    Breeding season of the South American fur seal (Arctocephalus Australis, Otariidae: Carnivora): new data for establishing independent evolutionary histories?

    The South American fur seal (SAFS) is distributed from Peru (Pacific Ocean) to Uruguay (Atlantic Ocean). However, there is a section of coastline of about 2,300 km along the Chilean coast where no SAFS are recorded. Based on morphological comparisons 3 geographic forms have been reported (Peruvian, South Chilean, Atlantic), whereas preliminary genetic studies suggest the presence of 3 distinct genetic types (Peruvian, South Chile, Atlantic). However, both of these approaches are support only the existence of 2 independent evolutionary histories for SAFS (Peruvian, Atlantic), in which case, we would also expect to find some differences in the species’ life histories among these locals populations (e.g., in the timing of the breeding season). In this study, we compare the breeding seasons of SAFS at Punta Pichalo (19°36’S-Chile), Isla Guafo (43°33’S-Chile), and Isla de Lobos (35°01’S-Uruguay) with data published for Punta San Juan (15°21’S-Peru). The periods for establishing territories, pupping, and mating took place on similar dates at Isla Guafo and Isla de Lobos, but occurred about a month earlier at Punta Pichalo and Punta San Juan. Pupping peaked at Isla Guafo on 15-December, at Isla de Lobos on 17-December, and at Punta Pichalo on 27-November. These differences in timing were not related to demographic, oceanographic or photoperiod conditions of the breeding sites under study, neither is related with the latitudinal effects on the breeding phenology.     The value of selected in vitro and in silico methods to predict acute oral toxicity in a regulatory context: results from the European Project ACuteTox

    ACuteTox is a project within the 6th European Framework Programme which had as one of its goals to develop, optimise and prevalidate a non-animal testing strategy for predicting human acute oral toxicity. In its last 6months, a challenging exercise was conducted to assess the predictive capacity of the developed testing strategies and final identification of the most promising ones. Thirty-two chemicals were tested blind in the battery of in vitro and in silico methods selected during the first phase of the project. This paper describes the classification approaches studied: single step procedures and two step tiered testing strategies. In summary, four in vitro testing strategies were proposed as best performing in terms of predictive capacity with respect to the European acute oral toxicity classification. In addition, a heuristic testing strategy is suggested that combines the prediction results gained from the neutral red uptake assay performed in 3T3 cells, with information on neurotoxicity alerts identified by the primary rat brain aggregates test method. Octanol-water partition coefficients and in silico prediction of intestinal absorption and blood-brain barrier passage are also considered. This approach allows to reduce the number of chemicals wrongly predicted as not classified (LD(50)>2000mg/kg b.w.).Peer Reviewe

    Phase II study of high-sensitivity genotyping of KRAS, NRAS, BRAF and PIK3CA to ultra-select metastatic colorectal cancer patients for panitumumab plus FOLFIRI: the ULTRA trial

    Background: Several studies show the importance of accurately quantifying not only KRAS and other low-abundant mutations because benefits of anti-EGFR therapies may depend on certain sensitivity thresholds. We assessed whether ultra-selection of patients using a high-sensitive digital PCR (dPCR) to determine KRAS, NRAS, BRAF and PIK3CA status can improve clinical outcomes of panitumumab plus FOLFIRI. Patients and methods: This was a single-arm phase II trial that analysed 38 KRAS, NRAS, BRAF and PIK3CA hotspots in tumour tissues of irinotecan-resistant metastatic colorectal cancer patients who received panitumumab plus FOLFIRI until disease progression or early withdrawal. Mutation profiles were identified by nanofluidic dPCR and correlated with clinical outcomes (ORR, overall response rate; PFS, progression-free survival; OS, overall survival) using cut-offs from 0% to 5%. A quantitative PCR (qPCR) analysis was also performed. Results: Seventy-two evaluable patients were enrolled. RAS (KRAS/NRAS) mutations were detected in 23 (32%) patients and RAS/BRAF mutations in 25 (35%) by dPCR, while they were detected in 7 (10%) and 11 (15%) patients, respectively, by qPCR. PIK3CA mutations were not considered in the analyses as they were only detected in 2 (3%) patients by dPCR and in 1 (1%) patient by qPCR. The use of different dPCR cut-offs for RAS (KRAS/NRAS) and RAS/BRAF analyses translated into differential clinical outcomes. The highest ORR, PFS and OS in wild-type patients with their lowest values in patients with mutations were achieved with a 5% cut-off. We observed similar outcomes in RAS/BRAF wild-type and mutant patients defined by qPCR. Conclusions: High-sensitive dPCR accurately identified patients with KRAS, NRAS, BRAF and PIK3CA mutations. The optimal RAS/BRAF mutational cut-off for outcome prediction is 5%, which explains that the predictive performance of qPCR was not improved by dPCR. The biological and clinical implications of low-frequent mutated alleles warrant further investigations

    Strategies to design clinical studies to identify predictive biomarkers in cancer research

    The discovery of reliable biomarkers to predict efficacy and toxicity of anticancer drugs remains one of the key challenges in cancer research. Despite its relevance, no efficient study designs to identify promising candidate biomarkers have been established. This has led to the proliferation of a myriad of exploratory studies using dissimilar strategies, most of which fail to identify any promising targets and are seldom validated. The lack of a proper methodology also determines that many anti-cancer drugs are developed below their potential, due to failure to identify predictive biomarkers. While some drugs will be systematically administered to many patients who will not benefit from them, leading to unnecessary toxicities and costs, others will never reach registration due to our inability to identify the specific patient population in which they are active. Despite these drawbacks, a limited number of outstanding predictive biomarkers have been successfully identified and validated, and have changed the standard practice of oncology. In this manuscript, a multidisciplinary panel reviews how those key biomarkers were identified and, based on those experiences, proposes a methodological framework—the DESIGN guidelines—to standardize the clinical design of biomarker identification studies and to develop future research in this pivotal field

    Exploring the potential of phone call data to characterize the relationship between social network and travel behavior

    [EN] Social network contacts have significant influence on individual travel behavior. However, transport models rarely consider social interaction. One of the reasons is the difficulty to properly model social influence based on the limited data available. Non-conventional, passively collected data sources, such as Twitter, Facebook or mobile phones, provide large amounts of data containing both social interaction and spatiotemporal information. The analysis of such data opens an opportunity to better understand the influence of social networks on travel behavior. The main objective of this paper is to examine the relationship between travel behavior and social networks using mobile phone data. A huge dataset containing billions of registers has been used for this study. The paper analyzes the nature of co-location events and frequent locations shared by social network contacts, aiming not only to provide understanding on why users share certain locations, but also to quantify the degree in which the different types of locations are shared. Locations have been classified as frequent (home, work and other) and non-frequent. A novel approach to identify co-location events based on the intersection of users' mobility models has been proposed. Results show that other locations different from home and work are frequently associated to social interaction. Additionally, the importance of non-frequent locations in co-location events is shown.     112 páginasEl presente estudio, desarrollado por un grupo de estudiantes del Colegio Técnico José Félix Restrepo IED, busca reinterpretar los servicios ecosistémicos de las coberturas vegetales de la cuenca alta del río Fucha, entre las carreras sexta y sexta Este. Este fue el tema principal de la investigación. Contiene información de los recursos naturales que nos ofrecen y que nos benefician no solo a los seres humanos, sino a toda a la biodiversidad que se encuentra sobre la cuenca del río. Entre los servicios se encuentran, por ejemplo, el agua potable limpia y los procesos de descomposición de desechos. Estos se pueden dividir en cuatro categorías amplias como aprovisionamientos (es decir la producción de agua y de alimentos), regulación (el control del clima y de las enfermedades), polinización de cultivos de aves e insectos y, finalmente, la cultural, de la que nos beneficiamos los estudiantes, vecinos y demás personas que hacemos uso de los servicios ecosistémicos que nos ofrece la cuenca. Con esta investigación se busca, por otra parte, conocer un poco más de los grandes beneficios que podemos recibir de la naturaleza y que hacen que la vida humana sea posible

    CIBERER : Spanish national network for research on rare diseases: A highly productive collaborative initiative

This innovative research structure comprises 11 different specific areas dedicated to the main public health priorities in the National Health System. CIBERER, the thematic area of CIBER focused on rare diseases (RDs) currently consists of 75 research groups belonging to universities, research centers, and hospitals of the entire country. CIBERER's mission is to be a center prioritizing and favoring collaboration and cooperation between biomedical and clinical research groups, with special emphasis on the aspects of genetic, molecular, biochemical, and cellular research of RDs. This research is the basis for providing new tools for the diagnosis and therapy of low-prevalence diseases, in line with the International Rare Diseases Research Consortium (IRDiRC) objectives, thus favoring translational research between the scientific environment of the laboratory and the clinical setting of health centers. In this article, we intend to review CIBERER's 15-year journey and summarize the main results obtained in terms of internationalization, scientific production, contributions toward the discovery of new therapies and novel genes associated to diseases, cooperation with patients' associations and many other topics related to RD research

    Croton schiedeanus Schltd prevents experimental hypertension in rats induced by nitric oxide deficit

    Croton schiedeanus Schltd (N.V.: "almizclillo") is a plant used in traditional medicine as an antihypertensive in Colombia. It contains flavonoid, diterpenoid and fenilbutanoid metabolites that have vasodilatation effects linked to the NO/cGMP pathway. This work aimed to assess the capacity of a 96% EtOH extract to prevent the hypertension induced by nitric oxide (NO) deficiency in rats. The NO synthase inhibitor L-NAME (10 mg/kg/d, i.p) was administered during five weeks to three groups of rats (6-7 animals): C. Schiedeanus (200 mg/kg/d, p.o), enalapril (reference, 10 mg/kg/d, p.o) and vehicle (control: olive oil 1 ml/kg/d, p.o). In addition, the blank group received only vehicle. The arterial blood pressure (BP) and heart rate (HR) were measured daily for six weeks. After sacrificing the animals, the aortic rings were isolated, contraction was triggered with phenylephrine (PE 10-6 M) and relaxant responses were achieved with cumulative concentrations of acetylcholine (ACh, 10-10 - 10-4 M). L-NAME increased the systolic arterial pressure in the control group, attaining mean values of 131 mm Hg at week 5, whereas the C. schiedeanus, enalapril and blank groups maintained blood pressure under 100 mm Hg. The capacity of PE to contract aortic rings was greater in the C. schiedeanus, enalapril and blank groups than in the control group (2157, 2005, 1910 and 1646 mg, respectively). The pEC50 values for ACh were as follows: C. Schiedeanus (6.89) >enalapril (6.39) > blank (5.68) > control (5.09). These results give support to C. Schiedeanus as a natural antihypertensive source

    Efecto del lavado con etilendiamina sobre las propiedades de las monopartículas de óxido de estaño

    El óxido de estaño es un compuesto ampliamente utilizado en aplicaciones tecnológicas destacándose como catalizador, sensor de gas, en fabricación de varistores, dispositivos electrónicos, electrodos electrocatalíticos y celdas fotovoltaicas, entre otros. En el presente trabajo se utilizó el método de precipitación controlada (MPC) para sintetizar óxido de estaño de tamaño nanométrico, empleando como precursor sulfato de estaño. El control de las diferentes etapas del proceso de síntesis se realizó a través de los registros de variación de pH, valoración potenciométrica, del sistema. Con base a esta información y el estudio de espectroscopía infrarroja realizada en la fase sólida, a diferentes valores de pH, se propone un mecanismo de formación de las partículas donde las reacciones de hidrólisis y condensación son muy importantes. Se analizó, además, el efecto del lavado del precipitado con etilendiamina y se verificó la eliminación del anión sulfato con este proceso, obteniéndose al final óxido de estaño, SnO y SnO2, como principales fases cristalinas, a temperatura ambiente.
