    Middle to late Holocene environmental evolution of the Pisa coastal plain (Tuscany, Italy) and early human settlements

    A cross-disciplinary (sedimentological, geochemical, micropalaeontological and archaeological) examination of 12 continuous cores, up to 20m long, integrated with stratigraphical, geomorphological and historical investigations, allows for reliable delineation of the middle-late Holocene environmental evolution in the Pisa old town area, with special emphasis on the Etruscan age transition. Depositional facies were identified through integration of sedimentological and micropalaeontological (benthic foraminifers, ostracods, phytoclasts and palynomorphs) data, while sediment dispersal patterns were reconstructed on the basis of geochemical analyses. Facies architecture was chronologically constrained by combined archaeological and radiocarbon dating. The turnaround from early Holocene, transgressive conditions to the ensuing (middle-late Holocene) phase of sea-level highstand is witnessed by a prominent shallowing-upward succession of lagoonal, paludal and then poorly drained floodplain deposits supplied by two river systems (Arno and Serchio). This 'regressive' trend, reflecting coastal progradation under nearly stable sea-level conditions, was interrupted by widespread swamp development close to the Iron-Etruscan age transition. The expansion of vast, low-lying paludal areas across the alluvial plain was mostly induced by the intricate, short-term evolution of the meandering Arno and Serchio river systems. These changes in the fluvial network, which occurred during a period of variable climate conditions, strongly influenced the early Etruscan culture (7th-5th century BC) in terms of human settlement and society behaviour. Conversely, a strong impact of human frequentation on depositional environments is observed at the transition to the Roman age (from the 1st century BC onwards), when the wetlands were drained and the modern alluvial plain started to form. The palaeoenvironmental reconstruction fits in with the original geographical descriptions mentioned in Strabo's Chronicles, and provides chronologically constrained data of fluvial evolution from the Pisa old town area. © 2013 Elsevier Ltd and INQUA

    Sand supply from shoreface to foredunes: aeolian transport measurements and morphological evolution of a Tuscany beach stretch (Italy)

    The coastal dunes are a highly dynamic sedimentary environment characterized by a continuous time-space readjustment in terms of morphology, shape and dimension. This is mainly due to the periodic fluctuation of the volume of sand available and by the force of the deflation processes, which are in turn driven by the interplay among pattern of vegetation cover, surface roughness and local-regional wind regime. The aim of our research is to quantify the deflation, transport and deposition of sands in a natural coastal field dune system located in the northern coast of Tuscany, Italy. The northern part of the investigated area is characterized by stable coastline condition while southwards strong erosive processes took place since 1800 year. Sedimentological data come from a series of sand collectors spaced along transects orthogonal to the coastline from the backshore to the semi stable dune field. The collectors were constructed of PVC pipe 100 x height 10 cm, with two openings 7 cm wide and 50 cm tall arranged on opposite sides of the tube. Opening willing to windward served for sand collection, and to leeward, covered with a metal wire 60ÎŒm opening. Collectors were buried along, until the base of the free window coincide with the surface of the ground about 1,5 m. The sand trapped within each collector was sampled every two hours for three consecutive times. In laboratory sand samples were weighed and subject to grain size analysis by means of mechanical sieves. The local winds parameters and their fluctuation with the time were acquired through a Meteorological mobile station. The station is equipped with three ammeters located to three different heights from ground surface: 40, 120 and 180 cm. A wireless sensor allows the constant output of data (each 5 sec) to a device. Temperature, and relative humidity value are furnished every 30 minutes. Analysis of data has evidenced the time-space fluctuation of sand volume in the two study area (stable area and under erosion).     We addressed the analysis of the physical and functional association of proliferating cell nuclear antigen (PCNA), a protein involved in many DNA transactions, with poly(ADP-ribose) polymerase (PARP-1), an enzyme that plays a crucial role in DNA repair and interacts with many DNA replication/repair factors. We demonstrated that PARP-1 and PCNA co-immunoprecipitated both from the soluble and the DNA-bound fraction isolated from S-phase-synchronized HeLa cells. Immunoprecipitation experiments with purified proteins further confirmed a physical association between PARP-1 and PCNA. To investigate the effect of this association on PARP-1 activity, an assay based on the incorporation of radioactive NAD was performed. Conversely, the effect of PARP-1 on PCNA-dependent DNA synthesis was assessed by a DNA polymerase delta assay. A marked inhibition of both reactions was found. Unexpectedly, PARP-1 activity also decreased in the presence of p21waf1/cip1. By pull-down experiments, we provided the first evidence for an association between PARP-1 and p21, which involves the C-terminal part of p21 protein. This association was further demonstrated to occur also in vivo in MNNG (N-methyl-N'-nitro-N-nitrosoguanidine)-treated human fibroblasts. These observations suggest that PARP-1 and p21 could cooperate in regulating the functions of PCNA during DNA replication/repair

    MAPPA cores: an interdisciplinary approach

    In the urban and periurban area of Pisa, 18 cores ranging in depth between 7 and 15 m were performed and analysed through an interdisciplinary approach, which led to an improved stratigraphic log representation. The acquired data also improved our knowledge about the palaeoenvironmental and human settlement evolution of Pisa area over the past 6000 years, evidencing the mutual interaction existing between landscape and human activities

    Eleven-year experience with the avidin-biotin pretargeting system in glioblastoma: Toxicity, efficacy and survival

    Background: The 3-step avidin-biotin pretargeting approach is applied in patients with recurrent glioblastoma (GBM), using biotinylated anti-tenascin monoclonal antibody as the first step of pretargeting followed by avidin and 90Ybiotin. Methods: The present study reviews objective response and overall survival rates in 502 glioblastoma patients treated with 3-step radioimmunotherapy in our institute from December 1994 to December 2005. Patients underwent standard treatment before receiving Pretargeted Antibody-Guided Radionuclide Therapy with 90Y-biotin (PAGRIT ¼). Results: Of the 502 patients, 272 (54%) were evaluable for response and 375 (75%) for overall survival. 174 patients (64%) continued to progress after PAGRIT ¼, 77 (28%) obtained disease stabilization, and 21 (8%) showed objective tumor regression. Survival of the 375 evaluable patients was 98.4% at 6 months, 79.2% at 12 months, 51.7% at 18 months, and 30.7% at 24 months after the first cycle of PAGRIT ¼. All 375 received 3-step PAGRIT ¼ at recurrence of GBM. The median survival time from diagnosis was 19 months. Conclusion: The results from this retrospective analysis suggest that 90Y-biotin PAGRIT ¼ interferes with the progression of glioblastoma, prolonging survival in a larger number of patients. Our analysis forms the basis for further prospective trials, where radioimmunotherapy, which is known to be more effective in minimal residual disease, could be offered immediately after surgery. © Grana et al.; Licensee Bentham Open

    Residential proximity to petrol stations and risk of childhood leukemia

    Petrol stations emit benzene and other contaminants that have been associated with an increased risk of childhood leukemia. We carried out a population-based case-control study in two provinces in Northern Italy. We enrolled 182 cases of childhood leukemia diagnosed during 1998-2019 and 726 age- and sex-matched population controls. We geocoded the addresses of child residences and 790 petrol stations located in the study area. We estimated leukemia risk according to distance from petrol stations within a 1000 m buffer and amount of supplied fuel within a buffer of 250 m from the child's residence. We used conditional logistic regression models to approximate risk ratios (RRs) and 95% confidence intervals (CIs) for associations of interest, adjusted for potential confounders. We also modeled non-linear associations using restricted cubic splines. In secondary analyses, we restricted to acute lymphoblastic leukemia (ALL) cases and stratifed by age (<5 and >= 5 years). Compared with children who lived >= 1000 m from a petrol station, the RR was 2.2 (95% CI 0.5-9.4) for children living<50 m from nearest petrol station. Associations were stronger for the ALL subtype (RR=2.9, 95% CI 0.6-13.4) and among older children (age >= 5 years: RR=4.4, 95% CI 0.6-34.1; age<5 years: RR=1.6, 95% CI 0.1-19.4). Risk of leukemia was also greater (RR=1.6, 95% CI 0.7-3.3) among the most exposed participants when assigning exposure categories based on petrol stations located within 250 m of the child's residence and total amount of gasoline delivered by the stations. Overall, residence within close proximity to a petrol station, especially one with more intense refueling activity, was associated with an increased risk of childhood leukemia, though associations were imprecise

    Physicochemical Characterization of Pectin-Gelatin Biomaterial Formulations for 3D Bioprinting.

    AbstractDeveloping biomaterial formulations with specific biochemical characteristics and physical properties suitable for bioprinting of 3D scaffolds is a pivotal challenge in tissue engineering. Therefore, the design of novel bioprintable formulations is a continuously evolving research field. In this work, the authors aim at expanding the library of biomaterial inks by blending two natural biopolymers: pectin and gelatin. Cytocompatible formulations are obtained by combining pectin and gelatin at different ratios and using (3‐glycidyloxypropyl)trimethoxysilane (GPTMS) as single crosslinking agent. It is shown that the developed formulations are all suitable for extrusion‐based 3D bioprinting. Self‐supporting scaffolds with a designed macroporosity and micropores in the bioprinted struts are successfully obtained by combining extrusion‐based bioprinting and freeze‐drying. The presence of gelatin in these formulations allows for the modulation of porosity, of water uptake and of scaffold stiffness in respect to pure pectin scaffolds. Results demonstrate that these new biomaterial formulations, processed with this specific approach, are promising candidates for the fabrication of tissue‐like scaffolds for tissue regeneration

    New Psychoactive Substances and Suicidality: A Systematic Review of the Current Literature

Epub 6 June 2021.Background and Objectives: Over the past twenty years a large number of New Psychoactive Substances (NPS) has entered and modified the recreational drug scene. Their intake has been associated with health-related risks, and especially so for vulnerable populations such as people with severe mental illness, who might be at higher risk of suicidality or self- injurious behaviour. This paper aims at providing an overview of NPS abuse and effects on mental health and suicidality issues, performing a literature review of the current related knowledge, thereby identifying substances that more than others are linked to suicidal behaviours. Materials and Methods: A comprehensive and updated overview of the literature regarding suicidality and NPS categories has been provided. An electronic search was performed, including all papers published up to March 2021, using the following keywords "NPS" OR "new psychoactive substances" OR "novel psychoactive substances" OR "synthetic cannabinoids" OR "phenethylamines" OR "synthetic cathinones" OR “tryptamines” OR “piperazines” OR “new synthetic opioids” OR “designer benzodiazepines”) AND ("suicide" OR "suicidality") NOT review NOT animal on the PubMed, Cochrane Library, and Web of Science online databases. Results: Suicidality and self-injurious behaviour appear to be frequently associated with some NPS such as cathinones, synthetic cannabinoids, and new synthetic opioids. Results are organised according to substances recorded. Conclusion: The growing use of NPS has become a significant clinical issue, posing increasing concern and challenges for clinicians working in both mental health and emergency departments. Thus, considering the associations between NPS and suicidality or self-injurious behaviours, areas where suicide-prevention efforts and strategies might be focused are the early detection, monitoring and restriction of NPS.Peer reviewe

    Impacts of biological globalization in the Mediterranean: Unveiling the deep history of human-mediated gamebird dispersal

    Humans have a long history of moving wildlife that over time has resulted in unprecedented biotic homogenization. It is, as a result, often unclear whether certain taxa are native to a region or naturalized, and how the history of human involvement in species dispersal has shaped present-day biodiversity. Although currently an eastern Palaearctic galliform, the black francolin (Francolinus francolinus) was known to occur in the western Mediterranean from at least the time of Pliny the Elder, if not earlier. During Medieval times and the Renaissance, the black francolin was a courtly gamebird prized not only for its flavor, but also its curative, and even aphrodisiac qualities. There is uncertainty, however, whether this important gamebird was native or introduced to the region and, if the latter, what the source of introduction into the western Mediterranean was. Here we combine historical documentation with a DNA investigation of modern birds and archival (13th–20th century) specimens from across the species’ current and historically documented range. Our study proves the black francolin was nonnative to the western Mediterranean, and we document its introduction from the east via several trade routes, some reaching as far as South Asia. This finding provides insight into the reach and scope of long-distance trade routes that serviced the demand of European aristocracy for exotic species as symbols of wealth and prestige, and helps to demonstrate the lasting impact of human-mediated long-distance species dispersal on current day biodiversity
