Physical mapping of chromosomes 3HS and 3DS
Relatório de estágio do mestrado em Ensino da Educação Física nos Ensinos Básico e Secundário, apresentado à Faculdade de Ciências do Desporto e Educação Física da Universidade de CoimbraO presente documento divide-se em duas partes: a reflexão dos aspetos relacionados com o estágio pedagógico e o desenvolvimento do tema-problema.
Numa primeira fase será dado enfoque à prática da docência, desenvolvida na escola E.B.2,3 Dr. Daniel de Matos. Serão apresentadas as expectativas iniciais em relação à prática, serão apresentadas e justificadas as opções metodológicas tomadas ao longo do estágio, serão relatadas as atividades desenvolvidas durante o estágio e será realçada a componente ética e profissional da profissão. De seguida será realizada uma justificação das opções tomadas. Para finalizar a primeira parte do presente trabalho será feita uma análise reflexiva sobre a prática pedagógica.
Na segunda parte temos o tema-problema e seu desenvolvimento, num documento que se pretende orientador da prática docente e auxiliar de como resolver comportamentos de desvio da tarefa por parte dos alunos. Servirá também para analisar e mostrar conclusões sobre o problema que a escola atual, enquanto organização enfrenta, a indisciplina.
Será efetuada uma pequena investigação, em que o objetivo é analisar a perceção dos docentes de uma escola pública sobre a gravidade dos comportamentos de indisciplina praticados.
O objetivo é chegar a alguma conclusão que possa ajudar/beneficiar os professores da escola na procura de estratégias e metodologias que possam minimizar o impacto deste tipo de comportamentos no normal funcionamento da aula.
This document is divided into two parts: a reflection of aspects related to teaching practice and development of the “theme-issue”.
Initially focus will be given to the practice of teaching, developed in E.B.2, 3 Dr. Daniel de Matos school. Initial expectations will be presented, as well as the methodological choices made along the stage and its reasons. The activities made during the internship will be reported and I will emphasize ethics and professional component of the teacher’s profession. Then I will do a justification of the choices made. To finish the first part of this work I will make a reflective analysis of teaching practice.
In the second part we have the “theme-issue” and its development, a document intended to be the guiding teaching practice and assist in solving the task behaviors of misuse by students. Will also serve to analyze and display conclusions about the problem that the current school, as an organization, faces, the indiscipline.
Will be performed a little investigation, in which the objective is to analyze the perceptions of teachers in a public school on the severity of disruptive behaviors committed.
The goal is to achieve some conclusion that can help / benefit school teachers in finding strategies and methodologies that can minimize the impact of such behaviors on normal class
Physical mapping of the short arm of chromosome 3 in aegilops tauschii and barley
A research program has been established to generate fine physical and genetic maps of wheat and barley chromosomes 3 and 7. In association with the International Wheat and Barley Genome Sequencing Consortia, the ultimate goal is to use these maps to produce a full genome sequence, target specific genes for isolation and analysis and investigate the recombinational behavior of the complex Triticeae chromosomes. We made a first physical map of chromosome 3S using the BAC library of Aegilops tauschii, the ancestor of D genome. The target region was the telomeric 20 cM of the short arm of Group 3 chromosomes, delineated by an X-ray induced deletion mutation called ph2a and estimated to be around 80 Mb in size. About 100 EST were anchored on the contigs by using information from previous studies of the deletion mutant, the physical map of 3BS, the wheat deletion bin map and the barley genetic map. The physical map on Ae tauschii and barley genetic maps were linked and used to calculate the recombination rate across the region. The maps allowed us to compare the organization of the genes along the chromosome 3S in the H, B and D-ancestor genomes. Deep brain stimulation of the subthalamic nucleus preferentially alters the translational profile of striatopallidal neurons in an animal model of Parkinson’s disease
Deep brain stimulation targeting the subthalamic nucleus (STN-DBS) is an effective surgical treatment for the motor symptoms of Parkinson’s disease (PD), the precise neuronal mechanisms of which both at molecular and network levels remain a topic of debate. Here we employ two transgenic mouse lines, combining translating ribosomal affinity purification (TRAP) with bacterial artificial chromosome expression (Bac), to selectively identify changes in translational gene expression in either Drd1a-expressing striatonigral or Drd2-expressing striatopallidal medium spiny neurons (MSNs) of the striatum following STN-DBS. 6-hydroxydopamine lesioned mice received either 5 days stimulation via a DBS electrode implanted in the ipsilateral STN or 5 days sham treatment (no stimulation). Striatal polyribosomal RNA was selectively purified from either Drd2 or Drd1a MSNs using the TRAP method and gene expression profiling performed. We identified 8 significantly altered genes in Drd2 MSNs (Vps33b, Ppp1r3c, Mapk4, Sorcs2, Neto1, Abca1, Penk1 and Gapdh) and 2 overlapping genes in Drd1a MSNs (Penk1 and Ppp1r3c) implicated in the molecular mechanisms of STN-DBS. A detailed functional analysis, using a further 728 probes implicated in STN-DBS, suggested an increased ability to receive excitation (mediated by increased dendritic spines, increased calcium influx and enhanced excitatory post synaptic potentials) accompanied by processes that would hamper the initiation of action potentials, transport of neurotransmitters from soma to axon terminals and vesicular release in Drd2-expressing MSNs. Finally, changes in expression of several genes involved in apoptosis as well as cholesterol and fatty acid metabolism were also identified. This increased understanding of the molecular mechanisms induced by STN-DBS may reveal novel targets for future non-surgical therapies for PD
