    Simultaneous constraints on the growth of structure and cosmic expansion from the multipole power spectra of the SDSS DR7 LRG sample

    The anisotropic galaxy clustering on large scales provides us with a unique opportunity to probe into the gravity theory through the redshift-space distortions (RSDs) and the Alcock-Paczynski effect. Using the multipole power spectra up to hexadecapole (ell=4), of the Luminous Red Galaxy (LRG) sample in the data release 7 (DR7) of the Sloan Digital Sky Survey II (SDSS-II), we obtain simultaneous constraints on the linear growth rate f, angular diameter distance D_A, and Hubble parameter H at redshift z = 0.3. For this purpose, we first extensively examine the validity of a theoretical model for the non-linear RSDs using mock subhalo catalogues from N-body simulations, which are constructed to match with the observed multipole power spectra. We show that the input cosmological parameters of the simulations can be recovered well within the error bars by comparing the multipole power spectra of our theoretical model and those of the mock subhalo catalogues. We also carefully examine systematic uncertainties in our analysis by testing the dependence on prior assumption of the theoretical model and the range of wavenumbers to be used in the fitting. These investigations validate that the theoretical model can be safely applied to the real data. Thus, our results from the SDSS DR7 LRG sample are robust including systematics of theoretical modeling; f(z = 0.3) sigma_8(z = 0.3) =0.49+-0.08(stat.)+-0.04(sys.), D_A (z = 0.3) =968+-42(stat.)+-17(sys.)[Mpc], H (z = 0.3) =81.7+-5.0(stat.)+-3.7(sys.)[km/s/Mpc]. We believe that our method to constrain the cosmological parameters using subhaloes catalogues will be useful for more refined samples like CMASS and LOWZ catalogues in the Baryon Oscillation Spectroscopic Survey in SDSS-III.Comment: accepted for publication in MNRA

    DRG-targeted helper-dependent adenoviruses mediate selective gene delivery for therapeutic rescue of sensory neuronopathies in mice

    Dorsal root ganglion (DRG) neuron dysfunction occurs in a variety of sensory neuronopathies for which there are currently no satisfactory treatments. Here we describe the development of a strategy to target therapeutic genes to DRG neurons for the treatment of these disorders. We genetically modified an adenovirus (Ad) to generate a helper virus (HV) that was detargeted for native adenoviral tropism and contained DRG homing peptides in the adenoviral capsid fiber protein; we used this HV to generate DRG-targeted helper-dependent Ad (HDAd). In mice, intrathecal injection of this HDAd produced a 100-fold higher transduction of DRG neurons and a markedly attenuated inflammatory response compared with unmodified HDAd. We also injected HDAd encoding the β subunit of β-hexosaminidase (Hexb) into Hexb-deficient mice, a model of the neuronopathy Sandhoff disease. Delivery of the DRG-targeted HDAd reinstated neuron-specific Hexb production, reversed gangliosidosis, and ameliorated peripheral sensory dysfunction. The development of DRG neuron–targeted HDAd with proven efficacy in a preclinical model may have implications for the treatment of sensory neuronopathies of diverse etiologies

    Designing a banking scale of human induced pluripotent stem cells based on suspension time-dependent quality variations in filling and cryopreservation processes

    To establish a robust commercial production system for a cell product, it is necessary to investigate a lot of variable factors inside and outside of the system and discuss the cell manufacturability. In case of trying a scale-up of banking system for human induced pluripotent stem cells (hiPSCs), the process time to fill the cell suspension into vials before cryopreservation is prolonged. And that will cause the decay of the cell quality, because cryoprotective agent (CPA) including dimethyl sulfoxide has toxicity to cells. Based on such fluctuation of cell product quality derived from time-dependency in down-stream process, novel strategy to design a process time and a banking scale is required. In this study, four performance indexes, survival ratio of cells during suspension in CPA before cryopreservation (γ), survival ratio, attachment efficiency and specific growth rate of cells after cryopreservation (β, α and μ, respectively) are proposed to evaluate the cellular state and potential of the product. And, the quality variations of suspended cells in CPA are elucidated by changing the process time of suspension at room temperature and 4 °C. At room temperature, γ decreased with process time (ts) exponentially, being γ = 0.72 at ts = 6 h. With respect to α, 4 hours suspension at room temperature had an insignificant effect, however, it dropped after the lag-time, being α = 0.73 at ts = 6 h. In contrast, β and μ were kept high level of 0.80 and 5.3 × 10-2 h-1, respectively, similarly to those without the process. In addition, the suspension at 4°C made the enhancement of γ and α at ts = 6 h (γ = 0.88 and α = 1.08, respectively), suggesting that the suppression in cell activity during suspension is important to preserve the cell quality. In conclusion, the proposed performance indexes are useful to estimate the state and potential of cell product in filling and cryopreservation processes, and the temperature control in filling process is one of the promising factors to maintain the cell product quality.     Psychological, social, and environmental factors to meeting physical activity recommendations among Japanese adults

    <p>Abstract</p> <p>Background</p> <p>Although the benefits of the recommended level of physical activity on reducing chronic diseases are well-established, most of the Japanese population is not sufficiently active. Thus, examining correlates is an important prerequisite for designing relevant polices and effective programs. The present study investigated psychological, social, and environmental factors associated with meeting physical activity recommendations among Japanese adults.</p> <p>Methods</p> <p>Data were analyzed for 1,932 men and women (43.6 ± 13.0 years), who responded to an Internet-based cross-sectional survey. Self-reported measure of physical activity, psychological (self-efficacy, pros, and cons), social (social support, health professional advice), environmental (home fitness equipment, access to facilities, neighborhood safety, enjoyable scenery, frequently observing others exercising, residential area), and demographic (gender, age, marital status, educational level, household income level, employment status) variables were obtained. Based on the current national guidelines for exercise in Japan (23 METs·hour per week), respondents were divided into two categories–recommended and not recommended (insufficient and inactive)–according to their estimated weekly physical activity level. An adjusted logistic regression model was utilized.</p> <p>Results</p> <p>When adjusting for all other variables, self-efficacy (men: OR = 2.13; 95% CI: 1.55–2.94, women: OR = 2.72; 95% CI: 1.82–4.08) and possessing home fitness equipment (men: OR = 1.55; 95% CI: 1.14–2.10, women: OR = 1.41; 95% CI: 1.01–1.99) for both genders, social support (OR = 1.44; 95% CI: 1.06–1.97) for men, and enjoyable scenery (OR = 1.60; 95% CI: 1.09–2.36) for women were positively associated with attaining the recommended level of physical activity. In women, cons (OR = 0.47; 95% CI: 0.33–0.67) and living in rural areas (OR = 0.50; 95% CI: 0.25–0.97) were negatively associated with meeting the physical activity recommendations.</p> <p>Conclusion</p> <p>In the psychological, social, and environmental domains, significant correlates of attaining the recommended level of physical activity were observed. Men and women had different patterns of psychological, social, and environmental correlates. These findings suggest that an intervention design that accounts for those correlates may more effectively promote physical activity among Japanese adults.</p

    Efficacy and Safety of Early Intravenous Landiolol on Myocardial Salvage in Patients with ST-segment Elevation Myocardial Infarction before Primary Percutaneous Coronary Intervention: A Randomized Study

    Early treatment with an oral β-blocker is recommended in patients with a ST-segment–elevation myocardial infarction (STEMI). In this multicenter study, we evaluated the effects of a continuous administration of landiolol, an ultrashort-acting β-blocker, before primary percutaneous coronary intervention (PCI) on myocardial salvage and its safety in STEMI patients. A total of 47 Japanese patients with anterior or lateral STEMI undergoing a primary PCI within 12 h of symptom onset were randomized to receive intravenous landiolol (started at 3 μg/min/kg dose and continued to a total of 50 mg; n=23) or not (control; n=24). Patients with Killip class III or more were excluded. The primary outcome was the myocardial salvage index on cardiac magnetic resonance imaging (MRI) performed 5-7 days after the PCI. Cardiac MRI was performed in 35 patients (74%). The myocardial salvage index in the landiolol group was significantly greater than that in the control group (44.4±14.6% vs. 31.7±18.9%, respectively; p=0.04). There were no significant differences in adverse events at 24 h between the landiolol and control groups. A continuous administration of landiolol before a primary PCI may increase the degree of myocardial salvage without additional hemodynamic adverse effects within the first 24 h after STEMI

    Joint associations of physical activity and screen time with overweight among japanese adults

    Background: although both insufficient physical activity (PA) and high screen time (ST) are independent risk factors for obesity, how the combination of sufficient/insufficient PA and high/low ST could increase obesity risk among the adult population of Japan is not known. This study examined joint associations of PA and ST with overweight among Japanese adults. Methods: an Internet-based survey collected data on height, weight, self-reported time spent in PA and ST, and sociodemographic variables from 2832 adults. Respondents were categorized into sufficient PA/low ST, sufficient PA/high ST, insufficient PA/low ST, or insufficient PA/high ST categories as per public PA guidelines and the median of ST. Logistic regression analysis examined the odds ratios (OR) of being overweight (body mass index, &#8805; 25 kg/m2) according to the categories of PA and ST. Results: in comparison with the sufficient PA/low ST category, participants in the insufficient PA/high ST category were significantly more likely overweight (OR, 1.48; 95% confidence interval [95%CI), 1.14, 1.93) after adjusting for sociodemographic variables. A significantly higher OR for overweight (including obesity) among insufficient PA/high ST category was also observed in men, but no significant association was found in women. Conclusions: both insufficient PA and prolonged ST contribute to overweight and obesity among Japanese adults.     Paediatric HIV and elimination of mother-to-child transmission of HIV in the ASEAN region: a call to action

    Recent achievements in scaling up paediatric antiretroviral therapy (ART) have changed the life of children living with HIV, who now stay healthy and live longer lives. However, as it becomes more of a chronic infection, a range of new problems have begun to arise. These include the disclosure of HIV serostatus to children, adherence to ART, long-term toxicities of antiretroviral drugs and their sexual and reproductive health, which are posing significant challenges to the existing health systems caring for children with HIV with limited resources, experiences and capacities. While intensified efforts and actions to improve care and treatment for these children are needed, it is crucial to accelerate the prevention of mother-to-child transmission (PMTCT) of HIV, which is the main cause of paediatric HIV in the ASEAN region so as to eliminate the fundamental cause of the problem. This report argues that given over 70% of women have access to at least one antenatal care visit in the region and acceptance of HIV testing after receiving counselling on PMTCT could be as high as 90%, there is an opportunity to strengthen PMTCT services and eventually eliminate new paediatric HIV infections in the ASEAN countries
