    Virus-Specific Read-Through Codon Preference Affects Infectivity of Chimeric Cucumber Green Mottle Mosaic Viruses Displaying a Dengue Virus Epitope

    A Cucumber green mottle mosaic virus (CGMMV) was used to present a truncated dengue virus type 2 envelope (E) protein binding region from amino acids 379 to 423 (EB4). The EB4 gene was inserted at the terminal end of the CGMMV coat protein (CP) open reading frame (ORF). Read-through sequences of TMV or CGMMV, CAA-UAG-CAA-UUA, or AAA-UAG-CAA-UUA were, respectively, inserted in between the CP and the EB4 genes. The chimeric clones, pRT, pRG, and pCG+FSRTRE, were transcribed into full-length capped recombinant CGMMV transcripts. Only constructs with the wild-type CGMMV read-through sequence yielded infectious viruses following infection of host plant, muskmelon (Cucumis melo) leaves. The ratio of modified to unmodified CP for the read-through expression clone developed was also found to be approximately 1:1, higher than what has been previously reported. It was also observed that infectivity was not affected by differences in pI between the chimera and its wild counterpart. Analysis of recombinant viruses after 21-days-postinculation (dpi) revealed that deletions occurred resulting in partial reversions of the viral population to near wild type and suggesting that this would be the limiting harvest period for obtaining true to type recombinants with this construct

    Diversity and ecological guild analysis of the oil palm fungal microbiome across root, rhizosphere, and soil compartments

    The rhizosphere microbiome is a major determinant of plant health, which can interact with the host directly and indirectly to promote or suppress productivity. Oil palm is one of the world’s most important crops, constituting over a third of global vegetable oil production. Currently there is little understanding of the oil palm microbiome and its contribution to plant health and productivity, with existing knowledge based almost entirely on culture dependent studies. We investigated the diversity and composition of the oil palm fungal microbiome in the bulk soil, rhizosphere soil, and roots of 2-, 18-, and 35-year old plantations in Selangor, Malaysia. The fungal community showed substantial variation between the plantations, accounting for 19.7% of community composition, with compartment (root, rhizosphere soil, and bulk soil), and soil properties (pH, C, N, and P) contributing 6.5 and 7.2% of community variation, respectively. Rhizosphere soil and roots supported distinct communities compared to the bulk soil, with significant enrichment of Agaricomycetes, Glomeromycetes, and Lecanoromycetes in roots. Several putative plant pathogens were abundant in roots in all the plantations, including taxa related to Prospodicola mexicana and Pleurostoma sp. The mycorrhizal status and dependency of oil palm has yet to be established, and using 18S rRNA primers we found considerable between-site variation in Glomeromycotinian community composition, accounting for 31.2% of variation. There was evidence for the selection of Glomeromycotinian communities in oil palm roots in the older plantations but compartment had a weak effect on community composition, accounting for 3.9% of variation, while soil variables accounted for 9% of community variation. While diverse Mucoromycotinian fungi were detected, they showed very low abundance and diversity within roots compared to bulk soil, and were not closely related to taxa which have been linked to fine root endophyte mycorrhizal morphology. Many of the fungal sequences showed low similarity to established genera, indicating the presence of substantial novel diversity with significance for plant health within the oil palm microbiome

    Adverse events following immunization and psychological distress among cancer patients/survivors following vaccination against SARS-CoV-2 infection

    PurposeThis study aims to describe the adverse events following immunization (AEFIs) of SARS-CoV-2 vaccination in cancer patients/survivors associated with their psychological distress.MethodsA cross-sectional study was conducted to assess AEFIs after the receipt of SARS-CoV-2 vaccines in cancer patients/survivors attending a university hospital in Malaysia. Psychological distress was measured using the Hospital Anxiety and Depression Scale (HADS) before and after the first and second doses of COVID-19 vaccine.ResultsA total of 217 complete responses were received. Compared with before vaccination, both HADS Anxiety (HADS-A) and HADS Depression (HADS-D) scores were significantly reduced after the first and second dose of the SARS-CoV-2 vaccine. Most of the participants had mild-or-moderate systemic and local AEFIs, with the most common being pain at the injection site, tiredness, and headache for both the first and second doses of the vaccine. Positive correlations between the total AEFI score and HADS-A (r = 0.309, p < 0.001) and HADS-D (r = 0.214, p = 0.001) scores were observed after the first dose of the SARS-CoV-2 vaccine. Similarly, positive associations were observed between the total AEFI score and HADS-A (r = 0.305, p < 0.001) and HADS-D (r = 0.235, p < 0.001) scores after the second dose of the SARS-CoV-2 vaccine.ConclusionMild-to-moderate AEFIs found in this study help address vaccine hesitancy in cancer patients/survivors. Receiving the SARS-CoV-2 vaccine had a positive effect on decreasing psychological distress in cancer patients/survivors. High severity of an AEFI was associated with higher anxiety and depressive symptoms

    Enhancing Uptake of Nature-Based Solutions for Informing Coastal Sustainable Development Policy and Planning: A Malaysia Case Study

    Nature-based Solutions (NbS) have been advocated to protect, sustainably manage, and restore natural or modified ecosystems, simultaneously providing human well-being and biodiversity benefits. The uptake of NbS differs regionally with some countries exhibiting greater uptake than others. The success of NbS also differs regionally with varying environmental conditions and social-ecological processes. In many regions, the body of knowledge, particularly around the efficacy of such efforts, remains fragmented. Having an “inventory” or “tool box” of regionally-trialed methods, outcomes and lessons learnt can improve the evidence base, inform adaptive management, and ultimately support the uptake of NbS. Using Malaysia as a case study, we provide a comprehensive overview of trialed and tested NbS efforts that used nature to address societal challenges in marine and coastal environments (here referring to mangroves, seagrass, coral reefs), and detailed these efforts according to their objectives, as well as their anticipated and actual outcomes. The NbS efforts were categorized according to the IUCN NbS approach typology and mapped to provide a spatial overview of IUCN NbS effort types. A total of 229 NbS efforts were collated, representing various levels of implementation success. From the assessment of these efforts, several key actions were identified as a way forward to enhance the uptake of Nature-based Solutions for informing coastal sustainable development policy and planning. These include increasing education, training, and knowledge sharing; rationalizing cooperation across jurisdictions, laws, and regulations; enhancing environmental monitoring; leveraging on existing policies; enabling collaboration and communication; and implementing sustainable finance instruments. These findings can be used to inform the improved application and uptake of NbS, globally.</jats:p

    With advances in technology, electrical devices follow the Moores Law, where the size of the devices tends smaller and smaller. With the diminishing supply of natural gas, new energy harvesting methods are being explored. One such method being explored is the kinetic energy harvesting. Researches have been conducted and electrostatic generators integrated with electrets are found to be well suited to harvest the kinetic energy that is vibrant in our environment, car and human traffic movement. In the initial experiments conducted to measure the natural frequency of the mechanical mechanism in this micro electrets/electrostatic power generator produced high inconsistent result which hinders the proper modelling of the generator. An analysis and review conducted on the experimentation set up raises multiple sources of external influences and possible sources of error, for example realignment effects due to vibration, electrical-mechanical interactions. In this report, efforts will be taken to optimise on the measurement of power generation efficiency of the micro electrets/electrostatic power generator. However, due to inconsistency in measurement readings for natural frequency of the system, efforts are focused into improving the experimentation setup to achieve better measurement results.
