    The effect of diatomaceous earth in live, attenuated infectious bronchitis vaccine, immune responses, and protection against challenge.

    Live virus vaccines are commonly used in poultry production, particularly in broilers. Massive application and generation of a protective local mucosal and humoral immunity with no adverse effects is the main goal for this strategy. Live virus vaccines can be improved by adding adjuvants to boost mucosal innate and adaptive responses. In a previous study we showed that diatomaceous earth (DE) can be used as adjuvant in inactivated vaccines. The aim of this study was to test DE as adjuvant in an Ark-DPI live infectious bronchitis virus (IBV) vaccine after ocular or spray application. Titrating the virus alone or after addition of DE showed that DE had no detrimental effect on the vaccine virus. However, adding DE to the vaccine did not induce higher IgG titers in the serum and IgA titers in tears. It also did not affect the frequency of CD4+ T cells, CD8+ T cells and monocytes/macrophages in the blood and the spleen determined by flow cytometry. In addition, protection generated against IBV homologous challenges, measured by viral load in tears, respiratory signs and histopathology in tracheas, did not vary when DE was present in the vaccine formulation. Finally, we confirmed through our observations that Ark vaccines administered by hatchery spray cabinet elicit weaker immune responses and protection against an IBV homologous challenge compared to the same vaccine delivered via ocular route

    DRIVE-THRU PHARMACY SERVICE: ASSESSMENT OF PERCEPTION AMONG PATIENTS OR CAREGIVERS IN HOSPITAL RAJA PEREMPUAN ZAINAB II

    Objective: The aim of this study was to evaluate the perception of patients or caregivers on drive-thru pharmacy in Raja Perempuan Zainab II Hospital (HRPZ II), Kelantan.Methods: A cross-sectional study was conducted from July to September 2014 on 387 patients or caregivers recruited from outpatient pharmacy, HRPZ II using a constructed survey form. The questionnaire was validated through a pilot study which yielded a Cronbach's alpha score of 0.897. It comprised common socio-demographic characteristics as well as 3 domains which described subject's experience in outpatient pharmacy, subject's perception of drive-thru pharmacy and subject's willingness to participate in the service.Results: More than half of respondents (n=231, 59.7 %) were aware of the existence of drive-thru pharmacy service. Almost all patients and caregivers (n=349, 90.2 %) perceived the service as useful to the public. Only a handful of them were reluctant to sign up, mainly due to the perception that it would cause lack of interaction between patients and pharmacists (n=16, 4.1%). Nearly three-quarters of participants (n=288; 74.4%) informed that they had never been asked to register as drive-thru pharmacy clients. They claimed that they were willing to use the service should it was offered to them (n=301; 77.8 %).Conclusion: This survey managed to garner positive feedbacks on drive-thru pharmacy. Most of the subjects perceived it as beneficial and were willing to use the service.Â

    The influence of persistent pathogens on circulating levels of inflammatory markers: A cross-sectional analysis from the Multi-Ethnic Study of Atherosclerosis

    Background: Systemic inflammation is linked to cardiovascular risk, but the influence of persistent pathogens, which are conventionally dichotomously categorized, on circulating levels of inflammatory markers is not clear. Antibody levels of pathogens have not been examined in relation to inflammation. Methods. Using data from a subsample of the Multi-Ethnic Study of Atherosclerosis, we examined circulating levels of interleukin-6 (IL-6), C-reactive protein (CRP) and fibrinogen in relation to five common persistent pathogens: cytomegalovirus, herpes simplex virus-1, Hepatitis A virus, Helicobacter pylori and Chlamydia pneumoniae. We tested the hypothesis that the number of seropositive pathogens (based on conventional cut-off points) would not be as sensitive a marker of inflammation as immune response measured by antibody levels to pathogens. Results. High antibody response to multiple pathogens showed graded and significant associations with IL-6 (p < 0.001), CRP (p = 0.04) and fibrinogen (p = 0.001), whereas seropositive pathogen burden did not. In multiple linear regression models, high antibody response to multiple pathogens maintained a positive association only with IL-6 (4.4% per pathogen exhibiting high antibody response, 95% CI 0.0-8.9). Conclusions. High antibody response to pathogens was a more consistent marker of inflammatory outcomes compared to seropositivity alone and high antibody response to multiple pathogens was a stronger marker compared to any single pathogen

    Solvent dependence of the rheological properties in hydrogel magnetorheological plastomer

    Chemically crosslinked hydrogel magnetorheological (MR) plastomer (MRP) embedded with carbonyl iron particles (CIPs) exhibits excellent magnetic performance (MR effect) in the presence of external stimuli especially magnetic field. However, oxidation and desiccation in hydrogel MRP due to a large amount of water content as a dispersing phase would limit its usage for long‐term applications, especially in industrial engineering. In this study, different solvents such as dimethyl sulfoxide (DMSO) are also used to prepare polyvinyl alcohol (PVA) hydrogel MRP. Thus, to understand the dynamic viscoelastic properties of hydrogel MRP, three different samples with different solvents: water, DMSO, and their binary mixtures (DMSO/water) were prepared and systematically carried out using the oscillatory shear. The outcomes demonstrate that the PVA hydrogel MRP prepared from precursor gel with water shows the highest MR effect of 15,544% among the PVA hydrogel MRPs. However, the samples exhibit less stability and tend to oxidise after a month. Meanwhile, the samples with binary mixtures (DMSO/water) show an acceptable MR effect of 11,024% with good stability and no CIPs oxidation. Otherwise, the sample with DMSO has the lowest MR effect of 7049% and less stable compared to the binary solvent samples. This confirms that the utilisation of DMSO as a new solvent affects the rheological properties and stability of the samples

    The Antimicrobial Peptide, LL-37, Inhibits in vitro Osteoclastogenesis

    Uncoupled bone resorption leads to net alveolar bone loss in periodontitis. The deficiency of LL-37, the only human antimicrobial peptide in the cathelicidin family, in patients with aggressive periodontitis suggests that LL-37 may play a pivotal role in the inhibition of alveolar bone destruction in periodontitis. We aimed to investigate a novel function of LL-37 in osteoimmunity by blocking osteoclastogenesis in vitro. Human osteoclast progenitor cells were isolated from a buffy coat of blood samples. The cells were cultured in the presence of various concentrations of LL-37 during an in vitro induction of osteoclastogenesis. Non-toxic doses of LL-37 could block multinuclear formation of the progenitor cells and significantly diminish the number of tartrate-resistant acid-phosphatase-positive cells and the formation of resorption pits (p < 0.05), whereas these concentrations induced cellular proliferation, as demonstrated by increased expression of proliferating cell nuclear antigen. Expression of several osteoclast genes was down-regulated by LL-37 treatment. It was demonstrated that nuclear translocation of nuclear- factor-activated T-cells 2 (NFAT2) was blocked by LL-37 treatment, consistent with a significant reduction in the calcineurin activity (p < 0.005). Collectively, our findings demonstrate that LL-37 inhibits the in vitro osteoclastogenesis by inhibiting the calcineurin activity, thus preventing nuclear translocation of NFAT2.     Rheological and resistance properties of magnetorheological elastomer with cobalt for sensor application

    Cobalt particles have been introduced as a filler due to the advantages of embedding their magnetic and electrical properties in magnetorheological elastomer (MRE). In the present research, the rheology and resistance of MRE are experimentally evaluated. Isotropic and anisotropic MRE samples containing silicone rubber and cobalt particles were fabricated. The magnetic properties of MRE are conducted using a vibrating sample magnetometer (VSM). The morphological aspects of MRE are observed by using field emission scanning electron microscopy (FESEM) and characterized by energy-dispersive X-ray spectroscopy (EDX). Rheological properties under various magnetic field strengths were measured for the magnetic field, strain amplitude, and frequency sweep test by using a parallel-plate rheometer. Subsequently, the resistance of MRE is tested under different applied forces and magnetic fields. The MRE storage modulus depicted an enhancement in field-dependent modulus across all the applied magnetic fields. The electrical resistance generated from the sample can be manipulated by external magnetic fields and mechanical loads. The conductivity of MRE is due to the existence of cobalt arrangements observed by FESEM. By introducing cobalt as filler and obtaining satisfactory results, the study might open new avenues for cobalt to be used as filler in MRE fabrication for future sensing applications
