Searching Data: A Review of Observational Data Retrieval Practices in Selected Disciplines
A cross-disciplinary examination of the user behaviours involved in seeking
and evaluating data is surprisingly absent from the research data discussion.
This review explores the data retrieval literature to identify commonalities in
how users search for and evaluate observational research data. Two analytical
frameworks rooted in information retrieval and science technology studies are
used to identify key similarities in practices as a first step toward
developing a model describing data retrieval
A convenient, optimized pipeline for isolation, fluorescence microscopy and molecular analysis of live single cells
BACKGROUND: Heterogeneity within cell populations is relevant to the onset and progression of disease, as well as development and maintenance of homeostasis. Analysis and understanding of the roles of heterogeneity in biological systems require methods and technologies that are capable of single cell resolution. Single cell gene expression analysis by RT-qPCR is an established technique for identifying transcriptomic heterogeneity in cellular populations, but it generally requires specialized equipment or tedious manipulations for cell isolation. RESULTS: We describe the optimization of a simple, inexpensive and rapid pipeline which includes isolation and culture of live single cells as well as fluorescence microscopy and gene expression analysis of the same single cells by RT-qPCR. We characterize the efficiency of single cell isolation and demonstrate our method by identifying single GFP-expressing cells from a mixed population of GFP-positive and negative cells by correlating fluorescence microscopy and RT-qPCR. CONCLUSIONS: Single cell gene expression analysis by RT-qPCR is a convenient means for investigating cellular heterogeneity, but is most useful when correlating observations with additional measurements. We demonstrate a convenient and simple pipeline for multiplexing single cell RT-qPCR with fluorescence microscopy which is adaptable to other molecular analyses
Illicit COVID-19 products online: A mixed-method approach for identifying and preventing online health risks

This is an open access article distributed under the terms of the Creative Commons Attribution License (CC BY), https://creativecommons.org/licenses/by/4.0/Aims: The COVID-19 pandemic triggered a demand for vaccines, cures, and the need of related documentation for travel, work and other purposes. Our project aimed to identify the illicit availability of such products across the Dark Web Markets (DWMs). Methods: A retrospective search for COVID-19 related products was carried out across 118 DWMs since the start of the pandemic (March 2020-October 2021). Data on vendors as well as advertised goods such as asking price, marketplace, listed date were collected and further validated through additional searches on the open web to verify the information relating to specific marketplaces. Both quantitative and qualitative methods were used for data analysis. Results: Forty-two listings of unlicenced COVID-19 cures and vaccination certificates were identified across 8 marketplaces sold by 25 vendors with significant variation in prices. The listings were found to be geographically specific and followed the progression of the pandemic in terms of availability. Correlations between vendor portfolios of COVID-19 products and variety of goods of other illicit nature such as illegal weaponry, medication/drugs of abuse also emerged from our analysis. Conclusion: This study is one of the first attempts to identify the availability of unlicenced COVID-19 products on DWMs. The easy accessibility to vaccines, fake test certificates and hypothetical/illegal cures poses serious health risks to (potential) buyers due to the uncontrolled nature of such products. It also exposes buyers to an unwanted contact with vendors selling a variety of other dangerous illicit goods. Further monitoring and regulatory responses should be implemented to protect the health and safety of citizens especially at times of global crisis.Peer reviewe
The Relationship of the Quality of Life and Activity Limitations of Persons with Orthopedic Disability
Walking difficulties influence both functional motor motion and fine motor motion, such as hand control. Included are conditions that make it hard to move, such as amputation, paralysis, cerebral palsy, stroke, multiple sclerosis, muscular dystrophy, arthritis, spinal cord injuries, and others. The study used WHOQOL-BREF-DIS and IMPACT-S to determine PWDs' quality of life and activity restrictions. It examines the link between quality of life and activity limitation, as well as discrimination, autonomy, and inclusion. Males have a higher quality of life, but females have more activity limitations. Discrimination and autonomy domains of WHOQOL-BREF-DIS are not connected with activity participation; inclusion and overall quality of life are. In conclusion, persons with orthopedic disabilities' level of inclusion and overall QOL in society, specifically their satisfaction with communicating with others, satisfaction with their ability to participate in social and local activities, and other people's acceptance and respect, are affected by their activity participation
Evaluation of GPT-3.5 and GPT-4 for supporting real-world information needs in healthcare delivery
Despite growing interest in using large language models (LLMs) in healthcare,
current explorations do not assess the real-world utility and safety of LLMs in
clinical settings. Our objective was to determine whether two LLMs can serve
information needs submitted by physicians as questions to an informatics
consultation service in a safe and concordant manner. Sixty six questions from
an informatics consult service were submitted to GPT-3.5 and GPT-4 via simple
prompts. 12 physicians assessed the LLM responses' possibility of patient harm
and concordance with existing reports from an informatics consultation service.
Physician assessments were summarized based on majority vote. For no questions
did a majority of physicians deem either LLM response as harmful. For GPT-3.5,
responses to 8 questions were concordant with the informatics consult report,
20 discordant, and 9 were unable to be assessed. There were 29 responses with
no majority on "Agree", "Disagree", and "Unable to assess". For GPT-4,
responses to 13 questions were concordant, 15 discordant, and 3 were unable to
be assessed. There were 35 responses with no majority. Responses from both LLMs
were largely devoid of overt harm, but less than 20% of the responses agreed
with an answer from an informatics consultation service, responses contained
hallucinated references, and physicians were divided on what constitutes harm.
These results suggest that while general purpose LLMs are able to provide safe
and credible responses, they often do not meet the specific information need of
a given question. A definitive evaluation of the usefulness of LLMs in
healthcare settings will likely require additional research on prompt
engineering, calibration, and custom-tailoring of general purpose models.Comment: 27 pages including supplemental informatio
