The JNK inhibitor, SP600125, potentiates the glial response and cell death induced by methamphetamine in the mouse striatum
This study investigates the effect of the selective Jun NH2-terminal kinase 1/2 (JNK1/2) inhibitor, (SP600125) on the striatal dopamine nerve terminal loss and on the increased interleukin-15 (IL-15) expression and glial response induced by methamphetamine (METH). Mice were given repeated low doses of METH (4Â mg/kg, i.p., three times separated by 3Â h) and killed 24Â h or 7 d after the last dose. SP600125 (30Â mg/kg, i.p) was administered 30Â min before the last METH injection. Results indicate that METH produced dopaminergic axonal neurotoxicity reflected as a marked decrease in the striatal density of tyrosine hydroxylase-immunoreactive (TH-ir) fibres and dopamine transporter-immunoreactivity (DAT-ir) 24Â h after dosing. These effects were not modified by SP600125. This compound also failed to prevent the long-term loss of dopamine levels and DAT observed 7 d following METH injection. Nevertheless, SP600125 potentiated METH-induced striatal cell loss reflected by an increase in Fluoro-Jade immunostaining, cleaved capase-3 immunoreactivity and the number of terminal deoxyncleotidyl transferase-mediated dUTP nick end labelling (TUNEL) positive cells. In line with a deleterious effect of JNK1/2 inhibition, SP600125 increased the astroglial and microglial response induced by METH and interfered with drug-induced IL-15 expression. Together these data indicate that, not only does SP600125 fail to protect against the dopaminergic damage induced by METH but also, in fact, it potentiates the glial response and the non-dopaminergic striatal cell loss caused by the drug. © 2013 CINP.Peer Reviewe
Isoreticular two-dimensional magnetic coordination polymers prepared through pre-synthetic ligand functionalization
Chemical functionalization is a powerful approach to tailor the physical and chemical properties of two-dimensional materials, increase their processability and stability, tune their functionalities and, even, create new 2D materials. This is typically achieved through post-synthetic functionalization by anchoring molecules on the surface of an exfoliated 2D crystal, but it inevitably alters the long-range structural order of the material. Here we present a pre-synthetic approach that allows the isolation of crystalline, robust, and magnetic functionalized monolayers of coordination polymers. A series of five isoreticular layered magnetic coordination polymers based on Fe(II) centres and different benzimidazole derivatives (bearing a Cl, H, CH3, Br or NH2 side group) were first prepared. On mechanical exfoliation, 2D materials are obtained that retain their long-range structural order and exhibit good mechanical and magnetic properties. This combination, together with the possibility to functionalize their surface at will, makes them good candidates to explore magnetism in the 2D limit and to fabricate mechanical resonators for selective gas sensing
Evaluation of appendicitis risk prediction models in adults with suspected appendicitis
Background
Appendicitis is the most common general surgical emergency worldwide, but its diagnosis remains challenging. The aim of this study was to determine whether existing risk prediction models can reliably identify patients presenting to hospital in the UK with acute right iliac fossa (RIF) pain who are at low risk of appendicitis.
Methods
A systematic search was completed to identify all existing appendicitis risk prediction models. Models were validated using UK data from an international prospective cohort study that captured consecutive patients aged 16–45 years presenting to hospital with acute RIF in March to June 2017. The main outcome was best achievable model specificity (proportion of patients who did not have appendicitis correctly classified as low risk) whilst maintaining a failure rate below 5 per cent (proportion of patients identified as low risk who actually had appendicitis).
Results
Some 5345 patients across 154 UK hospitals were identified, of which two‐thirds (3613 of 5345, 67·6 per cent) were women. Women were more than twice as likely to undergo surgery with removal of a histologically normal appendix (272 of 964, 28·2 per cent) than men (120 of 993, 12·1 per cent) (relative risk 2·33, 95 per cent c.i. 1·92 to 2·84; P < 0·001). Of 15 validated risk prediction models, the Adult Appendicitis Score performed best (cut‐off score 8 or less, specificity 63·1 per cent, failure rate 3·7 per cent). The Appendicitis Inflammatory Response Score performed best for men (cut‐off score 2 or less, specificity 24·7 per cent, failure rate 2·4 per cent).
Conclusion
Women in the UK had a disproportionate risk of admission without surgical intervention and had high rates of normal appendicectomy. Risk prediction models to support shared decision‐making by identifying adults in the UK at low risk of appendicitis were identified
Three catalytic sites in mitochondrial ATPase
AbstractKinetic data obtained after determining the hydrolytic activity of ATPase from rat liver in preparations where the enzyme had been purified, or in mitochondria, strongly suggest the existence of three different catalytic sites with different affinity for the substrate. The results obtained when measuring the ATPase activity at different substrate concentrations, and in the presence of the inhibitors KOCN or KSCN, or of the activators dinitrophenol and bicarbonate, show that the binding of these compounds to a regulatory site or sites affects in a different degree the hydrolytic activity of each catalytic site
Dinosaurs nesting on tidal flats
The discovery of a dinosaur clutch in Upper Cretaceous tidal flat deposits in the Tremp Formation in Biscarri,
Isona (Lleida, Spain), provides insight into the mode of life of some dinosaurs. The clutch occurs in grey marls that
were originally waterlogged, muddy sediments. The seven elongated eggs are parallel to each other and with their
long axis almost vertically oriented. A mound-nest is inferred in contrast to hole-nest described so far for similar eggshell types. The permeability of the eggshell (oospecies Megaloolithus siruguei) was highly enhanced by a braidednetwork of multicanaliculate pore canals. The nest style, eggshell porosity and tidal flat environment point to a periaquatic habitat for these dinosaurs. A modern analogue occurs in crocodiles, alligators and some birds living in periaquatic environments, with nesting on plant-mounds above waterlogged soils or even on floating vegetation
