    Adenovirus Gene Transfer to Amelogenesis Imperfecta Ameloblast-Like Cells

    To explore gene therapy strategies for amelogenesis imperfecta (AI), a human ameloblast-like cell population was established from third molars of an AI-affected patient. These cells were characterized by expression of cytokeratin 14, major enamel proteins and alkaline phosphatase staining. Suboptimal transduction of the ameloblast-like cells by an adenovirus type 5 (Ad5) vector was consistent with lower levels of the coxsackie-and-adenovirus receptor (CAR) on those cells relative to CAR-positive A549 cells. To overcome CAR -deficiency, we evaluated capsid-modified Ad5 vectors with various genetic capsid modifications including β€œpK7” and/or β€œRGD” motif-containing short peptides incorporated in the capsid protein fiber as well as fiber chimera with the Ad serotype 3 (Ad3) fiber β€œknob” domain. All fiber modifications provided an augmented transduction of AI-ameloblasts, revealed following vector dose normalization in A549 cells with a superior effect (up to 404-fold) of pK7/RGD double modification. This robust infectivity enhancement occurred through vector binding to both Ξ±vΞ²3/Ξ±vΞ²5 integrins and heparan sulfate proteoglycans (HSPGs) highly expressed by AI-ameloblasts as revealed by gene transfer blocking experiments. This work thus not only pioneers establishment of human AI ameloblast-like cell population as a model for in vitro studies but also reveals an optimal infectivity-enhancement strategy for a potential Ad5 vector-mediated gene therapy for AI

    Every year the number of free land plots for construction of buildings is steadily decreasing in cities. In this regard, the participants in the investment and construction process are increasingly seeking to maximize the use of land. A logical way for this is to use high-rise construction. However, until recently, builders encountered serious obstacles in the form of lack of special norms and rules for the design of high-rise buildings. It led to the need for individual coordination of each high-rise facility, the development and approval of special technical conditions, the passage of numerous administrative approvals. Thus, investment activity regarding the construction of high-rise buildings in the Russian Federation is reduced. In 2016, there were regulatory changes that substantially alleviated these difficulties. In this article, the authors analyze the features of the town-planning normative-legal field of high-rise construction and its development, track the interrelations between the regulatory regulation of the construction of similar facilities, with the real need, the technical feasibility and economic feasibility of their construction in the urban district of Samara. Conclusions and suggestions are also based on the analysis of the norms of urban zoning, the residential real estate market and the value of land plots

    Transformation of plant substrates by fungi

    The results of chemical and thermal analysis of substrates based on the post-extraction residue of balsam poplar (Populus balsamifera L.) buds before and after bioconversion by Fp5-15 Fomitopsis pinicola fungi were compared. The thermal characteristics obtained using thermogravimetry (TG/DTG) methods are presented. The stages of thermal decomposition, their temperature intervals and the temperatures of the maxima on the DTG curves, as well as the weight loss of the samples, have been established. Studies have shown that under the action of the enzyme complex of fungi, easily and difficultly hydrolysable polysaccharides are utilized, and lignin substances also undergo changes

    Discrimination between G/C Binding Sites by Olivomycin A Is Determined by Kinetics of the Drug-DNA Interaction

    Olivomycin A (OA) exerts its cytotoxic potency due to binding to the minor groove of the G/C-rich DNA and interfering with replication and transcription. Screening of the complete set of tetranucleotide G/C sites by electrophoretic mobility gel shift assay (EMSA) revealed that the sites containing central GC or GG dinucleotides were able to bind OA, whereas the sites with the central CG dinucleotide were not. However, studies of equilibrium OA binding in solution by fluorescence, circular dichroism and isothermal titration calorimetry failed to confirm the sequence preference of OA, indicating instead a similar type of complex and comparable affinity of OA to all G/C binding sites. This discrepancy was resolved by kinetics analysis of the drug–DNA interaction: the dissociation rate significantly differed between SGCS, SGGS and SCGS sites (S stands for G or C), thereby explaining the disintegration of the complexes during EMSA. The functional relevance of the revealed differential kinetics of OA–DNA interaction was demonstrated in an in vitro transcription assay. These findings emphasize the crucial role of kinetics in the mechanism of OA action and provide an important approach to the screening of new drug candidates

    Low-Frequency Content of THz Emission from Two-Color Femtosecond Filament

    We experimentally investigate the low-frequency (below 1 THz) spectral content of broadband terahertz (THz) emission from two-color femtosecond filament formed by the 2.7-mJ, 40-fs, 800+400-nm pulse focused into air. For incoherent detection, we screened the Golay cell by the bandpass filters and measured the THz angular distributions at the selected frequencies Ξ½=0.5, 1, 2 and 3 THz. The measured distributions of THz fluence were integrated over the forward hemisphere taking into account the transmittance of the filters, thus providing the estimation of spectral power at the frequencies studied. The spectral power decreases monotonically with the frequency increasing from 0.5 to 3 THz, thus showing that the maximum of THz spectrum is attained at ν≀0.5 THz. The THz waveform measured by electro-optical sampling (EOS) based on ZnTe crystal and transformed into the spectral domain shows that there exists the local maximum of the THz spectral power at Ξ½β‰ˆ1 THz. This disagrees with monotonic decrease of THz spectral power obtained from the filter-based measurements. We have introduced the correction to the spectral power reconstructed from EOS measurements. This correction takes into account different focal spot size for different THz frequencies contained in the broadband electromagnetic pulse. The corrected EOS spectral power is in semi-quantitative agreement with the one measured by a set of filters
