Synthese und Verarbeitung Nanoskopie-relevanter Farbstoffe auf der Basis von Bisthiophenethenen
Im Fokus dieser Arbeit steht die Substanzklasse der Bisthiophenethene (kurz BTEs), bei
welchen es sich um schaltbare MolekĂŒle handelt. Durch Ă€uĂere Stimuli, in diesem Fall
Bestrahlung, können diese Verbindungen reversibel zwischen zwei verschiedenen ZustÀnden
wechseln. Beide Formen unterscheiden sich in ihren physikalischen Eigenschaften, wobei die
FarbĂ€nderung der geschlossenen Form die elementarste Ănderung darstellt.
Das Ziel dieser Arbeit ist die gezielte Synthese von BTE-Farbstoffen, welche eine hohe
IntensitÀt der geschlossenen Form, ein schnelles Ansprechverhalten beider Reaktionen mit
sichtbarem Licht und eine hohe Anzahl an Schaltzyklen aufweisen. Die BTEs werden in einer
PMMA-Matrix eingebettet und in dĂŒnnen, homogenen Filmen rotationsbeschichtet. Im
Rahmen des NanoVidere-Projekts dienen diese PrĂ€parate als Detektionsmechanismus fĂŒr ein
hochauflösendes Konfokalmikroskop, welches Details unterhalb der Beugungsgrenze auflösen
kann.
ZunÀchst wird der Einfluss des Substitutionsmusters auf die BTEs untersucht. Es konnte ein
linearer Zusammenhang zwischen dem Substituenten und dem daraus resultierenden UV/Vis-Spektren festgestellt werden. DafĂŒr wird die spektroskopische HAMMETT-Funktion auf die
Farbstoffe angewendet. Somit können durch die HAMMETT-BROWN-Konstanten erstmals
spektroskopische Eigenschaften der BTEs vorhergesagt werden.
AuĂerdem zeigt sich, dass sich das Verhalten der BTE-Farbstoffe in Lösung oder in der
kristallinen Phase nicht auf die PMMA-Umgebung ĂŒbertragen lĂ€sst. Substitutionen des
GrundgerĂŒsts der BTEs, welche als stabilitĂ€tsfördernd gelten, zeigen ein nahezu exakt
gegensÀtzliches Verhalten, wenn diese im Film eingebettet sind. Es handelt sich demnach um
eine Umgebung, in welcher das Verhalten der BTEs grundlegend neu verstanden werden muss.
Durch neue AnsÀtze in der Synthese konnten so sukzessive neue Erkenntnisse gewonnen und
angewendet werden, sodass es gelang eine umfangreiche Grundlage fĂŒr das Verhalten der
Farbstoffe in Polymermatrizen zu gewinnen. Es konnte gezeigt werden, welchen Einfluss
verschiedene Substituenten auf die FarbintensitÀt, sowie auf die Farbe selbst, als auch das
Ansprech- und StabilitĂ€tsverhalten ausĂŒben. So gelang es Farbstoffe herzustellen, welche die
geforderten Eigenschaften erfĂŒllen und sich somit fĂŒr den Einsatz in der bildgebenden Super-Resolutions-Mikroskopie eignen.This thesis focuses on the substance class of bisthiophenethenes (BTEs for short), which are
switchable molecules. External stimuli, in this case irradiation, can cause these compounds to
switch reversibly between two different states. Both forms differ in their physical properties,
with the color change of the closed form representing the most elementary change.
The aim of this work is the targeted synthesis of BTE dyes that exhibit a high intensity of the
closed form, a fast response of both reactions with visible light and a high number of switching
cycles. The BTEs are embedded in a PMMA matrix and spin-coated in thin, homogeneous
films. As part of the NanoVidere project, these preparations serve as a detection mechanism for
a high-resolution confocal microscope, which can resolve details below the diffraction limit.
First, the influence of the substitution pattern on the BTEs is investigated. A linear relationship
between the substituent and the resulting UV/Vis spectra was found. For this purpose, the
spectroscopic HAMMETT function is applied to the dyes. This means that the HAMMETT-BROWN constants can be used to predict the spectroscopic properties of BTEs for the first
time.
It also shows that the behavior of the BTE dyes in solution or in the crystalline phase cannot be
transferred to the PMMA environment. Substitutions of the basic structure of the BTEs, which
are considered to promote stability, show almost exactly the opposite behavior when they are
embedded in the film. This is therefore an environment in which the behavior of the BTEs must
be fundamentally re-understood. Through new approaches in synthesis, new knowledge could
be successively gained and applied, so that a comprehensive basis for the behavior of the dyes
in polymer matrices could be obtained. The influence of different substituents on the color
intensity, the color itself, as well as the response and stability behavior could be shown. This
made it possible to produce dyes that meet the required properties and are therefore suitable for
use in super-resolution imaging microscopy
Switchable mesomeric betaines derived from pyridinium-phenolates and bis(thienyl)ethane
Syntheses of pushâpull substituted non-symmetric bis(thienyl)ethenes (BTEs) possessing a central perfluorocyclopentene core are described. The substituent effects of anisole, phenole, and phenolate as well as pyridine, pyridinium, and N-methylpyridinium substituents, joined through their 3- or 4-positions to the central BTE core, respectively, cover the range from very strongly electron-donating [Ï(4-phenolate)=â1.00] to extremely strongly electron-withdrawing [Ï(pyridinium-4-yl)=+2.57] in the title mesomeric betaines. The different isomers possessing 4-yl/4-yl, 4-yl/3-yl and 3-yl/3-yl substituents represent different combinations of conjugated and cross-conjugated partial structures and cause different spectroscopic properties. In addition, through-space conjugation between the 2- and 2âČ-position of the thiophenes can be observed which circumvents the charge-separation of through-bond cross-conjugation. The BTE possessing the pushâpull chromophore consisting of 3-anisole and 4-pyridinium substituents (24) displays the best extinction coefficients within the series of compounds described here (Ï”=33.8/15.7â
