    Effects of 3% trehalose as an adjuvant treatment after lasik

    Purpose: To evaluate the effect of 3% trehalose as an adjuvant in the standard treatment after laser-assisted in situ keratomileusis. Design: Interventional prospective comparative single-blind study. Setting: Department of Ophthalmology, Hospital Quirón Zaragoza, Spain. Methods: A total of 26 eyes (13 patients) were included, of which 12 eyes (group 1) received conventional treatment with lubricant drops of hyaluronic acid (0.15%) and 14 eyes (group 2) received, additionally, an ophthalmic solution of 3% trehalose. Pre- and postoperative quality-of-life tests and vital stains, tear breakup time, and osmolarity measurements were made. Results: We obtained statistically significant differences between the groups in the Symptom Assessment in Dry Eye test in all visits with respect to severity, and in the postoperative day 1 visit with respect to frequency, in all cases favoring the trehalose treatment. The values of osmolarity were not significantly different between groups. However, we did find significant differences in the Oxford scale in day 90 for the trehalose treatment (P<0.001), and in the National Eye Institute scale in day 30 (P=0.02). Conclusion: The results of this exploratory study indicate that the adjuvant treatment with 3% trehalose could be superior with respect to the standard treatment, with improvements in the objective and subjective parameters of tear quality

    Electroactive calcium-alginate/polycaprolactone/reduced graphene oxide nanohybrid hydrogels for skeletal muscle tissue engineering

    Graphene derivatives such as reduced graphene oxide (rGO) are used as components of novel biomaterials for their unique electrical properties. Electrical conductivity is a crucial factor for muscle cells, which are electrically active. This study reports the development of a new type of semi-interpenetrated polymer network based on two biodegradable FDA-approved biomaterials, sodium alginate (SA) and polycaprolactone (PCL), with Ca2+ ions as SA crosslinker. Several drawbacks such as the low cell adhesion of SA and weak structural stability can be improved with the incorporation of PCL. Furthermore, this study demonstrates how this semi-IPN can be engineered with rGO nanosheets (0.5% and 2% wt/wt rGO nanosheets) to produce electroactive nanohybrid composite biomaterials. The study focuses on the microstructure and the enhancement of physical and biological properties of these advanced materials, including water sorption, surface wettability, thermal behavior and thermal degradation, mechanical properties, electrical conductivity, cell adhesion and myogenic differentiation. The results suggest the formation of a complex nano-network with different interactions between the components: bonds between SA chains induced by Ca2+ ions (egg-box model), links between rGO nanosheets and SA chains as well as between rGO nanosheets themselves through Ca2+ ions, and strong hydrogen bonding between rGO nanosheets and SA chains. The incorporation of rGO significantly increases the electrical conductivity of the nanohybrid hydrogels, with values in the range of muscle tissue. In vitro cultures with C2C12 murine myoblasts revealed that the conductive nanohybrid hydrogels are not cytotoxic and can greatly enhance myoblast adhesion and myogenic differentiation.     Spatio-temporal trends of the bottom trawling activity in a mud volcano field of the north-eastern Gulf of Cádiz (south-western Iberian Peninsula)

    Multi-species bottom trawl fisheries are one of the human activities with a great impact on the benthic habitats and their associated biota. This study provides estimates of the bottom trawling activity (effort), catches and landings of the main commercial species as well as an estimation of the total revenue (TR) generated inside a mud volcano field located in the Spanish margin of the Gulf of Cádiz, during a time series from 2007 to 2012. To date, no studies have been carried out to analyse the temporal evolution of bottom trawling activity and TR in a mud volcano fied, or the economic consequences of possible potential bottom trawling regulation of certain sectors harbouring vulnerable and/or threatened habitats. In this study, Vessel Monitoring System data, logbooks and sales slips were used. The spatial distribution of the bottom trawling activity, catches and TR were related to the seafloor morphology and specific bottom types of the mud volcano field. During the time series, a high bottom trawling activity and associated catches was detected in flat sandy and muddy bottoms, including the Anastasya sector and between the Guadalquivir and Cádiz Diapiric Ridges. Low bottom trawling activity and catches were detected in the deepest areas but also in areas with hard and detritic bottoms such as Gazul and Chica sectors as well as in the Diapiric Ridges. A similar spatial pattern was detected for the TR asociated with these bottom trawling fisheries. An increase in bottom trawling activity was detected during the time series, mainly at the end, probably for increasing the TR and mantaining the economic profit due to the instability and increases in fuel prices and offset the increased costs. Based on the obtained information, bottom trawling regulations should be implemented in certain sectors harboring singular and/or threatened habitats and species. In some of these sectors, a low TR from bottom trawling was detected and, bottom trawling regulation may potentially have a low socioeconomic impact. This specific bottom trawling regulation could provide a sustainable balance between bottom trawling activities and habitat conservation in this mud volcano field according to the aims of the Habitats Directive (92/43/EEC) and the European Marine Strategy Framework Directive (2008/56/EEC).En prens

    The CaT strength in Seyfert nuclei revisited: analyzing young stars and non-stellar light contributions to the spectra

    In a former paper (Garcia-Rissmann et al. 2005; hereafter Paper I), we have presented spectra of 64 active, 9 normal and 5 Starburst galaxies in the region around the near-IR Calcium triplet absorption lines and the [SIII]9069 line. In the present paper we analyze the CaT strength (WCaT), and kinematical products derived in that study, namely stellar and ionized gas velocity dispersions. Our main results may be summarized as follows: (1) Seyfert 2s show no sign of dilution in WCaT with respect to the values spanned by normal galaxies, even when optical absorption lines such as the CaII K band at 3933 A are much weaker than in old, bulge-like stellar populations. (2) The location of Seyfert 2s in the WCaT-WCaK plane is consistent with evolutionary synthesis models. The implication is that the source responsible for the dilution of optical lines in these AGN is a young stellar population, rather than an AGN featureless continuum, confirming the conclusion of the pioneer study of Terlevich, Diaz & Terlevich. (3) In Seyfert 1s, both W[SIII] and WCaT tend to be diluted due to the presence of a non-stellar component, in agreement with the unification paradigm. (4) A comparison of stellar and gas velocity dispersions confirms the existence of a correlation between the typical velocities of stars and clouds of the Narrow Line Region. The strength and scatter around this correlation are similar to those previously obtained from the [OIII]5007 line width.Comment: 14 pages, 15 figures. Paper accepted for publication in MNRA

    Clinical factors associated with discontinuation of ts/bDMARDs in rheumatic patients from the BIOBADASER III registry

Persistence of therapy provides an index of a drug's overall effectiveness. The objective of the study was to identify factors associated with discontinuation of ts/bDMARDs in a real-world dataset. The study population comprised patients diagnosed with RA, PsA, and AS included in the BIOBADASER registry for whom follow-up data were available until November 2019. Patient features and treatment data were included in the analysis. The Kaplan-Meier method was used to study survival of the different drugs according to the reason for discontinuation. Factors associated with discontinuation were studied using Cox regression models and bivariate and multivariate analyses. P values of less than 0.05 were regarded as statistically significant. The study population comprised 4,752 patients who received a total of 8,377 drugs, of which 4,411 (52.65%) were discontinued. The Kaplan-Meier curves showed that survival for first-line treatment was greater in all 3 groups (p < 0.001). Patients with RA had a greater risk of discontinuation if they were younger (HR, 0.99; 95% CI 0.99-1.00), if they were receiving anti-TNFα agents (HR, 0.61; 95% CI 0.54-0.70), and if they had more comorbid conditions (HR, 1.09; 95% CI 1.00-1.17). Patients with PsA had a higher risk if they were women (HR, 1.36; 95% CI 1.15-1.62) and if they were receiving other ts/bDMARDs (HR, 1.29; 95% CI 1.05-1.59). In patients with AS, risk increased with age (HR, 1.01; 95% CI 1.00-1.02), as did the number of comorbid conditions (HR, 1.27; 95% CI 1.12-1.45). The factors that most affected discontinuation of ts/bDMARDs were line of treatment, age, type of drug, sex, comorbidity and the year of initiation of treatment. The association with these factors differed with each disease, except for first-line treatment, which was associated with a lower risk of discontinuation in all 3 diseases

    ACS imaging of star clusters in M51. I. Identification and radius distribution

    We use HST/ACS observations of the spiral galaxy M51 in F435W, F555W and F814W to select a large sample of star clusters with accurate effective radius measurements in an area covering the complete disc of M51. We present the dataset and study the radius distribution and relations between radius, colour, arm/interarm region, galactocentric distance, mass and age. We select a sample of 7698 (F435W), 6846 (F555W) and 5024 (F814W) slightly resolved clusters and derive their effective radii by fitting the spatial profiles with analytical models convolved with the point spread function. The radii of 1284 clusters are studied in detail. We find cluster radii between 0.5 and ~10 pc, and one exceptionally large cluster candidate with a radius of 21.6 pc. The median radius is 2.1 pc. We find 70 clusters in our sample which have colours consistent with being old GC candidates and we find 6 new "faint fuzzy" clusters in, or projected onto, the disc of M51. The radius distribution can not be fitted with a power law, but a log-normal distribution provides a reasonable fit to the data. This indicates that shortly after the formation of the clusters from a fractal gas, their radii have changed in a non-uniform way. We find an increase in radius with colour as well as a higher fraction of redder clusters in the interarm regions, suggesting that clusters in spiral arms are more compact. We find a correlation between radius and galactocentric distance which is considerably weaker than the observed correlation for old Milky Way GCs. We find weak relations between cluster luminosity and radius, but we do not observe a correlation between cluster mass and radius.Comment: 17 pages, 23 figures. Accepted for publication in A&

    Elucidation of the ebola virus VP24 cellular interactome and disruption of virus biology through targeted inhibition of host-cell protein function

    Viral pathogenesis in the infected cell is a balance between antiviral responses and subversion of host-cell processes. Many viral proteins specifically interact with host-cell proteins to promote virus biology. Understanding these interactions can lead to knowledge gains about infection and provide potential targets for antiviral therapy. One such virus is Ebola, which has profound consequences for human health and causes viral hemorrhagic fever where case fatality rates can approach 90%. The Ebola virus VP24 protein plays a critical role in the evasion of the host immune response and is likely to interact with multiple cellular proteins. To map these interactions and better understand the potential functions of VP24, label-free quantitative proteomics was used to identify cellular proteins that had a high probability of forming the VP24 cellular interactome. Several known interactions were confirmed, thus placing confidence in the technique, but new interactions were also discovered including one with ATP1A1, which is involved in osmoregulation and cell signaling. Disrupting the activity of ATP1A1 in Ebola-virus-infected cells with a small molecule inhibitor resulted in a decrease in progeny virus, thus illustrating how quantitative proteomics can be used to identify potential therapeutic targets

    Objectives: To determine if patients with the predominant extracranial large-vessel-vasculitis (LVV) pattern of giant cell arteritis (GCA) have a distinctive HLA-B association, different from that reported in biopsy-proven cranial GCA patients. In a further step we assessed if the combination of HLA-B and HLA-DRB1 alleles confers an increased risk for GCA susceptibility, either for the cranial and extracranial LVV phenotypes. Methods: A total of 184 patients with biopsy-proven cranial GCA, 105 with LVV-GCA and 486 healthy controls were included in our study. We compared HLA-B phenotype frequencies between the three groups. Results: HLA-B*15 phenotype was significantly increased in patients with classic cranial GCA compared to controls (14.7% versus 5.8%, respectively; p<0.01; OR [95% CI] =2.81 [1.54-5.11]). It was mainly due to the HLA-B*15:01 allele (12.5% versus 4.0%, respectively; p<0.01; OR [95% CI] =3.51 [1.77-6.99]) and remained statistically significant after Bonferroni correction. Similar HLA-B*15 association was observed in patients with the LVV-GCA (11.4% versus 5.8%, p=0.04, OR [95% CI] =2.11 [1.04-4.30]). This association was also mainly due to the HLA-B*15:01 allele (10.5% versus 4.0%, respectively; p=0.0054; OR [95% CI] =2.88 [1.19-6.59]). Noteworthy, the presence of HLA-B*15:01 together with HLA-DRB1*04:01 led to an increased risk of developing both cranial and extracranial LVV-GCA.     Deep-sea habitat characterization using acoustic data and underwater imagery in Gazul mud volcano (Gulf of Cádiz, NE Atlantic)

    Gazul is the shallowest mud volcano (MV) within the Shallow Field of Fluid Expulsion (SFFE) of the northeastern Gulf of Cádiz (NE Atlantic; 300–1200 m depth). The SFFE represents an important geo- and biodiversity area that was designated as a Site of Community Importance under the European Habitats Directive in 2014. In this study, geological features, habitats and associated biodiversity, as well as anthropogenic impacts, were characterized at Gazul MV from underwater imagery and multibeam bathymetry. Multivariate methods using the Bray-Curtis similarity index identified six main habitats, each of which harbored a characteristic faunal assemblage that included: (1) sandy ripple bottoms typified by the actiniarian Actinauge richardi; (2) sandy, muddy, coarse sand and bioclastic bottoms dominated by the solitary coral Flabellum chunii; (3) coarse sand and bioclastic bottoms, together with soft sediments covered by scattered methane-derived authigenic carbonates (MDACs) (mixed bottoms), characterized by the echinoid Cidaris cidaris; (4) hard bottoms comprising MDACs dominated by a wide variety of sponges and gorgonians; (5) coral-rubble bottoms typified by the presence of colonial scleractinian communities dominated by Madrepora oculata; and (6) mixed bottoms characterized by the presence of a styelid ascidian. Slope and water depth were the main factors explaining assemblages’ distribution, which was also supported by the presence of MDACs such as slabs, crusts and chimneys on the seafloor, as well as by the geomorphologic diversity of Gazul MV. The results highlight Gazul MV as an eco-biologically important area harboring different vulnerable marine ecosystem (VME) elements with indicator taxa such as scleractinians, sponges, gorgonians and black corals. ROV images revealed abandoned or lost fishing gears and marine debris on the seafloor, indicating anthropogenic impacts in Gazul MV and adjacent areas. Indeed trawling fisheries activities have also been detected in Vessel Monitoring System datasets. A fishery restricted area is recommended in Gazul MV due to the occurrence of diverse VMEs and species included in different conservation directives and conventions.Postprin
