    Myogenetic oligodeoxynucleotide complexed with berberine promotes differentiation of chicken myoblasts

    Myoblasts are myogenic precursors that develop into myotubes during muscle formation. Improving efficiency of myoblast differentiation is important for advancing meat production by domestic animals. We recently identified novel oligodeoxynucleotides (ODNs) termed myogenetic ODNs (myoDNs) that promote the differentiation of mammalian myoblasts. An isoquinoline alkaloid, berberine, forms a complex with one of the myoDNs, iSN04, and enhances its activities. This study investigated the effects of myoDNs on chicken myoblasts to elucidate their species-specific actions. Seven myoDNs (iSN01-iSN07) were found to facilitate the differentiation of chicken myoblasts into myosin heavy chain (MHC)-positive myotubes. The iSN04-berberine complex exhibited a higher myogenetic activity than iSN04 alone, which was shown to enhance the differentiation of myoblasts into myotubes and the upregulated of myogenic gene expression (MyoD, myogenin, MHC, and myomaker).     Comparison of Conventional 2D CC (Cranio-Caudal) + MLO (Medio-Lateral Oblique) Bi-Directional Photography and 2D-MLO + DBT-MLO (Digital Breast Tomosynthesis) in Mammography Examination

    Introduction: Mammography (MMG) is an excellent examination for breast cancer detection and is widely used in both screening and clinical practice. However, the breast thickness and background the concentration of the mammary gland is large among individuals, and the mammary gland and cancer may overlap and it may not be possible to point out a lesion. In recent years, digital breast tomosynthesis (hereinafter referred to as DBT) has been introduced to solve these problems, and its usefulness has been reported in clinical practice. Therefore, in anticipation of a higher cancer detection rate etc., we compared the conventional 2D CC + MLO 2-way MMG examination with 2DMLO + DBTMLO 2-way MMG examination.Materials and Methods: The combination of 2D CC + MLO and 2DMLO + DBTMLO was read for 95 cases of breast cancer taking 2D CC and MLO and DBT MLO from May 2016 to October 2017. It was judged that the category 3 or more required detailed examination, cancer detection, and cancer detection rates were compared. Subjects were 28 to 87 years of age (median: 55 years), breast composition: high concentration 15.8% (15 cases), heterogeneous high concentration 47.4% (45 cases), mammary gland 28.4% (27 cases), Fatty was 8.4% (8 cases).Results: The cancer detection rate was 87% for the 2D CC + MLO combination and 94% for the 2DMLO + DBTMLO combination in 95 cases of breast cancer. The interpretation time was 95 minutes for the 2D CC + MLO combination and 110 minutes for the 2DMLO + DBTMLO combination.The breast cancer detected only by the combination of 2D CC + MLO was scattered in 1 case and was classified as Category 3 by FAD. Breast cancer detected only by the combination of 2DMLO + DBTMLO: 8 cases with uneven distribution of breast composition, 3 cases with scattered mammary gland, 3 cases with 5 cases with disordered construction, Category 4 with 3 cases with FAD was.Conclusion: Both 2D CC + MLO combination and 2DMLO + DBTMLO combination are considered useful for screening MMG examination. In particular, we would like to consider the introduction to medical examinations for the addition of MMG in DBT, but the issue of the amount of image information, the problem of exposure to radiation, the direction of imaging, the establishment of interpretation methods etc. are future issues

    A Method of Describing a Self-occlusive Motion - A Reverse Motion History Image

    This paper proposes a new method of describing a self-occlusive human motion, particularly in the depth direction, which has been considered little in the motion/action recognition studies to date in spite of its importance in our daily life. A Motion History Image (MHI) is a well-known method of describing a motion by a single gray value image, but it suffers from a self-occlusion problem in which present motion overwrites past motion. To solve this difficulty, a Reverse description MHI (RMHI) is proposed in the paper. RMHI and the original MHI are both employed for motion representation in the proposed method; the former for approach motion, whereas the latter for leave motion. In the experiment on motion recognition, motions are described by RMHI or MHI according to motion direction, transformed then to Hu moment vectors, and finally recognized employing the k-nearest neighbor.     Distinct cell proliferation, myogenic differentiation, and gene expression in skeletal muscle myoblasts of layer and broiler chickens

    Myoblasts play a central role during skeletal muscle formation and growth. Precise understanding of myoblast properties is thus indispensable for meat production. Herein, we report the cellular characteristics and gene expression profiles of primary-cultured myoblasts of layer and broiler chickens. Broiler myoblasts actively proliferated and promptly differentiated into myotubes compared to layer myoblasts, which corresponds well with the muscle phenotype of broilers. Transcriptomes of layer and broiler myoblasts during differentiation were quantified by RNA sequencing. Ontology analyses of the differentially expressed genes (DEGs) provided a series of extracellular proteins as putative markers for characterization of chicken myogenic cells. Another ontology analyses demonstrated that broiler myogenic cells are rich in cell cycle factors and muscle components. Independent of these semantic studies, principal component analysis (PCA) statistically defined two gene sets: one governing myogenic differentiation and the other segregating layers and broilers. Thirteen candidate genes were identified with a combined study of the DEGs and PCA that potentially contribute to proliferation or differentiation of chicken myoblasts. We experimentally proved that one of the candidates, enkephalin, an opioid peptide, suppresses myoblast growth. Our results present a new perspective that the opioids present in feeds may influence muscle development of domestic animals.Articlejournal articl

    USP8 prevents aberrant NF-κB and Nrf2 activation by counteracting ubiquitin signals from endosomes

    K63-linked ubiquitin chains attached to plasma membrane proteins serve as tags for endocytosis and endosome-to-lysosome sorting. USP8 is an essential deubiquitinase for the maintenance of endosomal functions. Prolonged depletion of USP8 leads to cell death, but the major effects on cellular signaling pathways are poorly understood. Here, we show that USP8 depletion causes aberrant accumulation of K63-linked ubiquitin chains on endosomes and induces immune and stress responses. Upon USP8 depletion, two different decoders for K63-linked ubiquitin chains, TAB2/3 and p62, were recruited to endosomes and activated the TAK1-NF-κB and Keap1-Nrf2 pathways, respectively. Oxidative stress, an environmental stimulus that potentially suppresses USP8 activity, induced accumulation of K63-linked ubiquitin chains on endosomes, recruitment of TAB2, and expression of the inflammatory cytokine. The results demonstrate that USP8 is a gatekeeper of misdirected ubiquitin signals and inhibits immune and stress response pathways by removing K63-linked ubiquitin chains from endosomes.</p

    Mapping of Sea Surface Nutrients in the North Pacific: Basin-wide Distribution and Seasonal to Interannual Variability

    Monthly maps of sea surface nutrient (phosphate, nitrate and silicate) concentrations were produced for the North Pacific (10-60°N, 120°E-90°W) for the years 2001 to 2010 using a self-organizing map trained with temperature, salinity, chlorophyll-a concentration and mixed layer depth. Nutrient sampling was carried out mainly by ships of opportunity, providing good seasonal coverage of the surface ocean. Using the mapping results, we investigated the spatio-temporal variability of surface North Pacific nutrient and dissolved inorganic carbon (DIC) distributions on seasonal and interannual time scales. Nutrient and DIC concentrations were high in the subarctic in winter and low in the subtropics. In the summer, substantial amount of nutrients remained unutilized in subarctic and the northern part of the subarctic-subtropical boundary region while that was not the case in the southern part of the boundary region. In the subtropics, nutrients were almost entirely depleted throughout the year, while DIC concentrations showed a north-south gradient and significant seasonal change. Nutrients and DIC show a large seasonal drawdown in the western subarctic region, while the drawdown in the eastern subarctic region was weaker, especially for silica. The subarctic-subtropical boundary region also showed a large seasonal drawdown, which was most prominent for DIC and less obvious for nitrate and silicate. In the interannual time scale, the Pacific Decadal Oscillation was related to a seesaw pattern between the subarctic-subtropical boundary region and the Alaskan Gyre through the changes in horizontal advection, vertical mixing and biological production
