    Health monitoring of rolling element bearing using a spectrum searching strategy

    Aiming at achieving early fault diagnosis and tracking the degradation process of bearings, we propose a novel monitoring methodology using a spectrum searching strategy in this paper. Firstly, a vibration signal is collected with appropriate sampling frequency and length. Secondly, the structural information of spectrum (SIOS) on a predefined frequency grid is constructed through a searching algorithm after deriving the single-sided FFT spectrum. Finally, the two-dimensional (2-D) line plot of the frequency grid versus the average power in SIOS is employed to conduct fault detection and the sum of the largest six total-power (SLSTP) of the frequency grid in SIOS is calculated as a health indication to demonstrate the changes in the bearing’s health status. The performance of the proposed scheme is validated with both simulation and bearing data. Experimental results show that the monitoring algorithm could manifest satisfactory behaviors in early fault diagnosis and health assessment of bearings

    The phenolics, antioxidant activity and in vitro digestion of pomegranate (Punica granatum L.) peels: an investigation of steam explosion pre-treatment

    Pomegranate peels, the main byproduct of pomegranate production, are rich in phenolic compounds that are known for their effective antioxidant properties and have vast application prospects. In this study, steam explosion, an environmentally friendly technique, was applied to pretreat pomegranate peels for phenol extraction. We investigated the effects of explosion pressure, duration, and particle size on the content of total and individual phenolics, and antioxidant activity of pomegranate peels before and after in vitro digestion. The optimal conditions for a steam explosion for pomegranate peels in terms of total phenol content were a pressure of 1.5 MPa, a maintenance time of 90 s, and a particle size of 40 mesh. Under these conditions, pomegranate peel extract presented a higher yield of total phenols, gallic acid, and ellagic acid. However, it also had a lower content of punicalin and punicalagin, compared to the unexploded peels. There was no improvement in the antioxidant activity of pomegranate peels after the steam explosion. Moreover, the content of total phenol, gallic acid, ellagic acid, punicalin, and punicalagin, as well as the antioxidant activity of pomegranate peels, all increased after gastric digestion. Nevertheless, there was a large variation in the pomegranate peel processed by different pressure, duration, and sieve fractions. Overall, this study demonstrated that steam explosion pre-treatment could be an efficient method for improving the release of phenolics, especially gallic acid, and ellagic acid, from pomegranate peels

    Facile One-Pot Synthesis of Self-Assembled Folate-Biotin-Pullulan Nanoparticles for Targeted Intracellular Anticancer Drug Delivery

    The self-assembled folate-biotin-pullulan (FBP) nanoparticles (NPs) were prepared by facile one-pot synthesis and their physicochemical properties were characterized. The self-assembled FBP NPs were used as an anticancer drug nanocarrier entrapping doxorubicin (DOX) for targeting folate-receptors-overexpressing cancer cells. The identification of prepared NPs to folate-receptor-expressing cancer cells (KB cells) was affirmed by cell viability measurement, folate competition test, and flow cytometric analysis. Compared with the naked DOX and DOX/BP NPs, the DOX/FBP NPs had lower IC50 value compared to KB cells as a result of the folate-receptor-mediated endocytosis process. The cytotoxicity of DOX/FBP NPs to KB cells could be inhibited competitively by free folate. The cellular intake pattern of naked DOX and drug-loaded NPs was identified by confocal laser scanning microscopy (CLSM) observation and the higher cellular uptake of drug for DOX/FBP NPs over naked DOX was observed. The prepared FBP NPs had the potential to be used as a powerful carrier to target anticancer drugs to folate-receptor-expressing tumor cells and reduce cytotoxicity to normal tissues

    Spin-phonon scattering-induced low thermal conductivity in a van der Waals layered ferromagnet Cr2_2Si2_2Te6_6

    Layered van der Waals (vdW) magnets are prominent playgrounds for developing magnetoelectric, magneto-optic and spintronic devices. In spintronics, particularly in spincaloritronic applications, low thermal conductivity (κ\kappa) is highly desired. Here, by combining thermal transport measurements with density functional theory calculations, we demonstrate low κ\kappa down to 1 W m−1^{-1} K−1^{-1} in a typical vdW ferromagnet Cr2_2Si2_2Te6_6. In the paramagnetic state, development of magnetic fluctuations way above Tc=T_\mathrm{c}= 33 K strongly reduces κ\kappa via spin-phonon scattering, leading to low κ∼\kappa \sim 1 W m−1^{-1} K−1^{-1} over a wide temperature range, in comparable to that of amorphous silica. In the magnetically ordered state, emergence of resonant magnon-phonon scattering limits κ\kappa below ∼\sim 2 W m−1^{-1} K−1^{-1}, which would be three times larger if magnetic scatterings were absent. Application of magnetic fields strongly suppresses the spin-phonon scattering, giving rise to large enhancements of κ\kappa. Our calculations well capture these complex behaviours of κ\kappa by taking the temperature- and magnetic-field-dependent spin-phonon scattering into account. Realization of low κ\kappa which is easily tunable by magnetic fields in Cr2_2Si2_2Te6_6, may further promote spincaloritronic applications of vdW magnets. Our theoretical approach may also provide a generic understanding of spin-phonon scattering, which appears to play important roles in various systems.Comment: 14 pages, 6 figures, accepted for publication in Advanced Functional Material

    Promoter Hypermethylation Mediated Downregulation of FBP1 in Human Hepatocellular Carcinoma and Colon Cancer

    FBP1, fructose-1,6-bisphosphatase-1, a gluconeogenesis regulatory enzyme, catalyzes the hydrolysis of fructose 1,6-bisphosphate to fructose 6-phosphate and inorganic phosphate. The mechanism that it functions to antagonize glycolysis and was epigenetically inactivated through NF-kappaB pathway in gastric cancer has been reported. However, its role in the liver carcinogenesis still remains unknown. Here, we investigated the expression and DNA methylation of FBP1 in primary HCC and colon tumor. FBP1 was lowly expressed in 80% (8/10) human hepatocellular carcinoma, 66.7% (6/9) liver cancer cell lines and 100% (6/6) colon cancer cell lines, but was higher in paired adjacent non-tumor tissues and immortalized normal cell lines, which was well correlated with its promoter methylation status. Methylation was further detected in primary HCCs, gastric and colon tumor tissues, but none or occasionally in paired adjacent non-tumor tissues. Detailed methylation analysis of 29 CpG sites at a 327-bp promoter region by bisulfite genomic sequencing confirmed its methylation. FBP1 silencing could be reversed by chemical demethylation treatment with 5-aza-2′-deoxycytidine (Aza), indicating direct epigenetic silencing. Restoring FBP1 expression in low expressed cells significantly inhibited cell growth and colony formation ability through the induction of G2-M phase cell cycle arrest. Moreover, the observed effects coincided with an increase in reactive oxygen species (ROS) generation. In summary, epigenetic inactivation of FBP1 is also common in human liver and colon cancer. FBP1 appears to be a functional tumor suppressor involved in the liver and colon carcinogenesis

    Phenolic Profiling of Duchesnea indica Combining Macroporous Resin Chromatography (MRC) with HPLC-ESI-MS/MS and ESI-IT-MS

    Duchesnea indica (D. indica) is an important traditional Chinese medicine, and has long been clinically used to treat cancer in Asian countries. It has been described previously as a rich source of phenolic compounds with a broad array of diversified structures, which are the major active ingredients. However, an accurate and complete phenolic profiling has not been determined yet. In the present work, the total phenolic compounds in crude extracts from D. indica were enriched and fractionated over a macroporous resin column, then identified by HPLC-ESI-MS/MS and ESI-IT-MS (ion trap MS). A total of 27 phenolic compounds were identified in D. indica, of which 21 compounds were identified for the first time. These 27 phenolic compounds encompassing four phenolic groups, including ellagitannins, ellagic acid and ellagic acid glycosides, hydroxybenzoic acid and hydroxycinnamic acid derivatives, and flavonols, were then successfully quantified using peak areas against those of the corresponding standards with good linearity (R2 > 0.998) in the range of the tested concentrations. As a result, the contents of individual phenolic compounds varied from 6.69 mg per 100 g dry weight (DW) for ellagic acid to 71.36 mg per 100 g DW for brevifolin carboxylate. Not only did this study provide the first phenolic profiling of D. indica, but both the qualitative identification and the subsequent quantitative analysis of 27 phenolic compounds from D. indica should provide a good basis for future exploration of this valuable medicinal plant

    Quantitative Analysis and Comparison of Flavonoids in Lotus Plumules of Four Representative Lotus Cultivars

    The flavonoids in lotus plumules from four representative lotus cultivars have been analyzed using high-performance liquid chromatography coupled with ultraviolet detector and electrospray ionization triple quadrupole mass spectrometry. By this means, sixteen flavonoids were successfully measured and compared among four cultivars. Although similar flavonoid compositions were detected from these four cultivars, their flavonoid contents were significantly different. cv. Bailian from Guangchang was detected to have the most total flavonoid content, that is, 1595.86 mg/100 g, followed by cv. Xuanlian from Wuyi (1553.49 mg/100 g), cv. Xianglian from Xiangtan (1173.07 mg/100 g), and cv. Jianlian from Jianning (930.08 mg/100 g). However, similar percentages of flavonoid C-glycosides in the total flavonoids were found for four cultivars, which were 80.83% for cv. Bailian, 80.91% for cv. Xianglian, 79.25% for cv. Jianlian, and 78.53% for cv. Xuanlian. This work will be very useful for quality assurance and control for the lotus plumules from different origins in terms of qualitative and quantitative information about flavonoids. It will also be of special interest in the screening of lotus plumules with high flavonoid content, which are preferred due to their wide potential applications in food and pharmaceutical industries
