The use of FD-filamentous bacteriophage for the in vivo imaging of cancer

University of Missouri-Columbia 2006.Improved diagnosis of cancer through targeted imaging of neoplasms is needed for early detection and treatment of cancer. We propose that the physical characteristics of filamentous bacteriophage (phage) are well suited for use in the targeted imaging of cancer. This study describes the development of two different phage-based cancer imaging agents. One is specific for prostate carcinoma, and the other specific for melanoma, with each utilizing different imaging modalities - optical imaging and radioimaging. A human PC-3 prostate carcinoma was optically imaged using phage displaying a newly selected prostate homing peptide labeled with the near infrared fluorophore AF680, whereas, a mouse B16-F1 melanoma was radioimaged using single-photon-emission-computed-tomography through a two-step pretargeting procedure. This pretargeting strategy utilized bifunctional bacteriophage that were both biotin labeled and displayed [alpha]-melanoma stimulating hormone peptide analogs and included streptavidin-chelator complex radiolabeled with 111In. The successful imaging of prostate carcinoma and melanoma with different strategies exhibits the versatility of phage and highlights their utility in the imaging of cancer
THE Hα EMISSION OF NEARBY M DWARFS AND ITS RELATION TO STELLAR ROTATION
The high-energy emission from low-mass stars is mediated by the magnetic dynamo. Although the mechanisms by which fully convective stars generate large-scale magnetic fields are not well understood, it is clear that, as for solar-type stars, stellar rotation plays a pivotal role. We present 270 new optical spectra of low-mass stars in the Solar Neighborhood. Combining our observations with those from the literature, our sample comprises 2202 measurements or non-detections of Hα emission in nearby M dwarfs. This includes 466 with photometric rotation periods. Stars with masses between 0.1 and 0.6 M[subscript â] are well-represented in our sample, with fast and slow rotators of all masses. We observe a threshold in the massâperiod plane that separates active and inactive M dwarfs. The threshold coincides with the fast-period edge of the slowly rotating population, at approximately the rotation period at which an era of rapid rotational evolution appears to cease. The well-defined active/inactive boundary indicates that Hα activity is a useful diagnostic for stellar rotation period, e.g., for target selection for exoplanet surveys, and we present a mass-period relation for inactive M dwarfs. We also find a significant, moderate correlation between L[suscript Hα]/L[subscript bol] and variability amplitude: more active stars display higher levels of photometric variability. Consistent with previous work, our data show that rapid rotators maintain a saturated value of LHα/Lbol. Our data also show a clear power-law decay in L[subscript Hα]/L[subscript bol] with Rossby number for slow rotators, with an index of â1.7 ± 0.1.National Science Foundation (U.S.). Gymnosperms Demonstrate Patterns of Fine-root Trait Coordination Consistent with the Global Root Economics Space
Gymnosperms encompass a diverse group of mostly woody plants with high ecological and economic value, yet little is known about the scope and organization of fine-root trait diversity among gymnosperms due to the undersampling of most gymnosperm families and the dominance of angiosperm groups in recent syntheses. New and existing data were compiled for morphological traits (root diameter, length, tissue density, specific root length [SRL] and specific root area [SRA]), the architectural trait branching ratio, root nitrogen content [N] and mycorrhizal colonization. We used phylogenetic least squares regression and principal component analysis to determine trait-trait relationships and coordination across 66 species, representing 11 of the 12 extant gymnosperm families from boreal, temperate, subtropical and tropical biomes. Finally, we compared the relationship between family divergence time and mean trait values to determine whether evolutionary history structured variation in fine-root traits within the gymnosperm phylogeny. Wide variation in gymnosperm root traits could be largely captured by two primary axes of variation defined by SRL and diameter, and root tissue density and root nitrogen, respectively. However, individual root length and SRA each had significant correlations with traits defining both main axes of variation. Neither mycorrhizal colonization nor root branching ratio were closely related to other traits. We did not observe a directional evolution of mean trait values from older to more recently diverged gymnosperm families. Synthesis. Despite their unique evolutionary history, gymnosperms display a root economic space similar to that identified in angiosperms, likely reflecting common constraints on plants adapting to diverse environments in both groups. These findings provide greater confidence that patterns observed in broad syntheses justly capture patterns of trait diversity among multiple, distinct lineages. Additionally, independence between root architecture and other traits may support greater diversity in below-ground resource acquisition strategies. Unlike angiosperms, there were no clear trends towards increasingly thin roots over evolutionary time, possibly because of lower diversification rates or unique biogeographic history among gymnosperms, though additional observations are needed to more richly test evolutionary trends among gymnosperms
Are elites really less fair-minded?
US power âelitesâ are substantially less fair-minded than ânon-eliteâ general populations claims a study by Ray Fisman and coauthors [Science 349, 6254 (2015)]. This supposedly explains why US governments, run by people less fair than the citizens they represent, have been uninclined to tackle inequality. We critically replicated the study, because different experimental protocols for âelitesâ (interactive variants) and ânon-elitesâ (standard dictator games) were used to measure preferences. We find that the protocol potpourri drove the conclusion: under standard protocols, we find no significant class differences, if any opposite ones. Our work highlights the risk of producing false positives when ex post results appear invitingly plausible
Is shape in the eye of the beholder? Assessing landmarking error in geometric morphometric analyses on live fish
Geometric morphometrics is widely used to quantify morphological variation between biological specimens, but the fundamental influence of operator bias on data reproducibility is rarely considered, particularly in studies using photographs of live animals taken under field conditions. We examined this using four independent operators that applied an identical landmarking scheme to replicate photographs of 291 live Atlantic salmon (Salmo salar L.) from two rivers. Using repeated measures tests, we found significant inter-operator differences in mean body shape, suggesting that the operators introduced a systematic error despite following the same landmarking scheme. No significant differences were detected when the landmarking process was repeated by the same operator on a random subset of photographs. Importantly, in spite of significant operator bias, small but statistically significant morphological differences between fish from the two rivers were found consistently by all operators. Pairwise tests of angles of vectors of shape change showed that these between-river differences in body shape were analogous across operator datasets, suggesting a general reproducibility of findings obtained by geometric morphometric studies. In contrast, merging landmark data when fish from each river are digitised by different operators had a significant impact on downstream analyses, highlighting an intrinsic risk of bias. Overall, we show that, even when significant inter-operator error is introduced during digitisation, following an identical landmarking scheme can identify morphological differences between populations. This study indicates that operators digitising at least a sub-set of all data groups of interest may be an effective way of mitigating inter-operator error and potentially enabling data sharing

Carbohydrate antigens in nipple aspirate fluid predict the presence of atypia and cancer in women requiring diagnostic breast biopsy
<p>Abstract</p> <p>Background</p> <p>The goal of this prospective study was to determine (a) concentrations of the carbohydrate biomarkers Thomsen Friedenreich (TF) antigen and its precursor, Tn antigen, in nipple discharge (ND) collected from women requiring biopsy because of a suspicious breast lesion; and (b) if concentration levels predicted pathologic diagnosis.</p> <p>Methods</p> <p>Adult women requiring biopsy to exclude breast cancer were enrolled and ND obtained. The samples from 124 women were analyzed using an anti-TF and anti-Tn monoclonal antibodies in direct immunoassay.</p> <p>Results</p> <p>The highest median concentration in ND for TF and Tn was in women with ductal carcinoma <it>in situ </it>(DCIS). TF was higher in women with 1) cancer (DCIS or invasive) vs. either no cancer (atypia or benign pathology, p = .048), or benign pathology (p = .018); and 2) abnormal (atypia or cancer) versus benign pathology (p = .016); and was more predictive of atypia or cancer in post- compared to premenopausal women. Tn was not predictive of disease. High TF concentration and age were independent predictors of disease, correctly classifying either cancer or abnormal vs. benign pathology 83% of the time in postmenopausal women.</p> <p>Conclusions</p> <p>TF concentrations in ND were higher in women with precancer and cancer compared to women with benign disease, and TF was an independent predictor of breast atypia and cancer. TF may prove useful in early breast cancer detection.</p
Lake shape and the characteristics of migration behavior modify Atlantic salmon smolt migration success through lakes
Migration is a high-risk behavior. For the Atlantic salmon, Salmo salar, migrating from its river nursery area to marine feeding grounds, the magnitude of risk varies with habitat type. Passage through lakes, in particular, is associated with low rates of migration success. Downstream migrating salmon smolts are rheotactic when migrating in rivers, but lakes typically provide poorer directional currents for migrating salmon. In this study we tested if, in the absence of clear navigational cues in lakes, Atlantic salmon smolts switch to a random search strategy to find the outflowing river. We constructed random search simulations to test if lake basin shape has an effect on migration success. We also compared simulated migration characteristics with migrations of salmon smolts through five real lakes for which there are telemetry data for migrating salmon. Correlated random walk simulations showed that a random search strategy could be successful for all lake shapes tested but was more successful in curved (round and elliptical) than rectangular basin shapes. Rectangular basins with the migration start and stop points at the ends of the lake had a higher success than those where these points were perpendicular to the axis of the lake. In general, a random walk model predicted the migration success rate of fish tracked through real lakes. However, for two lakes the simulated migration success exceeded that of actual success, suggesting that fish passing through these lakes were not adopting a random search strategy. We speculate that this is the result of either conflicting navigational cues which inhibit smolts from finding the lake outlet or that they temporarily suspend migration (e.g., to feed). Modelling predicted that for small lakes, directional swimming in short steps (ca. 100âm) followed by turns with very low variation from the direction of travel resulted in the highest migration success. For larger lakes, longer step lengths but also with low turn variation (simulated turning angle drawn from distributions of standard deviation 2° and 5° around a mean of 0°) resulted in the highest migration success. We conclude that navigation in downstream migrating salmon smolts switches from rheotaxis in rivers to a random search tactic in lakes except where residual flow cues in some lakes prevent this, at times resulting in suboptimal navigation outcomes
The Contribution of HI-Rich Galaxies to the Damped Absorber Population at z=0
We present a study of HI-rich galaxies in the local universe selected from
blind emission-line surveys. These galaxies represent the emission-line
counterparts of local damped Lyman-alpha systems. We find that the HI
cross-section of galaxies is drawn from a large range of galaxy masses below
M_star, 66% of the area comes from galaxies in the range 8.5 < Log M_star <
9.7. Both because of the low mass galaxy contribution, and because of the range
of galaxy types and luminosities at any given HI mass, the galaxies
contributing to the HI cross-section are not exclusively L_star spirals, as is
often expected. The optical and near infrared counterparts of these galaxies
cover a range of types (from spirals to irregulars), luminosities (from L_star
to <0.01 L_star), and surface brightnesses. The range of optical and near
infrared properties as well as the kinematics for this population are
consistent with the properties for the low-z damped Lyman-alpha absorbers. We
also show that the number of HI-rich galaxies in the local universe does not
preclude evolution of the low-z damped absorber population, but it is
