    A system to enrich for primitive streak-derivatives, definitive endoderm and mesoderm, from pluripotent cells in culture

    Two lineages of endoderm develop during mammalian embryogenesis, the primitive endoderm in the pre-implantation blastocyst and the definitive endoderm at gastrulation. This complexity of endoderm cell populations is mirrored during pluripotent cell differentiation in vitro and has hindered the identification and purification of the definitive endoderm for use as a substrate for further differentiation. The aggregation and differentiation of early primitive ectoderm-like (EPL) cells, resulting in the formation of EPL-cell derived embryoid bodies (EPLEBs), is a model of gastrulation that progresses through the sequential formation of primitive streak-like intermediates to nascent mesoderm and more differentiated mesoderm populations. EPL cell-derived EBs have been further analysed for the formation of definitive endoderm by detailed morphological studies, gene expression and a protein uptake assay. In comparison to embryoid bodies derived from ES cells, which form primitive and definitive endoderm, the endoderm compartment of embryoid bodies formed from EPL cells was comprised almost exclusively of definitive endoderm. Definitive endoderm was defined as a population of squamous cells that expressed Sox17, CXCR4 and Trh, which formed without the prior formation of primitive endoderm and was unable to endocytose horseradish peroxidase from the medium. Definitive endoderm formed in EPLEBs provides a substrate for further differentiation into specific endoderm lineages; these lineages can be used as research tools for understanding the mechanisms controlling lineage establishment and the nature of the transient intermediates formed. The similarity between mouse EPL cells and human ES cells suggests EPLEBs can be used as a model system for the development of technologies to enrich for the formation of human ES cell-derived definitive endoderm in the future.Sveltana Vassilieva, Hweee Ngee Goh, Kevin X.     Structural evaluation of concrete expanded polystyrene sandwich panels for slab applications

    Sandwich panels are being extensively and increasingly used in building construction because they are light in weight, energy efficient, aesthetically attractive and can be easily handled and erected. This paper presents a structural evaluation of Concrete-Expanded Polystyrene (CEPS) sandwich panels for slab applications using finite element modeling approach. CEPS panels are made of expanded polystyrene foam sandwiched between concrete skins. The use of foam in the middle of sandwich panel reduces the weight of the structure and also acts as insulation against thermal, acoustics and vibration. Applying reinforced concrete skin to both sides of panel takes the advantages of the sandwich concept where the reinforced concrete skins take compressive and tensile loads resulting in higher stiffness and strength and the core transfers shear loads between the faces. This research uses structural software Strand7, which is based on finite element method, to predict the load deformation behaviour of the CEPS sandwich slab panels. Non linear static analysis was used in the numerical investigations. Predicted results were compared with the existing experimental results to validate the numerical approach used

    A proposal for a scalable universal bosonic simulator using individually trapped ions

    We describe a possible architecture to implement a universal bosonic simulator (UBS) using trapped ions. Single ions are confined in individual traps, and their motional states represent the bosonic modes. Single-mode linear operators, nonlinear phase-shifts, and linear beam splitters can be realized by precisely controlling the trapping potentials. All the processes in a bosonic simulation, except the initialization and the readout, can be conducted beyond the Lamb-Dicke regime. Aspects of our proposal can also be applied to split adiabatically a pair of ions in a single trap

    Differential Forms and Wave Equations for General Relativity

    Recently, Choquet-Bruhat and York and Abrahams, Anderson, Choquet-Bruhat, and York (AACY) have cast the 3+1 evolution equations of general relativity in gauge-covariant and causal ``first-order symmetric hyperbolic form,'' thereby cleanly separating physical from gauge degrees of freedom in the Cauchy problem for general relativity. A key ingredient in their construction is a certain wave equation which governs the light-speed propagation of the extrinsic curvature tensor. Along a similar line, we construct a related wave equation which, as the key equation in a system, describes vacuum general relativity. Whereas the approach of AACY is based on tensor-index methods, the present formulation is written solely in the language of differential forms. Our approach starts with Sparling's tetrad-dependent differential forms, and our wave equation governs the propagation of Sparling's 2-form, which in the ``time-gauge'' is built linearly from the ``extrinsic curvature 1-form.'' The tensor-index version of our wave equation describes the propagation of (what is essentially) the Arnowitt-Deser-Misner gravitational momentum.Comment: REVTeX, 26 pages, no figures, 1 macr

    Extensions and block decompositions for finite-dimensional representations of equivariant map algebras

    Suppose a finite group acts on a scheme XX and a finite-dimensional Lie algebra g\mathfrak{g}. The associated equivariant map algebra is the Lie algebra of equivariant regular maps from XX to g\mathfrak{g}. The irreducible finite-dimensional representations of these algebras were classified in previous work with P. Senesi, where it was shown that they are all tensor products of evaluation representations and one-dimensional representations. In the current paper, we describe the extensions between irreducible finite-dimensional representations of an equivariant map algebra in the case that XX is an affine scheme of finite type and g\mathfrak{g} is reductive. This allows us to also describe explicitly the blocks of the category of finite-dimensional representations in terms of spectral characters, whose definition we extend to this general setting. Applying our results to the case of generalized current algebras (the case where the group acting is trivial), we recover known results but with very different proofs. For (twisted) loop algebras, we recover known results on block decompositions (again with very different proofs) and new explicit formulas for extensions. Finally, specializing our results to the case of (twisted) multiloop algebras and generalized Onsager algebras yields previously unknown results on both extensions and block decompositions.Comment: 41 pages; v2: minor corrections, formatting changed to match published versio

    Observed Recent Trends in Tropical Cyclone Rainfall Over Major Ocean Basins

    In this study, we use Tropical Rainfall Measuring Mission (TRMM) and Global Precipitation Climatology Project (GPCP) rainfall data together with historical storm track records to examine the trend of tropical cyclone (TC) rainfall in major ocean basins during recent decades (1980-2007). We find that accumulated total rainfall along storm tracks for all tropical cyclones shows a weak positive trend over the whole tropics. However, total rainfall associated with weak storms, and intense storms (Category 4-5) both show significant positive trends, while total rainfall associated with intermediate storms (Category1-3) show a significant negative trend. Storm intensity defined as total rain produced per unit storm also shows increasing trend for all storm types. Basin-wide, from the first half (1980-1993) to the second half (1994-2007) of the data period, the North Atlantic shows the pronounced increase in TC number and TC rainfall while the Northeast Pacific shows a significant decrease in all storm types. Except for the Northeast Pacific, all other major basins (North Atlantic, Northwest Pacific, Southern Oceans, and Northern Indian Ocean) show a significant increase in total number and rainfall amount in Category 4-5 storms. Overall, trends in TC rainfall in different ocean basins are consistent with long-term changes in the ambient large-scale environment, including SST, vertical wind shear, sea level pressure, mid-tropospheric humidity, and Maximum Potential Intensity (MPI). Notably the pronounced positive (negative) trend of TC rainfall in the North Atlantic (Northeast Pacific) appears to be related to the most (least) rapid increase in SST and MPI, and the largest decrease (increase) in vertical wind shear in the region, relative to other ocean basins
