    Hypertemporal Imaging of NYC Grid Dynamics

    Hypertemporal visible imaging of an urban lightscape can reveal the phase of the electrical grid granular to individual housing units. In contrast to in-situ monitoring or metering, this method offers broad, persistent, real-time, and non-permissive coverage through a single camera sited at an urban vantage point. Rapid changes in the phase of individual housing units signal changes in load (e.g., appliances turning on and off), while slower building- or neighborhood-level changes can indicate the health of distribution transformers. We demonstrate the concept by observing the 120 Hz flicker of lights across a NYC skyline. A liquid crystal shutter driven at 119.75 Hz down-converts the flicker to 0.25 Hz, which is imaged at a 4 Hz cadence by an inexpensive CCD camera; the grid phase of each source is determined by analysis of its sinusoidal light curve over an imaging "burst" of some 25 seconds. Analysis of bursts taken at ~15 minute cadence over several hours demonstrates both the stability and variation of phases of halogen, incandescent, and some fluorescent lights. Correlation of such results with ground-truth data will validate a method that could be applied to better monitor electricity consumption and distribution in both developed and developing cities.Comment: This paper uses astronomical techniques applied to the study of urban lights.     Effect of soil on microbial diversity of flora and fauna in ecosystem

    Soil is a complex and dynamic biological system. We are limited in the determination of microbial mediated reactions because present assays for determining the overall rate of entire metabolic processes (such as respiration) or specific enzyme activities (such as urease, protease and phosphomonoesterase activity) do not allow any identification of the microbial species directly involved in the measured processes. The central problem posed by the link between microbial diversity and soil function is to understand the relations between genetic diversity and community structure and between community structure and function. Soil seems to be characterized by a redundancy of functions; for example, no relationship has been shown to exist between microbial diversity and decomposition of organic matter. Generally, a reduction in any group of species has little effect on overall processes in soil because other microorganisms can take on its function

    An energy efficient modified passive power filter for power quality enhancement in electric drives

    In most industrial applications, AC drives are used. These drives require power electronic modules to convert AC to DC and also DC to AC. The power modules used for power conversion consist of power semiconductor switches. There is distortion in the voltage and current obtained from the power modules due to non-linear behaviors of semiconductor switches. To reduce the distortion in the input current, inductors are used along with the line impedance. A high value of inductance is required to maintain the percentage of THD within limits set by the latest standards. Along with the increased size and cost, it also reduces the power factor and output DC voltage at higher loads. The use of a passive power filter (PPF) is the conventional method followed for the reduction of percentage THD and promotion of the power factor. PPFs comprise passive elements such as the resistor, inductor, and capacitor. These passive elements are connected at the point of common coupling (PCC) in shunt to compensate for the harmonics present in the input current. In this study, a modified multi-tuned passive filter is considered to reduce the source current harmonics. A bridge rectifier with resistive load, three-phase induction motor drive, and linear resistive–inductive load is connected at the point of common coupling to analyze the harmonics present in the source current, and also unbalanced created in one phase. To achieve proper selection of the resistance value for the passive filter, the class topper optimization technique is used. To validate the simulation results obtained for the multi-tuned passive filter, the hardware is implemented with a three-phase AC induction motor drive load, in which the speed of the motor is controlled with voltage by a frequency control algorithm using an FPGA controller; The 50% THD is reduced by using the fifth-order filter alone, 75% by combining the fifth- and seventh-order filters, 85% by combining the 5th-, 7th-, 11th-, 13th-, and higher-order filters, and 90% for varying loads. For single-phase AC induction motor load, THD % is reduced to 4%, and for three-phase AC induction motor drive, THD % is reduced to 10% with the same value of the filter

    Mycobacterium tuberculosis Transcriptional Adaptation, Growth Arrest and Dormancy Phenotype Development Is Triggered by Vitamin C

    BACKGROUND: Tubercle bacilli are thought to persist in a dormant state during latent tuberculosis (TB) infection. Although little is known about the host factors that induce and maintain Mycobacterium tuberculosis (M. tb) within latent lesions, O(2) depletion, nutrient limitation and acidification are some of the stresses implicated in bacterial dormancy development/growth arrest. Adaptation to hypoxia and exposure to NO/CO is implemented through the DevRS/DosT two-component system which induces the dormancy regulon. METHODOLOGY/PRINCIPAL FINDINGS: Here we show that vitamin C (ascorbic acid/AA) can serve as an additional signal to induce the DevR regulon. Physiological levels of AA scavenge O(2) and rapidly induce the DevR regulon at an estimated O(2) saturation of <30%. The kinetics and magnitude of the response suggests an initial involvement of DosT and a sustained DevS-mediated response during bacterial adaptation to increasing hypoxia. In addition to inducing DevR regulon mechanisms, vitamin C induces the expression of selected genes previously shown to be responsive to low pH and oxidative stress, triggers bacterial growth arrest and promotes dormancy phenotype development in M. tb grown in axenic culture and intracellularly in THP-1 cells. CONCLUSIONS/SIGNIFICANCE: Vitamin C mimics multiple intracellular stresses and has wide-ranging regulatory effects on gene expression and physiology of M. tb which leads to growth arrest and a 'dormant' drug-tolerant phenotype, but in a manner independent of the DevRS/DosT system. The 'AA-dormancy infection model' offers a potential alternative to other models of non-replicating persistence of M. tb and may be useful for investigating host-'dormant' M. tb interactions. Our findings offer a new perspective on the role of nutritional factors in TB and suggest a possible role for vitamin C in TB

    Chemoenzymatic synthesis, nanotization and anti- aspergillus activity of optically enriched fluconazole analogues

    Despite recent advances in diagnostic and therapeutic advances in antifungal research, aspergillosis still remains a leading cause of morbidity and mortality. One strategy to address this problem is to enhance the activity spectrum of known antifungals, and we now report the first successful application of Candida antarctica lipase (CAL) for the preparation of optically enriched fluconazole analogs. Anti-Aspergillus activity was observed for an optically enriched derivative, (-)-S-2-(2’ ,4’ -difluorophenyl)-1-hexyl-amino-3-(1‴,2‴,4‴) triazol-1‴-yl-propan-2-ol, which exhibits MIC values of 15.6 μg/mL and 7.8 μg/disc in microbroth dilution and disc diffusion assays, respectively. This compound is tolerated by mammalian erythrocytes and cell lines (A549 and U87) at concentrations of up to 1000 μg/mL. When incorporated into dextran nanoparticles, the novel, optically enriched fluconazole analog exhibited improved antifungal activity against Aspergillus fumigatus (MIC = 1.63 μg/mL). These results not only demonstrate the ability of biocatalytic approaches to yield novel, optically enriched fluconazole derivatives but also suggest that enantiomerically pure fluconazole derivatives, and their nanotised counterparts, exhibiting anti-Aspergillus activity may have reduced toxicity
