Detección de enterotoxinas estafilocócicas y TSST-1 mediante una técnica de transferencia electroforética

Este método presenta como ventajas sobre las tecnicas actualmente utilizadas su gran especificidad, elevada rapidez, capacidad de procesar un elevado numero de muestras y adecuada sensibilidad. Además, también se investigo la capacidad de producción de enterotoxinas y tsst-1 por cepas de estafilococos aisladas de mastitis de diversas especies de animales (ovejas, cabras y vacas). Para investigar la capacidad enterotoxigenica de dichas cepas se utilizaron una variante del método elisa y la técnica de transferencia electroforetica desarrollada en la presente tesis. Development of an Artificial Neural Network for the Detection of Supporting Hindlimb Lameness: A Pilot Study in Working Dogs
[EN] Subjective lameness assessment has been a controversial subject given the lack of agreement between observers; this has prompted the development of kinetic and kinematic devices in order to obtain an objective evaluation of locomotor system in dogs. After proper training, neural networks are potentially capable of making a non-human diagnosis of canine lameness. The purpose of this study was to investigate whether artificial neural networks could be used to determine canine hindlimb lameness by computational means only. The outcome of this study could potentially assess the efficacy of certain treatments against diseases that cause lameness. With this aim, input data were obtained from an inertial sensor positioned on the rump. Data from dogs with unilateral hindlimb lameness and sound dogs were used to obtain differences between both groups at walk. The artificial neural network, after necessary adjustments, was integrated into a web management tool, and the preliminary results discriminating between lame and sound dogs are promising. The analysis of spatial data with artificial neural networks was summarized and developed into a web app that has proven to be a useful tool to discriminate between sound and lame dogs. Multiparametric Comparison of Two TTA-Based Surgical Techniques in Dogs with Cranial Cruciate Ligament Tears
[EN] Tearing of the cranial cruciate ligament causes hindlimb lameness in dogs. Different surgical procedures have been proposed to treat this condition. In this study, two different TTA-based techniques and implants were compared. A total of 30 dogs were separated into two groups according to the technique and implant used (Porous TTA® or Model Xgen®). The aim of the study was to assess whether one of these techniques has better functional recovery of the joint, better bone consolidation after the osteotomy procedure and fewer osteoarthritic changes. We compared both groups up to 3 months after surgery. No significant differences were found in any of the assessed parameters. Thus, both procedures were found to be equally effective and safe.SIThanks to the dog owners for their cooperation.This research received no external funding
Control de campo de ayudas agrarias basado en telefonía móvil
Hoy día, la mayoría de los datos que se manejan llevan asociados una componente geográfica. Bajo este contexto
asistimos a un acercamiento de tecnologías empleadas por usuarios expertos hacia usuarios genéricos. En este
marco se plantea la integración de las nuevas tecnologías en el control de las ayudas agrarias. Actualmente, los
procesos de control de ayudas agrarias se basan fundamentalmente en visitas de campo con objeto de certificar el
uso del suelo. Esta metodología de control origina una movilización importante de personal y equipos, empleando
simultáneamente dispositivos como cámara fotográfica, tablet-PC, GPS, etc. El objetivo de este proyecto ha sido el
desarrollo de una aplicación para telefonía móvil aprovechando los distintos sensores que se encuentran integrados
en él, utilizando terminales que añaden nuevas funcionalidades, asegurando la integridad de la información,
mejorando su correcta interpretación así como estableciendo diferentes canales de difusión. El proyecto se ha
desarrollado bajo Android, empleando librerías de código abierto y de ejecución independiente al fabricante del
dispositivo telefónico. El resultado es un software que integra información de diferentes sensores, presentando el
terminal móvil apto para los trabajos de control de campo, reduciendo el material a emplear y mejorando la
integridad de la información.Today, most data we use is associated with a geographic component. In this context we are witnessing a
convergence in technologies used by expert users to generic users. In this framework we look at the integration of
new technologies in the control of agricultural aid. Currently, control processes agricultural subsidies are
fundamentally based on field visits in order to certify the land use. This control methodology results in a significant
mobilization of personnel and equipment, using devices like a camera, tablet PC, GPS, etc. The objective of this
project has been to develop a mobile application taking advantage of the different sensors that are integrated into it,
using terminals that add new functionality, ensuring data integrity, improving its correct interpretation as well as
establishing different channels diffusion. The project has been developed under Android, using open source libraries
independent to the device manufacturer. The result is software that integrates information from different sensors,
introducing the mobile terminal suitable for the work of field control, reducing the material to be used and improved
data integrity
Effects of testosterone and exercise training on bone microstructure of rats
[EN] Background and Aim: Male hypogonadism results from failure to produce physiological levels of testosterone. Testosterone in men is essential in masculine development, sperm production, and adult man’s health. Osteoporosis is one of the consequences of hypogonadism. Regular physical exercise and exogenous testosterone administration are frequently used to prevent or treat this condition. This study aimed to understand the effects of lifelong exercise training and testosterone levels (isolated and together) in the main bone structure parameters. Materials and Methods: A total of 24 rats were used and randomly divided into four groups: Control group (CG; n=6), exercised group (EG, n=6), testosterone group (TG, n=6), and testosterone EG (TEG, n=6). A micro-computed tomography equipment was used to evaluate 15 bone parameters. Results: Both factors (exercise training and testosterone) seem to improve the bone resistance and microstructure, although in different bone characteristics. Testosterone influenced trabecular structure parameters, namely, connectivity density, trabecular number, and trabecular space. The exercise promoted alterations in bone structure as well, although, in most cases, in different bone structure parameters as bone mineral density and medullar mineral density. Conclusion: Overall, exercise and testosterone therapy seems to have a synergistic contribution to the general bone structure and resistance. Transcriptomic Profile of Canine DH82 Macrophages Infected by Leishmania infantum Promastigotes with Different Virulence Behavior
Zoonotic visceral leishmaniosis caused by Leishmania infantum is an endemic disease in the Mediterranean Basin affecting mainly humans and dogs, the main reservoir. The leishmaniosis outbreak declared in the Community of Madrid (Spain) led to a significant increase in human disease incidence without enhancing canine leishmaniosis prevalence, suggesting a better adaptation of the outbreak’s isolates by other host species. One of the isolates obtained in the focus, IPER/ES/2012/BOS1FL1 (BOS1FL1), has previously demonstrated a different phenotype than the reference strain MCAN/ES/1996/BCN150 (BCN150), characterized by a lower infectivity when interacting with canine macrophages. Nevertheless, not enough changes in the cell defensive response were found to support their different behavior. Thus, we decided to investigate the molecular mechanisms involved in the interaction of both parasites with DH82 canine macrophages by studying their transcriptomic profiles developed after infection using RNA sequencing. The results showed a common regulation induced by both parasites in the phosphoinositide-3-kinase–protein kinase B/Akt and NOD-like receptor signaling pathways. However, other pathways, such as phagocytosis and signal transduction, including tumor necrosis factor, mitogen-activated kinases and nuclear factor-κB, were only regulated after infection with BOS1FL1. These differences could contribute to the reduced infection ability of the outbreak isolates in canine cells. Our results open a new avenue to investigate the true role of adaptation of L. infantum isolates in their interaction with their different hosts
Development and Evaluation of a Disease Large Animal Model for Preclinical Assessment of Renal Denervation Therapies
[EN] New-generation catheters-based renal denervation (RDN) is under investigation for the treatment of uncontrolled hypertension (HTN). We assessed the feasibility of a large animal model of HTN to accommodate the human RDN devices. Ten minipigs were instrumented to measure blood pressure (BP) in an awake-state. HTN was induced with subcutaneous 11-deoxycorticosterone (DOCA, 100 mg/kg) implants. Five months after, the surviving animals underwent RDN with the Symplicity® system. Norepinephrine (NE) renal gradients were determined before and 1 month after RDN. Renal arteries were processed for histological (hematoxylin-eosin, Movat pentachrome) and immunohistochemical (S100, tyrosine-hydroxylase) analyses. BP significantly rose after DOCA implants. Six animals died prematurely, mainly from infectious causes. The surviving animals showed stable BP levels after 5 months. One month after RDN, nerve damage was showed in three animals, with impedance drop >10%, NE gradient drop and reduction in BP. The fourth animal showed no nerve damage, impedance drop <10%, NE gradient increase and no change in BP. In conclusion, the minipig model of DOCA-induced HTN is feasible, showing durable effects. High mortality should be addressed in next iterations of this model. RDN may partially offset the DOCA-induced HTN. Hydroxyl Groups Induce Bioactivity in Silica/Chitosan Aerogels Designed for Bone Tissue Engineering. In Vitro Model for the Assessment of Osteoblasts Behavior
Silica (SiO2)/chitosan (CS) composite aerogels are bioactive when they are submerged in simulated body fluid (SBF), causing the formation of bone-like hydroxyapatite (HAp) layer. Silica-based hybrid aerogels improve the elastic behavior, and the combined CS modifies the network entanglement as a crosslinking biopolymer. Tetraethoxysilane (TEOS)/CS is used as network precursors by employing a sol-gel method assisted with high power ultrasound (600 W). Upon gelation and aging, gels are dried in supercritical CO2 to obtain monoliths. Thermograms provide information about the condensation of the remaining hydroxyl groups (400-700 degrees C). This step permits the evaluation of the hydroxyl group's content of 2 to 5 OH nm(-2). The formed Si-OH groups act as the inductor of apatite crystal nucleation in SBF. The N-2 physisorption isotherms show a hysteresis loop of type H3, characteristic to good interconnected porosity, which facilitates both the bioactivity and the adhesion of osteoblasts cells. After two weeks of immersion in SBF, a layer of HAp microcrystals develops on the surface with a stoichiometric Ca/P molar ratio of 1.67 with spherulite morphology and uniform sizes of 6 mu m. This fact asserts the bioactive behavior of these hybrid aerogels. Osteoblasts are cultured on the selected samples and immunolabeled for cytoskeletal and focal adhesion expression related to scaffold nanostructure and composition. The initial osteoconductive response observes points to a great potential of tissue engineering for the designed composite aerogels
Epidemiological and clinical characteristics of Streptococcus tigurinus endocarditis
Background: Streptococcus tigurinus was recently described as a new streptococcal species within the viridans group streptococci (VGS). The objectives of the present work were to analyse the clinical and microbiological characteristics of S. tigurinus isolated from patients with bacteraemias, to determine the prevalence of S. tigurinus among VGS endocarditis in Spain, and to compare the clinical characteristics and outcomes of endocarditis caused by S. tigurinus and other VGS. Methods: Retrospective nationwide study, performed between 2008 and 2016 in 9 Spanish hospitals from 7 different provinces comprising 237 cases of infective endocarditis. Streptococcal isolates were identified by sequencing fragments of their 16S rRNA, sodA and groEL genes. Clinical data of patients with streptococcal endocarditis were prospectively collected according to a pre-established protocol. Results: Patients with endocarditis represented 7/9 (77.8%) and 26/86 (30.2%) of the bacteraemias caused by S. tigurinus and other VGS, respectively (p < 0.001), in two of the hospital participants. Among patients with streptococcal endocarditis, 12 different Streptococcus species were recognized being S. oralis, S. tigurinus and S. mitis the three more common. No relevant statistical differences were observed in the clinical characteristics and outcomes of endocarditis caused by the different VGS species. Conclusions: In this multicenter study performed in Spain, S. tigurinus showed a higher predilection for the endocardial endothelium as compared to other VGS. Role of age and comorbidities in mortality of patients with infective endocarditis
[Purpose]: The aim of this study was to analyse the characteristics of patients with IE in three groups of age and to assess the ability of age and the Charlson Comorbidity Index (CCI) to predict mortality.
[Methods]: Prospective cohort study of all patients with IE included in the GAMES Spanish database between 2008 and 2015.Patients were stratified into three age groups:<65 years,65 to 80 years,and ≥ 80 years.The area under the receiver-operating characteristic (AUROC) curve was calculated to quantify the diagnostic accuracy of the CCI to predict mortality risk.
[Results]: A total of 3120 patients with IE (1327 < 65 years;1291 65-80 years;502 ≥ 80 years) were enrolled.Fever and heart failure were the most common presentations of IE, with no differences among age groups.Patients ≥80 years who underwent surgery were significantly lower compared with other age groups (14.3%,65 years; 20.5%,65-79 years; 31.3%,≥80 years). In-hospital mortality was lower in the <65-year group (20.3%,<65 years;30.1%,65-79 years;34.7%,≥80 years;p < 0.001) as well as 1-year mortality (3.2%, <65 years; 5.5%, 65-80 years;7.6%,≥80 years; p = 0.003).Independent predictors of mortality were age ≥ 80 years (hazard ratio [HR]:2.78;95% confidence interval [CI]:2.32–3.34), CCI ≥ 3 (HR:1.62; 95% CI:1.39–1.88),and non-performed surgery (HR:1.64;95% CI:11.16–1.58).When the three age groups were compared,the AUROC curve for CCI was significantly larger for patients aged <65 years(p < 0.001) for both in-hospital and 1-year mortality.
[Conclusion]: There were no differences in the clinical presentation of IE between the groups. Age ≥ 80 years, high comorbidity (measured by CCI),and non-performance of surgery were independent predictors of mortality in patients with IE.CCI could help to identify those patients with IE and surgical indication who present a lower risk of in-hospital and 1-year mortality after surgery, especially in the <65-year group
