    A novel programmable lysozyme-based lysis system in Pseudomonas putida for biopolymer production

    Indexación: Scopus; Web of Science.Cell lysis is crucial for the microbial production of industrial fatty acids, proteins, biofuels, and biopolymers. In this work, we developed a novel programmable lysis system based on the heterologous expression of lysozyme. The inducible lytic system was tested in two Gram-negative bacterial strains, namely Escherichia coli and Pseudomonas putida KT2440. Before induction, the lytic system did not significantly arrest essential physiological parameters in the recombinant E. coli (ECPi) and P. putida (JBOi) strain such as specific growth rate and biomass yield under standard growth conditions. A different scenario was observed in the recombinant JBOi strain when subjected to PHA-producing conditions, where biomass production was reduced by 25% but the mcl-PHA content was maintained at about 30% of the cell dry weight. Importantly, the genetic construct worked well under PHA-producing conditions (nitrogen-limiting phase), where more than 95% of the cell population presented membrane disruption 16 h post induction, with 75% of the total synthesized biopolymer recovered at the end of the fermentation period.     Discovering Rehabilitation trends in Spain: A bibliometric analysis

    The main purpose of this study is to offer an overview of the rehabilitation research area in Spain from 1970 to 2018 through a bibliometric analysis. Analysis of performance and a co-word science mapping analysis were conducted to highlight the topics covered. The software tool SciMAT was used to analyse the themes concerning their performance and impact measures. A total of 3,564 documents from the Web of Science were retrieved. Univ Deusto, Univ Rey Juan Carlos and Basque Foundation for Science are the institutions with highest relative priority.     Entrevistar : una experiencia de enseñanza de la entrevista en profundidad

    La siguiente presentación da cuenta de una propuesta diseñada y llevada a cabo durante dos cuatrimestres en la carrera de Ciencias de la Educación en el marco de un espacio curricular denominado “Seminario de Recolección y Análisis de datos". Está destinado a estudiantes del último ciclo de la carrera, especialmente a quienes están interesados en formarse como investigadores. Reconstruimos la experiencia desarrollando en cada etapa del proceso: la estrategia metodológica de la enseñanza, explicitamos las lecturas y actividades propuestas, y el sentido formativo de cada una de ellas al tiempo que introducimos posibles variaciones, discusiones y algunas apreciaciones que fuimos generando en el proceso. Asumimos una perspectiva socio antropológica basada en el enfoque teórico de Pierre Bourdieu y otros aportes específicamente metodológicos (Valles,1999; Blanchet,1989; Guber, 1991; Achili, 2005; García Salord, 2000) que ayudan a dar precisión al trabajo. Todo el proceso propone un doble desafío: por un lado, aprender a entrevistar y, por el otro, generar una práctica reflexiva de auto análisis que permita a cada estudiante conocerse un poco mejor en esta tarea. Promovemos un trabajo denso en términos teóricos y una experiencia subjetiva intensa; el diálogo entre ambas constituye de algún modo algo de ese “aprender a investigar investigando", factible en los marcos curriculares y normativos previstos.     Direct observation of melting in a 2-D superconducting vortex lattice

    Topological defects such as dislocations and disclinations are predicted to determine the twodimensional (2-D) melting transition. In 2-D superconducting vortex lattices, macroscopic measurements evidence melting close to the transition to the normal state. However, the direct observation at the scale of individual vortices of the melting sequence has never been performed. Here we provide step by step imaging through scanning tunneling spectroscopy of a 2-D system of vortices up to the melting transition in a focused-ion-beam nanodeposited W-based superconducting thin film. We show directly the transition into an isotropic liquid below the superconducting critical temperature. Before that, we find a hexatic phase, characterized by the appearance of free dislocations, and a smectic-like phase, possibly originated through partial disclination unbinding. These results represent a significant step in the understanding of melting of 2-D systems, with impact across several research fields, such as liquid crystal molecules, or lipids in membranes.Comment: Submitted to Nature Physic

    Investigation of the Mechanism of the Gene Regulation of OPG (Osteoprotegerin) by Cx43

    poster abstractThe main objective is to determine whether the gene regulation of OPG, osteoprotegerin, by Cx43, is at the promoter level. In a recent project, it was found that deletion in Cx43 from osteocytes resulted in a decreased OPG expression. Furthermore, it was found that deletion of Cx43 from osteocytes resulted in enhanced osteoclast differentiation. Osteoprotegerin (OPG), an osteoblast-secreted decoy receptor that modulates osteoclast formation could be directly controlled by Cx43 at the promoter binding sites of p53 and Sp1. Cx43 and OPG in turn are widely up regulated by Wnt, lipid-modified signaling proteins that influence cell proliferation, differentiation, and survival. The activation of Wnt signaling results in the binding of the transcription factor Tcf in gene promoters, which leads to increased gene expression. The investigation was carried out using reporter constructs in which the activation of the promoter resulted in the transcription of the enzyme luciferase. Luciferase activity, in turn, can be measured using a commercially available substrate that emits luminescence when luciferase is present. OPG-Luc and Tcf-Luc were grown in E. coli and purified using a kit from Qiagen. Transfection of OPG 1 and Tcf-Luc reporter constructs on MLO-Y4 osteocyte cells deleted for Cx43 (Cx43shRNA) and Cx37 (Cx37shRNA) was conducted after seeding of the cells a day in advance. For each cell line, regular and Lithium Chloride (to mimic the effects of Wnt) induced medium was used, and cells were cultured for 24h. From the assay, it was deemed that luciferase activity was higher in Wnt induced cells. OPG is a target of Wnt signaling downstream of the transcription factor Tcf. We therefore also measure Tcf-mediated transcription using a Tcf-luciferase construct. Expression of OPG-Luc and Tcf-Luc was higher in cell lines that are not silenced for Cx43 and Cx37. According to ANOVA test, the results did reach statistical significance. However, future trials will be conducted to mimic the results
