    PRODUCTION OF POLYCLONAL ANTIBODIES AGAINST RECOMBINANT VP6 ANTIGEN OF ROTAVIRUS

    Rotavirus is the most common cause of diarrhea in infants and young children under five years old. The VP6 protein of rotavirus was considered as an important antigen in the development of novel vaccine generation and diagnostic tools. Polyclonal antibodies of VP6 antigen could be used to generate a rapid test based on the lateral flow immunoassay. In the present study, we showed the results of synthesis of polyclonal antibodies against recombinant VP6 protein of rotavirus. Recombinant VP6 protein was strongly produced by E. coli in suitable conditions. The properties of recombinant VP6 protein were characterized by Western blot and ELISA methods. Pure recombinant VP6 protein was used as an antigen to inject into the rabbits. The recombinant VP6 protein was highly immunogenic in rabbits. Anti-VP6 antibodies were purified from the rabbit serum using the Protein A affinity chromatography. The specificity of the antibody towards its antigen was characterized by Western blot and ELISA immunoassay. The purity of antibody was reached as 97 %. The efficiency of antibody production in rabbit was 3.94 mg/ml serum. The amount and quality of the obtained anti-rotavirus VP6 antibody was suitable for application in the development of the diagnostic kits

    Stable isotopes as an effective tool for N nutrient source identification in a heavily urbanized and agriculturally intensive tropical lowland basin

We present the application of dual stable isotope analyses of NO3 (δ15N-NO3 and δ18O-NO3) to provide a comprehensive assessment of the provenance, partitioning, and conversion of nitrate across the Day River Basin (DRB), Vietnam, which is heavily impacted by agriculture and urbanization. Stable isotope compositions of river water δ18O-H2O, in addition to their δ15N-NO3 and δ18O-NO3 signatures, were sampled at 12 locations in the DRB. Sample collection was conducted during three different periods to capture changes in regional weather and agricultural fertilization regimes; April (the dry season and key fertilization period), July (the rainy season and another key fertilization period) and October (the rainy season with no regional fertilization). Ranges of NO3 stable isotopes are − 7.1 to + 9.2‰ and − 3.9 to + 13.2‰ for δ18O and δ15N, respectively. Interpretation of the stable isotope data characterizes 4 main sources of NO3 in the DRB; (1) nitrified urea fertilizer derived from an intensive agricultural irrigation network, (2) soil and groundwater leaching from within the basin (3) manure and sewage inputs (which is more prevalent in downstream river sections) and (4) upstream inflow from the Red River which discharges into the Day River through the Dao River. We applied a mixing model for the DRB consisting of 4 variables, representing these 4 different sources. The partition calculation shows that during the fertilization and rainy period of July, more than 45% of river NO3 is derived from nitrified urea sources. During the other sampling periods (April and October), manure and sewage contribute more than 50% of river NO3 and are derived from the middle portion of the DRB, where the Day River receives domestic wastewater from the Vietnamese capital, Hanoi. Stable isotope data of O and N reveal that nitrification processes are more prevalent in the rainy season than in dry season and that this predominantly takes place in paddy field agricultural zones. In general, data demonstrate that nitrate loss in the DRB is due to denitrification which takes place in polluted stretches of the river and dominates in the dry season. This study highlights that (i) domestic waste should be treated prior to its discharge into the Day River and (ii) the need for better catchment agricultural fertilization practices as large portions of fertilizer currently discharge into the river, which greatly impacts regional water quality

    Using stable isotopes to estimate young water fractions in a heavily regulated, tropical lowland river basin

    The young water fraction of streamflow (Fyw), an important hydrological variable, has been calculated for the first time, for a monsoon‐fed coastal catchment in northern Vietnam. Oxygen stable isotopes (δ18O) from six river sites in the Day River Basin (DRB) were analysed monthly, between January 2015 and December 2018. River δ18O signatures showed sine wave variability, reflecting the amount effect and tropical (dry‐rainy) seasonality of the region. The δ18O composition of precipitation ranged from −12.67 to +1.68‰, with a mean value of −5.14‰, and in‐streamflow signatures ranged from −11.63 to −1.37‰ with a mean of −5.02‰. Fractions of young water (Fyw) were calculated from the unweighted and flow‐weighted δ18O composition of samples. Unweighted Fyw ranged between 29 ± 8% and 82 ± 21% with a mean value of 51 ± 19%, and was not significantly different from flow‐weighted Fyw (range between 33 ± 25% and 92 ± 73%, mean 52 ± 36%). Both unweighted and flow‐weighted Fyw were highest in the middle of stream and lowest in downstream sites, capturing the impacts of landuse changes, hydrology and human activities in the catchment. Our calculations imply that more than a half of rainwater reaches the DRB river mainstream within the first 3 months. The Fyw is much higher than the global average (of one‐third) and insensitive to discharge due to the combination of a humid catchment with high rainfall, low storage capacity, flat landscape and an intensive drainage system in the DRB. Also the low discharge sensitivity of Fyw in the DRB implies that the regional hydrology is severely altered by humans

    Feature selection methods and sampling techniques to financial distress prediction for Vietnamese listed companies

    The research is taken to integrate the effects of variable selection approaches, as well as sampling techniques, to the performance of a model to predict the financial distress for companies whose stocks are traded on securities exchanges of Vietnam. A firm is financially distressed when its stocks are delisted as requirement from Vietnam Stock Exchange because of making a loss in 3 consecutive years or having accumulated a loss greater than the company’s equity. There are 12 models, constructed differently in feature selection methods, sampling techniques, and classifiers. The feature selection methods are factor analysis and F-score selection, while 3 sets of data samples are chosen by choice-based method with different percentages of financially distressed firms. In terms of classifying technique, logistic regression together with SVM are used in these models. Data are collected from listed firms in Vietnam from 2009 to 2017 for 1, 2 and 3 years before the announcement of their delisting requirement. The experiment’s results highlight the outperformance of the SVM model with F-score selection method in a data sample containing the highest percentage of non-financially distressed firms

    Dry Eyes Status on Des Scale and Related Factors in Outpatients at Vietnam National Institute of Ophthalmology

    BACKGROUND: Dry eye (DE) can effect on quality of life by pain, inability to perform certain activities that require prolonged attention (driving, reading,…) and productivity at work and finally effect to Q0L associated with DE. OSDI is scale questionnaire is created team to measure the quality of life related to ocular surface disease. AIM: To describe the dry eye disease according to OSDI scale and related factors of this disease. METHODS: A cross-sectional descriptive study was carried out on outpatients (≥ 16-year-old) who were examined and diagnosed with dry eyes at Vietnam National Institute Of Ophthalmology from April to July 2018. Data was collected using the OSDI questionnaire. RESULTS: The average age of participants was 44.6 years; 80.9% of patients were female; 39.9% were identified having mild dry eye. The related factors have been identified that associated with severe dry eye, including age OR = 1.03 (95%CI: 1.01-1.05, p = 0.005), binocular good vision OR = 0.11 (95%CI: 0.05-0.23; p < 0.0001), medical history OR = 17.09 (95%CI: 2.24-130.25; p < 0.0001), chronic conjunctivitis OR = 0.36 (95%CI: 0.14-0.91; p = 0.027), refractive errors OR = 0.14 (95%CI: 0.04-0.48; p < 0.0001), Sjogren's syndrome OR = 31.13 (95%CI: 7.08-136.76; p < 0.0001). CONCLUSION: Several related factors have been identified associated with severe dry eye, including: age, binocular good vision, medical history, chronic conjunctivitis, refractive errors, Sjogren's syndrome

    Transformation Chlorophyll a of Spirulina platensis to Chlorin e6 Derivatives and Several Applications

    BACKGROUND: Spirulina platensis contains a large amount of chlorophylls, chlorophyll a, that are starting materials to synthesize functionalized chlorins. Chlorin e6 (Ce6) as well as its derivatives are second generation sensitizers using in photodynamic therapy (PDT) of various cancers. In this study, we transfer chlorophyll a of S. platensis to Ce6derivatives and determine their several applications. AIM: to evaluate the effects of Ce6 derivatives to treat cancer cells. METHODS: Ce6 trimethylester was created from methyl pheophorbide a2 in S. platensis provided by the Hidumi Company, Nghe An province, Viet Nam. Hela cells were incubated with Ce6 trimethylester and the irradiated with the diode laser dose of 1.2 J/cm2/min through the system of filters £ 650 nm. MTT assay and clonogenic assay were used to determine survival rate and cloning efficiency of cells. Antimicrobial effect of Ce6 trimethylester with halogen light were studied with Propionibacterium acnes VTCC 0218 and Staphylococcus aureus VTCC 0173. RESULTS: From dry biomass (700 g) of S. platensis, after extracting chlorophyll a and methanolysis, 4.2 g of methyl pheophorbide a was obtained. The reaction to give Ce6 trimethylester with 82% yield was performed with potassium hydroxide (KOH) in MeOH/THF/CHCl3. After irradiation with a 650 nm laser at 1.2 J, the cell viability in all samples decreased with Ce6 trimethylester treatment, the survival declining trend of Hela cells treated with Ce6 trimethylester were proportional when concentration of Ce6 trimethylester increased. The rate of colony formation was declined as the concentration of Ce6 trimethylester treated was increased. The growth of both S. aureus and P. acnes can be inactivated by Ce6 trimethylester PDT. The MIC99 value against P. acnes VTCC 0218 and S. aureus VTCC 0173 of Ce6 trimethylester with halogen light was 1.25 μg/ml. CONCLUSION: The Ce6 trimethylester from S. platensis cultivated in Viet Nam could be used as a potential photosentizer for photodynamic therapy for treatment of cancer and acne

    Identifying the controls on nitrate and metabolic state within the Red River delta (Vietnam) with the use of stable isotopes

    In many places around the world, anthropogenic activities have resulted in nitrate (NO3−) pollution and changes in the metabolic state of aquatic ecosystems. Here we combined stable isotope and physico-chemical monitoring to assess the sources of NO3− and the overall metabolic state within the Red River delta, Vietnam. River water stable isotope compositions (δ18O-H2O) ranged between −11.2 and −2.7 ‰, δ18O-NO3− between −7.1 and + 29.7 ‰ and δ15N-NO3− between −3.9 and + 14.0 ‰. We identified the dominant NO3− sources as: 1) soil leachate, 2) domestic waste flushed from urban areas, and 3) NH4+ fertilizers washed from paddy fields. The relative impact of each source depends on geographical location within the delta and the time of year, due to dilution and concentration effects during wet and dry seasons. The primary NO3−source upstream is natural soil leachates, predominantly from tributaries connected to the Red River’s main stream. Within the middle-lower section of Red River delta, urban pollution from manure and septic waste reaches as high as 50 % of the total NO3− load during dry season. NO3− leached from fertilizers is also high at sites in the middle of the delta, related to agricultural activities. Dissolved oxygen isotope (δ18O-O2) values calculated from δ18O-H2O and δ18O-NO3− values indicate that the aquatic metabolism is net autotrophic (oxygen from primary production exceeds consumption by respiration), but high inputs of biodegradable organic matter from untreated domestic waste and high rates of sediment oxygen demand (SOD) and chemical oxygen demand (COD) have resulted in the whole river system becoming undersaturated in oxygen.
