with this agent in IPF at a clinically developable dose. Methods We explored evidence for pathway signalling in IPF lung tissue and examined the potency of GSK2126458 in fibroblast functional assays and precision-cut IPF lung tissue. We further explored the potential of IPF patient-derived bronchoalveolar lavage (BAL) cells to serve as pharmacodynamic biosensors to monitor GSK2126458 target engagement within the lung. Results We provide evidence for PI3K pathway activation in fibrotic foci, the cardinal lesions in IPF. GSK2126458 inhibited PI3K signalling and functional responses in IPF-derived lung fibroblasts, inhibiting Akt phosphorylation in IPF lung tissue and BAL derived cells with comparable potency. Integration of these data with GSK2126458 pharmacokinetic data from clinical trials in cancer enabled modelling of an optimal dosing regimen for patients with IPF. Conclusions Our data define PI3K as a promising therapeutic target in IPF and provide a scientific and dosing framework for progressing GSK2126458 to clinical testing in this disease setting.     The Golden Ratio Prediction for the Solar Angle from a Natural Model with A5 Flavour Symmetry

    We formulate a consistent model predicting, in the leading order approximation, maximal atmospheric mixing angle, vanishing reactor angle and tan {\theta}_12 = 1/{\phi} where {\phi} is the Golden Ratio. The model is based on the flavour symmetry A5 \times Z5 \times Z3, spontaneously broken by a set of flavon fields. By minimizing the scalar potential of the theory up to the next-to-leading order in the symmetry breaking parameter, we demonstrate that this mixing pattern is naturally achieved in a finite portion of the parameter space, through the vacuum alignment of the flavon fields. The leading order approximation is stable against higher-order corrections. We also compare our construction to other models based on discrete symmetry groups.Comment: 28 pages, 2 figures.     A novel stepwise micro-TESE approach in non obstructive azoospermia

    Background: The purpose of the study was to investigate whether micro-TESE can improve sperm retrieval rate (SRR) compared to conventional single TESE biopsy on the same testicle or to contralateral multiple TESE, by employing a novel stepwise micro-TESE approach in a population of poor prognosis patients with non-obstructive azoospermia (NOA). Methods: Sixty-four poor prognosis NOA men undergoing surgical testicular sperm retrieval for ICSI, from March 2007 to April 2013, were included in this study. Patients inclusion criteria were a) previous unsuccessful TESE, b) unfavorable histology (SCOS, MA, sclerahyalinosis), c) Klinefelter syndrome. We employed a stepwise micro-TESE consisting three-steps: 1) single conventional TESE biopsy; 2) micro-TESE on the same testis; 3) contralateral multiple TESE. Results: SRR was 28.1 % (18/64). Sperm was obtained in both the initial single conventional TESE and in the following micro-TESE. The positive or negative sperm retrieval was further confirmed by a contralateral multiple TESE, when performed. No significant pre-operative predictors of sperm retrieval, including patients’ age, previous negative TESE or serological markers (LH, FSH, inhibin B), were observed at univariate or multivariate analysis. Micro-TESE (step 2) did not improve sperm retrieval as compared to single TESE biopsy on the same testicle (step 1) or multiple contralateral TESE (step 3). Conclusions: Stepwise micro-TESE could represent an optimal approach for sperm retrieval in NOA men. In our view, it should be offered to NOA patients in order to gradually increase surgical invasiveness, when necessary.
