    Comparison of HER2 Expression in Primary Tumor and Disseminated Tumor Cells in the Bone Marrow of Breast Cancer Patients

    Objective: The aim of this study was to measure the human epidermal growth factor receptor 2 (HER2) status of disseminated tumor cells (DTCs) from bone marrow (BM) aspirates and to assess correspondence or discrepancy with the primary tumor. Methods: DTCs were isolated from the BM of 156 breast cancer patients. Cytokeratin-positive DTCs were further analyzed by the chromogenic in situ hybridization method to detect HER2 gene amplification. Results: A significant correlation (p = 0.021) was found between the HER2 status of DTCs and the primary tumors. Sixty-one (68.5%) patients had a corresponding status. However, a shift of phenotype between primary tumor and DTCs was found in the remaining patients. Conclusion: This study showed a significant grade of discordance of the HER2 status between primary tumors and DTCs in the BM of a relevant subgroup of patients. Detection of HER2 amplification on DTCs could therefore help to better stratify patients for a more tailored therapy, since they would benefit from a HER2-targeted therapy. (C) 2016 S.     Rhodamine-Tagged Polymethacrylate Dyes as Alternative Tools for Analysis of Plant Cells

    A rhodamine B (RhB)-based initiator for atom transfer radical polymerization (ATRP) was synthesized and applied for preparation of poly(2-trimethylammoniumethyl methacrylate) (PChMA), poly(2-hydroxyethyl methacrylate) (PHEMA) and poly(2-trimethylsilyloxyethyl methacrylate) (PHEMATMS). Polymer fluorescence was confirmed by determination of quantum yield by comparative method with piroxicam as the standard exhibiting dependency of emission intensity on the polymer chain hydrophilicity and the kind of solvent. The RhB functionalized polymers were used for biological tests in plant materials except for RhB-PHEMATMS because of weak fluorescence. These two polymers slightly differed in cellular localization. RhB-PChMA was mostly observed in cell walls of root tissues and cotyledon epidermis. It was also observed in cytoplasm and cell organelles of root cap cells and rhizodermis, in contrast with cytoplasm of cotyledon epidermis. RhB-PHEMA was also present in apoplast. A strong signal in protoxylem cell walls and a weak signal in cell walls of rhizodermis and cortex were visible. Moreover, it was also present in cell walls of cotyledon epidermis. However, RhB-PHEMA was mostly observed in cytoplasm and cell organelles of all root tissues and epidermis of cotyledons. Both RhB-polymers did not cause cell death which means that they can be used in living plant material

    Mercury (Hg) is a pervasive contaminant that can adversely affect predatory wildlife. Bird eggs provide insights into breeding females' Hg burdens, and are easily collected and archived. We present data on Hg trends in herring gull (Larus argentatus) eggs from five sites in Atlantic Canada from 1972 to 2008. We found a significant decrease in Hg at Manawagonish Island, New Brunswick and Île du Corossol, Quebec, but after correcting Hg for dietary shifts using stable isotopes (δ15N), these trends disappeared. Decreasing temporal trends of stable isotopes in gull eggs were observed at four sites, suggesting shifts in gull diets. At Gull Island, Newfoundland, diet-adjusted Hg increased from 1977 to 1992, dropped sharply between 1992 and 1996, and rose again from 1996 to 2008. After adjusting Hg trends for dietary shifts of herring gulls, it appears that environmental Hg in coastal ecosystems has remained relatively constant at most sites in Atlantic Canada over the last 36 years
