Characterization of a Murine Anti-laminin-1 Monoclonal Antibody (AK8) Produced by Immunization with Mouse-derived Laminin-1
Laminin-1 is a structural glycoprotein that forms an integral part of
the scaffolding of basement membranes, and plays an important role during
embryonic development. We have recently demonstrated a significant association
between anti-laminin-1 antibodies (Abs) and reproductive failure, such as recurrent
spontaneous abortions and infertility-associated endometriosis in both human and
mouse studies. In the present study, we established an IgM (μ,κ) monoclonal
anti-laminin-1 Ab (AK8) by immunizing mice with mouse Engelbreth-Holm-Swarm
sarcoma (EHS)-derived laminin-α1. The AK8 monoclonal antibody (mAb) reacted with
particular peptide sequences from the globular G domain of mouse laminin-α1 chain
of using ELISA and Western blot techniques. The peptide tertiary structure of
the epitope recognized by AK8 mAb was predicted using eight synthesized domain
peptide sequences and three consensus sequences obtained by phage displayed
random peptide library. Basement membranes of endometrium of pregnant mice
and humans were immunostained with AK8 mAb. Thus, AK8 mAb recognized
a common structure present in the G domain of the laminin-1 chain in both
mice and humans. The passive immunization of mice with AK8 mAb may represent
a suitable animal model for anti-laminin-1 Ab-mediated reproductive failure
Development of Disaster Education Digital Material for Elementary Schoolchild: Fostering an Attitude of Subjective Behavior toward Earthquakes
This study developed the disaster education digital material for elementary schoolchildren, and examined the teaching effectiveness of this material. Eighty-five schoolchildren attended the class using this material twice, and did homework with their parents. Before and after the class, they answered the questionnaire to measure their learning effect (attitude toward subjective behavior, motivation toward disaster prevention action in cooperation with parents: both 3 items) and their evaluation of the class (3 items). The results generally indicated that their attitude of subjective behavior, and their motivation toward disaster prevention action was higher after the education, and their evaluation of class were high. These results generally showed the effectiveness of this material. In future studies, generalizability of these results should be examined.研究ノー
Differences in verbal instructions on the experience and practice of Dohsa-Hou
The purpose of this study was to discuss the influence of the deference on the task instructions to the feeling of Dohsa-experience and Dohsa-performance in application of Dohsa-hou. One instruction is “Please raise your arm and down your arm. (Movement instruction)” and another instruction is “Please raise your arm and down your arm with relaxation of your body. (Relaxation instruction)”. During the movement, the experimenter saw how long it takes for that. After that, subjects checked the scale of feeling of Dohsa-experience. As a result, in the movement instruction condition, subjects feel “change of their body”, “refresh”, “active”, “attention to the body”, “self attitude of active trial and error”, “self attitude of concentration” more than another. In the movement instruction, subjects feel “vague” and “confusion” more than another. In addition, it takes more long time for relaxation condition than another. This result means that clear instruction of relaxation made subjects experience actual feelings. In addition, because subjects were not used to relaxation activity, they experience new feelings. So, therapists should consider the features of instruction in the therapy of application of Dohsa-hou
