Evaluation of animal models of neurobehavioral disorders
Animal models play a central role in all areas of biomedical research. The process of animal model building, development and evaluation has rarely been addressed systematically, despite the long history of using animal models in the investigation of neuropsychiatric disorders and behavioral dysfunctions. An iterative, multi-stage trajectory for developing animal models and assessing their quality is proposed. The process starts with defining the purpose(s) of the model, preferentially based on hypotheses about brain-behavior relationships. Then, the model is developed and tested. The evaluation of the model takes scientific and ethical criteria into consideration
Low Birth Weight Impairs Acquisition of Spatial Memory Task in Pigs
In commercial pig farming, an increasing number of low birth weight (LBW) piglets are born, due to selection for large litter sizes. While LBW piglets have a higher risk of pre-weaning mortality, a considerable number of these piglets survive to slaughter age. In humans, LBW is a risk factor for long-term cognitive impairments. In pigs, studies examining the post-weaning effects of LBW on cognition have reported contradictory results. Therefore, the current study aimed to assess the effects of LBW on cognitive development in pigs using an improved study design, by (1) testing a larger sample size than previous studies, (2) assessing acute and chronic stress responses to account for a potential altered stress response in LBW pigs, and (3) testing both female and male pigs to account for potential confounding effects of sex. Learning and memory of 20 LBW pigs and 20 normal birth weight (NBW) pigs, both groups consisting of 10 females and 10 males, were compared using a spatial holeboard task. In this task, pigs had to learn and remember the locations of hidden food rewards. After a pig had successfully acquired the task, it was presented with two successive reversal phases during which it was presented with a new configuration of reward locations. The holeboard allows for simultaneous assessment of working and reference memory, as well as measures of motivation, exploration, and behavioral flexibility. Mixed model ANOVAs revealed a transiently impaired reference memory performance of LBW pigs, implying they had more difficulty learning their reward configuration in the holeboard. Also, LBW piglets showed increased pre-weaning hair cortisol concentrations compared to their NBW siblings. No other effects of LBW were found. Sex had no direct or interaction effects on any measures of holeboard performance or stress. It is possible that the enriched housing conditions applied during our study had an ameliorating effect on our pigs' cognitive development. Overall, our results suggest LBW has a negative effect on post-weaning cognitive performance in pigs. This could have welfare consequences as cognitive skills are required for pigs to learn how to correctly respond to their environment
Toward assessing the role of dietary fatty acids in lamb's neurological and cognitive development
Understanding and measuring sheep cognition and behavior can provide us with measures to safeguard the welfare of these animals in production systems. Optimal neurological and cognitive development of lambs is important to equip individuals with the ability to better cope with environmental stressors. However, this development can be affected by nutrition with a special role from long-chain fatty acid supply from the dam to the fetus or in lamb's early life. Neurological development in lambs takes place primarily during the first two trimesters of gestation. Through late fetal and early postnatal life, the lamb brain has a high level of cholesterol synthesis. This rate declines rapidly at weaning and remains low throughout adulthood. The main polyunsaturated fatty acids (PUFA) in the brain are ω-6 arachidonic acid and ω-3 docosahexaenoic acid (DHA), which are elements of plasma membranes' phospholipids in neuronal cells. DHA is essential for keeping membrane integrity and is vital for normal development of the central nervous system (CNS), and its insufficiency can damage cerebral functions and the development of cognitive capacities. In sheep, there is evidence that supplying PUFA during gestation or after birth may be beneficial to lamb productive performance and expression of species-specific behaviors. The objective of this perspective is to discuss concepts of ruminant behavior and nutrition and reflect on future research directions that could help to improve our knowledge on how dietary fatty acids (FA) relate to optimal neurological and cognitive development in sheep

Climate change, public health, and animal welfare: towards a One Health approach to reducing animal agriculture’s climate footprint
Animal agriculture contributes significantly to global greenhouse gas (GHG) emissions—an estimated 12%-20% of total anthropogenic emissions. This has led both governmental and private actors to propose various ways to mitigate those climate impacts. This paper applies a One Health lens to the issue, arguing that the choice of solutions should not only consider the potential to reduce GHG emissions—which is not always a given—but also the implications for public health and animal welfare. With this perspective, we examine the potential public health and animal welfare impacts of three types of strategies that are often proposed: (1) “sustainable intensification” methods, aimed at maintaining or increasing production while limiting emissions and avoiding further land conversion; (2) “species shift” approaches, which focus on changing diets to consume meat from animals produced with lower GHG emissions instead of that of animals associated with higher emissions; and (3) “systemic dietary change” approaches that promote shifts towards whole plant-based foods or novel alternatives to conventional animal products. We discuss how some approaches—particularly those associated with sustainable intensification and species shift—could introduce new and significant risks to public health and animal welfare. Promoting systemic dietary change helps to overcome some of these challenges, but requires careful attention to equity to ensure that vulnerable populations still have access to the nutrients they need. We end with recommendations for a more holistic approach to reducing emissions from farmed animals that can help avoid trade-offs and increase synergies with other societal goals
Early-life interventions to prevent feather pecking and reduce fearfulness in laying hens
Severe feather pecking, the pulling out of feathers of conspecifics, is a major welfare issue in laying hens. Possible underlying causes are fearfulness and lack of foraging opportunities. Because early life is a crucial stage in behavioral development, adapting the incubation and rearing environment to the birds' needs may reduce fearfulness and prevent the development of feather pecking. In a 2 × 2 factorial design study, we investigated whether a green light-dark cycle throughout incubation, which resembles natural incubation circumstances more than the standard dark incubation, and foraging enrichment with live larvae during rearing reduce fearfulness and feather pecking and increase foraging behavior of laying hen pullets from an early age onwards. In this 2-batch experiment, 1,100 ISA Brown eggs were incubated under either 0 h of light/24 h of darkness or 12 h of green LED light/12 h of darkness. After hatching, 400 female chicks (200 per batch) were housed in 44 pens (8–10 chicks per pen). During the entire rearing phase (0–17 wk of age), half of the pens received black soldier fly larvae in a food puzzle as foraging enrichment. We assessed fear of novel objects and humans, feather pecking, plumage condition, foraging behavior, and recovery time after a 3-fold vaccination (acute stressor). A slight increase in the number of foraging bouts was only seen with larvae provisioning (rate ratio 1.19, 95% CI 1.02–1.29, P = 0.008). Neither lighted incubation nor larvae provisioning affected fearfulness, feather pecking, plumage condition or recovery time after vaccination. In conclusion, the present study showed no effects of light during incubation and minor effects of foraging enrichment during rearing on the behavior of laying hen pullets. Further research is recommended on other welfare aspects
Assessing learning and memory in pigs
In recent years, there has been a surge of interest in (mini) pigs (Sus scrofa) as species for cognitive research. A major reason for this is their physiological and anatomical similarity with humans. For example, pigs possess a well-developed, large brain. Assessment of the learning and memory functions of pigs is not only relevant to human research but also to animal welfare, given the nature of current farming practices and the demands they make on animal health and behavior. In this article, we review studies of pig cognition, focusing on the underlying processes and mechanisms, with a view to identifying. Our goal is to aid the selection of appropriate cognitive tasks for research into pig cognition. To this end, we formulated several basic criteria for pig cognition tests and then applied these criteria and knowledge about pig-specific sensorimotor abilities and behavior to evaluate the merits, drawbacks, and limitations of the different types of tests used to date. While behavioral studies using (mini) pigs have shown that this species can perform learning and memory tasks, and much has been learned about pig cognition, results have not been replicated or proven replicable because of the lack of validated, translational behavioral paradigms that are specially suited to tap specific aspects of pig cognition. We identified several promising types of tasks for use in studies of pig cognition, such as versatile spatial free-choice type tasks that allow the simultaneous measurement of several behavioral domains. The use of appropriate tasks will facilitate the collection of reliable and valid data on pig cognition
Effects of Maternal Care During Rearing in White Leghorn and Brown Nick Layer Hens on Cognition, Sociality and Fear
Both genetic background and maternal care can have a strong influence on cognitive and emotional development. To investigate these effects and their possible interaction, White Leghorn (LH) and Brown Nick (BN) chicks, two hybrid lines of layer hen commonly used commercially, were housed either with or without a mother hen in their first five weeks of life. From three weeks of age, the chicks were tested in a series of experiments to deduce the effects of breed and maternal care on their fear response, foraging and social motivation, and cognitive abilities. The LH were found to explore more and showed more attempts to reinstate social contact than BN. The BN were less active in all tests and less motivated than LH by social contact or by foraging opportunity. No hybrid differences were found in cognitive performance in the holeboard task. In general, the presence of a mother hen had unexpectedly little effect on behavior in both LH and BN chicks. It is hypothesized that hens from commercially used genetic backgrounds may have been inadvertently selected to be less responsive to maternal care than ancestral or non-commercial breeds. The consistent and strong behavioral differences between genetic strains highlights the importance of breed-specific welfare management processes
