    La formació permanent del professorat d'educació infantil: elements de reflexió

    Las características específicas del colectivo de profesionales de la etapa de Educación infantil proyectan una dinámica muy activa que genera nuevas metodologías, estrategias y recursos para una intervención educativa de calidad, en una etapa de vital importancia para el correcto desarrollo infantil. Estas características han de tenerse en cuenta en la formación de formadores del profesorado de esta etapa educativa, para favorecer su desarrollo profesional. Son necesarios proyectos de formación alternativos que conecten los «ámbitos de educación» a partir de propuestas integradas: escuela, familia, instituciones locales y asociaciones. What are needed are projects of alternative education that connect up the “fields of education” in terms of the following: school, family, local institutions and associations. La dispersión y posible variabilidad de esta organización debe garantizar el contenido y calidad del aprendizaje. En el curso 2013-14 planteamos evaluar los resultados de las prácticas con un doble objetivo: valorar las competencias de los alumnos y si nuestra organización conseguía los objetivos prácticos. El instrumento fue la observación de la actuación del alumno en un entorno simulado al final del periodo práctico. Metodologia: - Participaron los 60 alumnos matriculados, 30 evaluadores (15 enfermeras y 15 anestesiólogos) y 12 coordinadores de rotaciones y de estaciones. - Diseño de 6 estaciones con contenido relevante para la formación, ejecución en 10 min seguido de 15 min de “debriefing”, plantillas de evaluación de habilidades clínicas y no clínicas y criterios acordados por los evaluadores. Las estaciones fueron valoración preanestésica, bloqueo de plexo, anestesia general, preparación de perfusiones electrónicas de fármacos, depresión respiratoria y shock anafiláctico. - Se entrenó a los instructores sobre contenido y finalidad de la prueba, con 2 reuniones prácticas..

    Evaluation of micromorphological changes in tooth enamel after mechanical and ultrafast laser preparation of surface cavities

    The aim of this in vitro study was to evaluate the morphological changes that occur in tooth enamel after mechanical instrumentation and after femtosecond laser irradiation with different parameters via light and scanning electron microscopy (SEM). Twelve totally impacted third molars were collected and sectioned to provide several cut surfaces. These surfaces were exposed to infrared (λ = 795 nm, 120 fs, 1-kHz repetition rate, maximum mean power 1 W) laser pulses and machined by means of a conventional mechanical technique. Two very different geometrical patterns were performed with femtosecond laser pulses: shallow rectangular cavities and deep cylindrical ones. The results of both machining procedures were examined using light and scanning electron microscopy. The SEM images show the femtosecond laser ability to produce high-precision cavities in tooth enamel. No signs of collateral damage, burning, melting, or cracks were observed despite the far different laser pulse energies used (ranging from 7 to 400 μJ), unlike what is seen with conventional mechanical techniques.     Is basal ultrasensitive measurement of calcitonin capable of substituting for the pentagastrin-stimulation test?

    OBJECTIVE: To evaluate a second-generation assay for basal serum calcitonin (CT) measurements compared with the pentagastrin-stimulation test for the diagnosis of inherited medullary thyroid carcinoma (MTC) and the follow-up of patients with MTC after surgery. Recent American Thyroid Association recommendations suggest the use of basal CT alone to diagnose and assess follow-up of MTC as the pentagastrin (Pg) test is unavailable in many countries. DESIGN: Multicentric prospective study. PATIENTS: A total of 162 patients with basal CT <10 ng/l were included: 54 asymptomatic patients harboured noncysteine \u27rearranged during transfection\u27 (RET) proto-oncogene mutations and 108 patients had entered follow-up of MTC after surgery. MEASUREMENT: All patients underwent basal and Pg-stimulated CT measurements using a second-generation assay with 5-ng/l functional sensitivity. RESULTS: Ninety-five per cent of patients with basal CT ≥ 5 ng/l and 25% of patients with basal CT <5 ng/l had a positive Pg-stimulation test (Pg CT >10 ng/l). Compared with the reference Pg test, basal CT ≥ 5 ng/l had 99% specificity, a 95%-positive predictive value but only 35% sensitivity (P < 0.0001). Overall, there were 31% less false-negative results using a 5-ng/l threshold for basal CT instead of the previously used 10-ng/l threshold. CONCLUSION: The ultrasensitive CT assay reduces the false-negative rate of basal CT measurements when diagnosing familial MTC and in postoperative follow-up compared with previously used assays. However, its sensitivity to detect C-cell disease remains lower than that of the Pg-stimulation test

We investigated immune-related gene expression patterns in liver tissues surrounding early-stage HCCs and chemopreventive agents that might alter these patterns to prevent liver tumorigenesis. Methods: We analyzed gene expression profiles of nontumor liver tissues from 392 patients with early-stage HCC (training set, N = 167 and validation set, N = 225) and liver tissue from patients with cirrhosis without HCC (N = 216, controls) to identify changes in expression of genes that regulate the immune response that could contribute to hepatocarcinogenesis. We defined 172 genes as markers for this deregulated immune response, which we called the immune-mediated cancer field (ICF). We analyzed the expression data of liver tissues from 216 patients with cirrhosis without HCC and investigated the association between this gene expression signature and development of HCC and outcomes of patients (median follow-up, 10 years). Human liver tissues were also analyzed by histology. C57BL/6J mice were given a single injection of diethylnitrosamine (DEN) followed by weekly doses of carbon tetrachloride to induce liver fibrosis and tumorigenesis. Mice were then orally given the multiple tyrosine inhibitor nintedanib or vehicle (controls); liver tissues were collected and histology, transcriptome, and protein analyses were performed. We also analyzed transcriptomes of liver tissues collected from mice on a choline-deficient high-fat diet, which developed chronic liver inflammation and tumors, orally given aspirin and clopidogrel or the anti-inflammatory agent sulindac vs mice on a chow (control) diet. Results: We found the ICF gene expression pattern in 50% of liver tissues from patients with cirrhosis without HCC and in 60% of nontumor liver tissues from patients with early-stage HCC. The liver tissues with the ICF gene expression pattern had 3 different features: increased numbers of effector T cells; increased expression of genes that suppress the immune response and activation of transforming growth factor β signaling; or expression of genes that promote inflammation and activation of interferon gamma signaling. Patients with cirrhosis and liver tissues with the immunosuppressive profile (10% of cases) had a higher risk of HCC (hazard ratio, 2.41; 95% confidence interval, 1.21-4.80). Mice with chemically induced fibrosis or diet-induced steatohepatitis given nintedanib or aspirin and clopidogrel down-regulated the ICF gene expression pattern in liver and developed fewer and smaller tumors than mice given vehicle. Conclusions: We identified an immune-related gene expression pattern in liver tissues of patients with early-stage HCC, called the ICF, that is associated with risk of HCC development in patients with cirrhosis. Administration of nintedanib or aspirin and clopidogrel to mice with chronic liver inflammation caused loss of this gene expression pattern and development of fewer and smaller liver tumors. Agents that alter immune regulatory gene expression patterns associated with carcinogenesis might be tested as chemopreventive agents in patients with cirrhosis

    Molecular portrait of high alpha-fetoprotein in hepatocellular carcinoma: implications for biomarker-driven clinical trials

    The clinical utility of serum alpha-fetoprotein (AFP) in patients with hepatocellular carcinoma (HCC) is widely recognised. However, a clear understanding of the mechanisms of AFP overexpression and the molecular traits of patients with AFP-high tumours are not known. We assessed transcriptome data, whole-exome sequencing data and DNA methylome profiling of 520 HCC patients from two independent cohorts to identify distinct molecular traits of patients with AFP-high tumours (serum concentration?>?400?ng/ml), which represents an accepted prognostic cut-off and a predictor of response to ramucirumab. Those AFP-high tumours (18% of resected cases) were characterised by significantly lower AFP promoter methylation (p?<?0.001), significant enrichment of progenitor-cell features (CK19, EPCAM), higher incidence of BAP1 oncogene mutations (8.5% vs 1.6%) and lower mutational rates of CTNNB1 (14% vs 30%). Specifically, AFP-high tumours displayed significant activation of VEGF signalling (p?<?0.001), which might provide the rationale for the reported benefit of ramucirumab in this subgroup of patients

    Rhizobium Promotes Non-Legumes Growth and Quality in Several Production Steps: Towards a Biofertilization of Edible Raw Vegetables Healthy for Humans

    The biofertilization of crops with plant-growth-promoting microorganisms is currently considered as a healthy alternative to chemical fertilization. However, only microorganisms safe for humans can be used as biofertilizers, particularly in vegetables that are raw consumed, in order to avoid sanitary problems derived from the presence of pathogenic bacteria in the final products. In the present work we showed that Rhizobium strains colonize the roots of tomato and pepper plants promoting their growth in different production stages increasing yield and quality of seedlings and fruits. Our results confirmed those obtained in cereals and alimentary oil producing plants extending the number of non-legumes susceptible to be biofertilized with rhizobia to those whose fruits are raw consumed. This is a relevant conclusion since safety of rhizobia for human health has been demonstrated after several decades of legume inoculation ensuring that they are optimal bacteria for biofertilization

    Phosphate solubilizers enhance NPK fertilizer use efficiency in rice and legume cultivation

    It has been reported that phosphate solubilizing bacteria (PSB) are the most promising bacteria among the plant growth promoting rhizobacteria (PGPR); which may be used as biofertilizers for plant growth and nutrient use efficiency. Moreover, these soil micro-organisms play a significant role in regulating the dynamics of organic matter decomposition and the availability of plant nutrients such as nitrogen (N), phosphorus (P), potassium (K) and other nutrients. Through this study, the management of nutrient use efficiency by the application of PSB was targeted in order to make the applied nutrients more available to the plants in the rice (Oryza sativa) and yardlong bean (Vigna unguiculata) cultivation. Results have shown that the treatments with PSB alone or in the form of consortia of compatible strains with or without the external application of chemical NPK gave more germination index (G. I.) from 2.5 to 5 in rice and 2.7 to 4.8 in bean seeds. They also showed a higher growth in both shoot and root length and a higher biomass as compared to the control. This gives us an idea about the potentiality of these PSB strains and their application in rice and yardlong bean cultivation to get a better harvest index. Their use will also possibly reduce the nutrient runoff or leaching and increase in the use efficiency of the applied fertilizers. Thus, we can conclude that the NPK uptake and management can be improved by the use of PSB in rice and yardlong bean cultivation, and their application may be much more beneficial in the agricultural field

    Enhancement of clover growth by inoculation of P-solubilizing fungi and arbuscular mycorrhizal fungi

    This study evaluated the synergism between several P-solubilizing fungi isolates and arbuscular mycorrhizal fungi to improve clover ( Trifolium pratense) growth in the presence of Araxá apatite. Clover was sown directly in plastic pots with 300g of sterilized washed sand, vermiculite and sepiolite 1:1:1 (v:v:v) as substrate, and grown in a controlled environment chamber. The substrate was fertilized with 3 g L-1 of Araxá apatite. A completely randomized design, in 8×2 factorial scheme (eight P-solubilizing fungi treatments with or without arbuscular mycorrhizal fungi)and four replicates were used. The P-solubilizing fungi treatments consisted of five Brazilian P-solubilizing fungi isolates (PSF 7, 9, 20, 21 and 22), two Spanish isolates ( Aspergillus niger and the yeast Yarowia lipolytica) and control (non-inoculated treatment). The greatest clover growth rate was recorded when Aspergillus niger and PSF 21 were co-inoculated with arbuscular mycorrhizal fungi. Aspergillus niger, PSF 7 and PSF 21 were the most effective isolates on increasing clover growth in the presence of arbuscular mycorrhizal fungi. Greater mycorrhizal colonization resulted in greater clover growth rate in most PSF treatments. PSF 7 was the best isolate to improve the establishment of mycorrhizal and rhizobia symbiosis.Este estudo avaliou o sinergismo entre diversos isolados defungos solubilizadores de fosfato e micorrízicos arbusculares para beneficiar o crescimento de trevo ( Trifolium pratense) na presença de apatita de Araxá. A cultura foi semeada diretamente em potes plásticos com 300 g de substrato esterilizado formado por areia lavada, vermiculita e sepiolita 1:1:1 (v:v:v) e cultivada em câmara climática. O substrato foi fertilizado com 3 g L-1 de apatita de Araxá. O experimento foi instalado em delineamento completamente casualizado, esquema fatorial 8×2 (oito tratamentos de inoculação de fungos solubilizadores de fosfato com ou sem fungos micorrízicos arbusculares) e quatro repetições. Os tratamentos de fungos solubilizadores de fosfato consistiram em cinco isolados brasileiros de fungos solubilizadores de fosfato (FSF 7, 9, 20, 21 e 22), dois isolados procedentes da Espanha ( Aspergillus niger e a levedura Yarowia lipolytica) e o controle (tratamento não inoculado). A maior taxa de crescimento da cultura foi obtida quando Aspergillus niger e FSF 21 foram co-inoculados com fungos micorrízicos arbusculares. Aspergillus niger, FSF 7 e o FSF 21 foram os isolados mais efetivos para incrementar o crescimento de trevo na presença de fungos micorrízicosarbusculares. A maior taxa de colonização micorrízica resultou em alta taxa de crescimento de trevo na maioria dos tratamentos com fungos solubilizadores de fosfato. O isolado FSF 7 foi o melhor para favorecer o estabelecimento das simbioses com fungos micorrízicos e com rizóbio
