    The Shared and Distinct White Matter Networks Between Drug-Naive Patients With Obsessive-Compulsive Disorder and Schizophrenia

    Background: Obsessive-compulsive disorder (OCD) and schizophrenia (SZ) as two severe mental disorders share many clinical symptoms, and have a tight association on the psychopathological level. However, the neurobiological substrates between these two diseases remain unclear. To the best of our knowledge, no study has directly compared OCD with SZ from the perspective of white matter (WM) networks.Methods: Graph theory and network-based statistic methods were applied to diffusion MRI to investigate and compare the WM topological characteristics among 29 drug-naive OCDs, 29 drug-naive SZs, and 65 demographically-matched healthy controls (NC).Results: Compared to NCs, OCDs showed the alterations of nodal efficiency and strength in orbitofrontal (OFG) and middle frontal gyrus (MFG), while SZs exhibited widely-distributed abnormalities involving the OFG, MFG, fusiform gyrus, heschl gyrus, calcarine, lingual gyrus, putamen, and thalamus, and most of these regions also showed a significant difference from OCDs. Moreover, SZs had significantly fewer connections in striatum and visual/auditory cortices than OCDs. The right putamen consistently showed significant differences between both disorders on nodal characteristics and structural connectivity.Conclusions: SZ and OCD present different level of anatomical impairment and some distinct topological patterns, and the former has more serious and more widespread disruptions. The significant differences between both disorders are observed in many regions involving the frontal, temporal, occipital, and subcortical regions. Particularly, putamen may serve as a potential imaging marker to distinguish these two disorders and may be the key difference in their pathological changes

    Unlocking the mystery of the hard-to-sequence phage genome: PaP1 methylome and bacterial immunity

    BACKGROUND: Whole-genome sequencing is an important method to understand the genetic information, gene function, biological characteristics and survival mechanisms of organisms. Sequencing large genomes is very simple at present. However, we encountered a hard-to-sequence genome of Pseudomonas aeruginosa phage PaP1. Shotgun sequencing method failed to complete the sequence of this genome. RESULTS: After persevering for 10 years and going over three generations of sequencing techniques, we successfully completed the sequence of the PaP1 genome with a length of 91,715 bp. Single-molecule real-time sequencing results revealed that this genome contains 51 N-6-methyladenines and 152 N-4-methylcytosines. Three significant modified sequence motifs were predicted, but not all of the sites found in the genome were methylated in these motifs. Further investigations revealed a novel immune mechanism of bacteria, in which host bacteria can recognise and repel modified bases containing inserts in a large scale. This mechanism could be accounted for the failure of the shotgun method in PaP1 genome sequencing. This problem was resolved using the nfi(-) mutant of Escherichia coli DH5α as a host bacterium to construct a shotgun library. CONCLUSIONS: This work provided insights into the hard-to-sequence phage PaP1 genome and discovered a new mechanism of bacterial immunity. The methylome of phage PaP1 is responsible for the failure of shotgun sequencing and for bacterial immunity mediated by enzyme Endo V activity; this methylome also provides a valuable resource for future studies on PaP1 genome replication and modification, as well as on gene regulation and host interaction.     To solve the ecological disasters in Pubugou Reservoir Fluctuating Zone, ecological blanket governance model is proposed in this paper, which may provide good early environment for plants’ survival in fluctuation zone, and then play the function of greening and sustainable development to ensure the slopes’ stability. Meanwhile, based on the result of vetiver ecological blanket in Hanyuan experimental zone, we find that three kinds of typical Fluctuating Zone slope’s greening effect is good, which includes the dirt piling up slope, the whole lump of rock slope and the gravel piling up slope, and it gets an average coverage of 90.3 % as well as good strength. Due to the different geological conditions, the ecological blankets’ governance effect differs from slope to slope. Using analytic hierarchy process to calculate the weight, we get the dirt piling up slope, the whole lump of rock slope and the gravel piling up slope’s weights are 0.41, 0.17, 0.42, respectively, namely, the dirt piling up slope and the gravel piling up slope have good results overall, followed by the whole lump of rock slope

    Drought impacts on blue and green water: A spatial and temporal analysis

    Global warming is exacerbating water security risks in watershed ecosystems. Hence, it is especially crucial to examine how blue and green water respond to drought in watershed ecosystems. This study utilized the Soil and Water Assessment Tool (SWAT) to investigate temporal and spatial variations in blue and green water resources within the Wujiang River Basin (WRB) across three distinct ecosystems: agriculture, forest, and grassland. Concurrently, the Standardized Precipitation Evapotranspiration Index (SPEI) was employed to assess drought risk in the basin, and the relationship between typical ecosystem blue and green water resources and drought response was further explored. The study's results revealed the following key findings: (1) From 1985 to 2014, the basin recorded an average total of 54129.8 × 106 m3 and 46227.7 × 106 m3 of blue and green water, respectively. Notably, forest ecosystems contributed 50 % and 62 % to the blue and green water resources, respectively. (2) In typical ecosystems, temporal changes in both blue and green water exhibited a pattern of initial increase followed by a decrease. Furthermore, spatial changes along the upstream-to-downstream gradient showed an initial decrease, followed by an increase, and ultimately another decrease. (3) In typical ecosystems, blue water is more affected by dry and wet conditions, while green water is influenced by a combination of dry and wet conditions and land use changes. (4) As drought intensity rises, both blue and green water resources in typical ecosystems decrease. Notably, green water in forest ecosystems is less affected by drought intensity, which contributes to maintaining water resource security in watershed ecosystems. These study findings provide a robust scientific foundation for water resource management and drought mitigation strategies in the basin
