    Characterization of the Lipopolysaccharide from a \u3cem\u3eRhizobium phaseoli\u3c/em\u3e Mutant that is Defective in Infection Thread Development

    The lipopolysaccharide (LPS) from a Rhizobium phaseoli mutant, CE109, was isolated and compared with that of its wild-type parent, CE3. A previous report has shown that the mutant is defective in infection thread development, and sodium dodecyl sulfate-polyacrylamide gel electrophoresis shows that it has an altered LPS (K. D. Noel, K. A. VandenBosch, and B. Kulpaca, J. Bacteriol. 168:1392-1462, 1986). Mild acid hydrolysis of the CE3 LPS released a polysaccharide and an oligosaccharide, PS1 and PS2, respectively. Mild acid hydrolysis of CE109 LPS released only an oligosaccharide. Chemical and immunochemical analyses showed that CE3-PS1 is the antigenic O chain of this strain and that CE109 LPS does not contain any of the major sugar components of CE3-PS1. CE109 oligosaccharide was identical in composition to CE3-PS2. The lipid A\u27s from both strains were very similar in composition, with only minor quantitative variations. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis of CE3 and CE109 LPSs showed that CE3 LPS separated into two bands, LPS I and LPS II, while CE109 had two bands which migrated to positions similar to that of LPS II. Immunoblotting with anti-CE3 antiserum showed that LPS I contains the antigenic O chain of CE3, PS1. Anti-CE109 antiserum interacted strongly with both CE109 LPS bands and CE3 LPS II and interacted weakly with CE3 LPS I. Mild-acid hydrolysis of CE3 LPS I, extracted from the polyacrylamide gel, showed that it contained both PS1 and PS2.     Histopathological spectrum of salivary gland lesions in Ajmer region, Rajasthan, India

    Background: There is a wide spectrum of salivary gland lesions with morphological and clinical diversity which makes it a difficult task for histopathological interpretation. Tumours of salivary glands are uncommon accounting for 3-10% of the total tumors of head and neck region and less than one percent of all tumours. The aim of this study was to recognize various histomorphological patterns of salivary gland lesions, their frequency, age, gender and site wise distribution.Methods: This study was carried out from June 2015 to May 2018 in the department of Pathology, JLN Medical College and associated Group of Hospitals, Ajmer. Total 121 cases of salivary gland lesions were included. Specimens were processed and stained by Hematoxylin and Eosin stain followed by histopathological examination.Results: Out of total 121 cases, 43.8% were non-neoplastic and 56.2% were neoplastic. In non-neoplastic lesions predominant was chronic sialadenitis (50.9%) followed by mucocele (28.3%) which commonly seen in submandibular gland (47.17%). Among neoplastic cases, 79.4% were benign and 20.6% were malignant lesions. Neoplastic lesions commonly were seen in parotid (75%). Pleomorphic adenoma was the commonest benign tumour (81.4%). Mucoepidermoid carcinoma was the most common malignant salivary tumour. Benign tumours were common in third and fourth decades, whereas malignant tumours were more common in fifth and sixth decades. Male predominance was seen in overall salivary gland lesions.Conclusions: Histopathological examination is mandatory in the diagnosis of salivary gland lesions because of their wide spectrum of histomorphology

    Secreted frizzled related protein 2 (Sfrp2) is the key Akt-mesenchymal stem cell-released paracrine factor mediating myocardial survival and repair

    Stem cell therapy has emerged as a promising tool for the treatment of a variety of diseases. Previously, we have shown that Akt-modified mesenchymal stem cells mediate tissue repair through paracrine mechanisms. Using a comprehensive functional genomic strategy, we show that secreted frizzled related protein 2 (Sfrp2) is the key stem cell paracrine factor that mediates myocardial survival and repair after ischemic injury. Sfrp2 is known to modulate Wnt signaling, and we demonstrate that cardiomyocytes treated with secreted frizzled related protein increase cellular beta-catenin and up-regulate expression of antiapoptotic genes. These findings reveal the key role played by Sfrp2 in mediating the paracrine effects of Akt-mesenchymal stem cells on tissue repair and identify modulation of Wnt signaling as a therapeutic target for heart disease

    Induction of Heme Oxygenase-1, Biliverdin Reductase and H-Ferritin in Lung Macrophage in Smokers with Primary Spontaneous Pneumothorax: Role of HIF-1α

    Few data concern the pathophysiology of primary spontaneous pneumothorax (PSP), which is associated with alveolar hypoxia/reoxygenation. This study tested the hypothesis that PSP is associated with oxidative stress in lung macrophages. We analysed expression of the oxidative stress marker 4-HNE; the antioxidant and anti-inflammatory proteins heme oxygenase-1 (HO-1), biliverdin reductase (BVR) and heavy chain of ferritin (H-ferritin); and the transcription factors controlling their expression Nrf2 and HIF-1alpha, in lung samples from smoker and nonsmoker patients with PSP (PSP-S and PSP-NS), cigarette smoke being a risk factor of recurrence of the disease.mRNA was assessed by RT-PCR and proteins by western blot, immunohistochemistry and confocal laser analysis. 4-HNE, HO-1, BVR and H-ferritin were increased in macrophages from PSP-S as compared to PSP-NS and controls (C). HO-1 increase was associated with increased expression of HIF-1alpha mRNA and protein in alveolar macrophages in PSP-S patients, whereas Nrf2 was not modified. To understand the regulation of HO-1, BVR and H-ferritin, THP-1 macrophages were exposed to conditions mimicking conditions in C, PSP-S and PSP-NS patients: cigarette smoke condensate (CS) or air exposure followed or not by hypoxia/reoxygenation. Silencing RNA experiments confirmed that HIF-1alpha nuclear translocation was responsible for HO-1, BVR and H-ferritin induction mediated by CS and hypoxia/reoxygenation.PSP in smokers is associated with lung macrophage oxidative stress. The concept of true power mainly depends on how correctly the issues like voltage sag and voltage swell are mitigated. Among all the power quality problems, like voltage sag andvoltage swell mentioned are of key importance. This is because of the most widely use of newly invented as well as the outdated sensitive equipments connected to the load. Sensitivity is the main cause of the above power quality problems and it cannot be eliminated completely as it has many other operating properties. So for solve voltage sag and swell problem caused by the sensitive equipments connected to the faulty loads solve by connect some other system like DVR. The occurrence of sag and swell varies with equipment, environment, process operations, desired control schemes etc. by referring IEEE standards, the power quality events will last only for small instants. Thus the real time detection becomes a really tough task and even a small instant variation will cause a series of problems to the process industry. The dynamic voltage restorer is a special type of power device used for providing consistent and reliable supply power to the load devices.     Tunable-Q Wavelet Transform Based Multivariate Sub-Band Fuzzy Entropy with Application to Focal EEG Signal Analysis

    This paper analyses the complexity of multivariate electroencephalogram (EEG) signals in different frequency scales for the analysis and classification of focal and non-focal EEG signals. The proposed multivariate sub-band entropy measure has been built based on tunable-Q wavelet transform (TQWT). In the field of multivariate entropy analysis, recent studies have performed analysis of biomedical signals with a multi-level filtering approach. This approach has become a useful tool for measuring inherent complexity of the biomedical signals. However, these methods may not be well suited for quantifying the complexity of the individual multivariate sub-bands of the analysed signal. In this present study, we have tried to resolve this difficulty by employing TQWT for analysing the sub-band signals of the analysed multivariate signal. It should be noted that higher value of Q factor is suitable for analysing signals with oscillatory nature, whereas the lower value of Q factor is suitable for analysing signals with non-oscillatory transients in nature. Moreover, with an increased number of sub-bands and a higher value of Q-factor, a reasonably good resolution can be achieved simultaneously in high and low frequency regions of the considered signals. Finally, we have employed multivariate fuzzy entropy (mvFE) to the multivariate sub-band signals obtained from the analysed signal. The proposed Q-based multivariate sub-band entropy has been studied on the publicly available bivariate Bern Barcelona focal and non-focal EEG signals database to investigate the statistical significance of the proposed features in different time segmented signals. Finally, the features are fed to random forest and least squares support vector machine (LS-SVM) classifiers to select the best classifier. Our method has achieved the highest classification accuracy of 84.67% in classifying focal and non-focal EEG signals with LS-SVM classifier. The proposed multivariate sub-band fuzzy entropy can also be applied to measure complexity of other multivariate biomedical signals
