Integrative functional analysis uncovers metabolic differences between Candida species
Candida species are a dominant constituent of the human mycobiome and associated with the development of several diseases. Understanding the Candida species metabolism could provide key insights into their ability to cause pathogenesis. Here, we have developed the BioFung\ua0database, providing an efficient annotation of protein-encoding genes. Along, with BioFung, using carbohydrate-active enzyme (CAZymes) analysis, we have uncovered core and accessory features across Candida species demonstrating plasticity, adaption to the environment and acquired features. We show a greater importance of amino acid metabolism, as functional analysis revealed that all Candida species can employ amino acid metabolism. However, metabolomics revealed that only a specific cluster of species (AGAu species-C. albicans, C. glabrata and C. auris) utilised amino acid metabolism including arginine, cysteine, and methionine metabolism potentially improving their competitive fitness in pathogenesis. We further identified critical metabolic pathways in the AGAu cluster with biomarkers and anti-fungal target potential in the CAZyme profile, polyamine, choline and fatty acid biosynthesis pathways. This study, combining genomic analysis, and validation with gene expression and metabolomics, highlights the metabolic diversity with AGAu species that underlies their remarkable ability to dominate they mycobiome and cause disease
Methylation of the Gpat2 promoter regulates transient expression during mouse spermatogenesis
Spermatogenesis is a highly regulated process that involves both mitotic and meiotic divisions, as well as cellular differentiation to yield mature spermatozoa from undifferentiated germinal stem cells. Although Gpat2 was originally annotated as encoding a glycerol-3-phosphate acyltransferase by sequence homology to Gpat1, GPAT2 is highly expressed in testis but not in lipogenic tissues and is not up-regulated during adipocyte differentiation. New data show that GPAT2 is required for the synthesis of piRNAs (piwi-interacting RNAs), a group of small RNAs that protect the germ cell genome from retrotransposable elements. In order to understand the relationship between GPAT2 and its role in the testis, we focused on Gpat2 expression during the first wave of mouse spermatogenesis. Gpat2 expression was analysed by qPCR (quantitative real-time PCR), in situ hybridization, immunohistochemistry and Western blotting. Gpat2 mRNA content and protein expression were maximal at 15 dpp (days post-partum) and were restricted to pachytene spermatocytes. To achieve this transient expression, both epigenetic mechanisms and trans-acting factors are involved. In vitro assays showed that Gpat2 expression correlates with DNA demethylation and histone acetylation and that it is up-regulated by retinoic acid. Epigenetic regulation by DNA methylation was confirmed in vivo in germ cells by bisulfite sequencing of the Gpat2 promoter. Consistent with the initiation of meiosis at 11 dpp, methylation decreased dramatically. Thus, Gpat2 is expressed at a specific stage of spermatogenesis, consistent with piRNA synthesis and meiosis I prophase, and its on–off expression pattern responds predominantly to epigenetic modifications
Proposed nomenclature for Pseudallescheria, Scedosporium and related genera
As a result of fundamental changes in the International Code of Nomenclature on the use of separate names for sexual and asexual stages of fungi, generic names of many groups should be reconsidered. Members of the ECMM/ISHAM working group on Pseudallescheria/Scedosporium infections herein advocate a novel nomenclature for genera and species in Pseudallescheria, Scedosporium and allied taxa. The generic names Parascedosporium, Lomentospora, Petriella, Petriellopsis, and Scedosporium are proposed for a lineage within Microascaceae with mostly Scedosporium anamorphs producing slimy, annellidic conidia. Considering that Scedosporium has priority over Pseudallescheria and that Scedosporium prolificans is phylogenetically distinct from the other Scedosporium species, some name changes are proposed. Pseudallescheria minutispora and Petriellidium desertorum are renamed as Scedosporium minutisporum and S. desertorum, respectively. Scedosporium prolificans is renamed as Lomentospora prolificans
O efeito da infecção por Schistosoma mansoni na morbidade infantil no Estado da Bahia, Brasil: I — Análise do nível ecológico
Esta investigação foi levada a efeito em 10 pequenas cidades do Estado da Bahia (Nordeste do Brasil), escolhidas através de um processo amostrai. O objetivo foi estudar as correlações entre as prevalências de hepatomegalia e esplenomegalia, em diferentes áreas, com a prevalência e a intensidade da infecção pelo S. mansoni nas mesmas áreas. Nossas conclusões são: a) a morbidade esquistossomótica (prevalências de hepatomegalia e esplenomegalia) foi diretamente correlacionada com a prevalência e a intensidade da infecção; b) a intensidade da infecção (medida pelo número de ovos nas fezes) mostrou ser um bom indicador do grau de morbidade, explicando a variação da morbidade entre as áreas melhor que a prevalência da infecção; c) a prevalência de esplenomegalia e o tamanho médio do fígado abaixo do rebordo costal na linha me dio-esternal são bom indicadores da prevalência e da intensidade da infecção pelo S. mansoni na comunidade. Nossas conclusões trazem a possibilidade da construção de equações de regressão definitivas entre indicadores de morbidade e indicadores de infecção, de tal forma que o valor de um possa ser usado para predizer o valor do outro
The search campaign to identify and Image the Philae Lander on the surface of comet 67P/Churyumov-Gerasimenko
On the 12th of November 2014, the Rosetta Philae Lander descended to make the first soft touchdown on the surface of a comet – comet 67P/Churyumov-
Gerasimenko. That soft touchdown did occur but due to the failure in the firing of its two harpoons, Philae bounced and travelled across the comet making contact with the surface twice more before finally landing in a shaded rocky location somewhere on the southern hemisphere of the comet. The search campaign, led by ESA, involved multiple teams across Europe with a wide range of techniques used in support of it. This search campaign would continue through 2015 where a prime candidate on the surface was identified and on into 2016 to end on the 2nd of September 2016 when a definitive and conclusive image was taken of the lander on the surface of the comet, confirming the prime candidate to indeed be Philae
