    Analysis of Compressive Toughness and Deformability of High Ductile Fiber Reinforced Concrete

    The compressive toughness evaluation index of HDC (high ductile fiber reinforced concrete) is studied through three groups of uniaxial compressive tests of HDC specimens with different fiber mixing amounts, and an equivalent analysis of their deformability is carried out, coming to the following conclusion: (1) the peak strain of HDC under uniaxial compression can be up to 3.41~3.67 times as large as that of the mortar matrix; (2) the equivalent compressive toughness index reflects the unit volume deformation energy of specimens under uniaxial compression and it can be used as the compressive toughness evaluation index of HDC; (3) the fiber bridging effect of HDC increases the equivalent compressive toughness index and the compressive deformability up to 3 times of the mortar matrix; (4) the relationship between the equivalent compressive toughness index Wcu0.85 and the fiber mixing amount φ is established according to the test results; and (5) the fiber bridging effect of the matrix in HDC can be equaling as a large number of constraint stirrups installed in the specimens, which significantly enhances the compressive toughness and the compressive deformability of specimens

    Pricing strategy research in the dual-channel pharmaceutical supply chain considering service

    In the context of developing a new era, the pharmaceutical supply chain market has gradually transformed from a seller’s market to a buyer’s market. The closer the consumers are, the greater the market pricing power, so the pharmaceutical market power of manufacturers and retailers has also changed. This study considers the effect of service on the pricing strategy of the pharmaceutical platform supply chain. The study aimed to coordinate optimization, and the coordination strategy of the pharmaceutical platform supply chain of complementary products is discussed mainly by researching the price and service factors. Various situations are studied by hypothesis and model solving. This study uses Stackelberg game decision-making. Manufacturers are at the forefront of platform supply chain decisions. The research found that the price was lower under centralized decision-making than under decentralized decision-making. Coordination between price and service levels needs attention in the pharmaceutical platform supply chain of complementary products, and the service level should be controlled within a certain range. Only by improving the service level can enterprises maximize profits, providing a theoretical basis for pharmaceutical supply chain pricing strategy research. Supply chain members must strive to improve service levels to improve medical supply consumers’ (patients) psychological satisfaction level. Service levels do not fully mitigate channel conflict. Therefore, pharmaceutical complements have become a way to alleviate the conflicts in the pharmaceutical platform supply chain

    Genome-wide identification and expression profiling of B3 transcription factor genes in Populus alba × Populus glandulosa

    B3-domain containing transcription factors (TFs) are well known to play important roles in various developmental processes, including embryogenesis, seed germination, etc. Characterizations and functional studies of the B3 TF superfamily in poplar are still limited, especially on their roles in wood formation. In this study, we conducted comprehensive bioinformatics and expression analysis of B3 TF genes in Populus alba × Populus glandulosa. A total of 160 B3 TF genes were identified in the genome of this hybrid poplar, and their chromosomal locations, syntenic relationships, gene structures, and promoter cis-acting elements were analyzed. Through domain structure and phylogenetic relationship analyses, these proteins were classified into four families LAV, RAV, ARF, and REM. Domain and conservation analyses revealed different gene numbers and different DNA-binding domains among families. Syntenic relationship analysis suggested that approximately 87% of the genes resulted from genome duplication (segmental or tandem), contributing to the expansion of the B3 family in P. alba × P. glandulosa. Phylogeny in seven species revealed the evolutionary relationship of B3 TF genes across different species. B3 domains among the eighteen proteins that were highly expressed in differentiating xylem had a high synteny, suggesting a common ancestor for these seven species. We performed co-expression analysis on the representative genes in two different ages of poplar, followed by pathways analysis. Among those genes co-expressed with four B3 genes, 14 were involved in lignin synthases and secondary cell walls biosynthesis, including PagCOMT2, PagCAD1, PagCCR2, PagCAD1, PagCCoAOMT1, PagSND2, and PagNST1. Our results provide valuable information for the B3 TF family in poplar and show the potential of B3 TF genes in engineering to improve wood properties

Mullis developed a revolutionary method name polymerase chain reaction (PCR) in 1983, which can synthesize new strand of DNA complementary to the template strand of DNA and produce billions of copies of a DNA fragment only in few hours. Denaturation, annealing, and extension are the three primary steps involved in the PCR process, which generally requires thermocyclers, DNA template, a pair of primers, Taq polymerase, nucleotides, buffers, etc. With the development of PCR, from traditional PCR, quantitative PCR, to next digital PCR, PCR has become a powerful tool in life sciences and medicine. Applications of PCR techniques for infectious diseases include specific or broad-spectrum pathogen detection, assessment and surveillance of emerging infections, early detection of biological threat agents, and antimicrobial resistance analysis. Applications of PCR techniques for genetic diseases include prenatal diagnosis and screening of neonatal genetic diseases. Applications of PCR techniques for cancer research include tumor-related gene detection. This chapter aimed to discuss about the different types of PCR techniques, including traditional PCR, quantitative PCR, digital PCR, etc., and their applications for rapid detection, mutation screen or diagnosis in infectious diseases, inherited diseases, cancer, and other diseases

    Spectral self-adaptive absorber/emitter for harvesting energy from the sun and outer space

    The sun (~6000 K) and outer space (~3 K) are the original heat source and sink for human beings on Earth. The energy applications of absorbing solar irradiation and harvesting the coldness of outer space for energy utilization have attracted considerable interest from researchers. However, combining these two functions in a static device for continuous energy harvesting is unachievable due to the intrinsic infrared spectral conflict. In this study, we developed spectral self-adaptive absorber/emitter (SSA/E) for daytime photothermal and nighttime radiative sky cooling modes depending on the phase transition of the vanadium dioxide coated layer. A 24-hour day-night test showed that the fabricated SSA/E has continuous energy harvesting ability and improved overall energy utilization performance, thus showing remarkable potential in future energy applications.Comment: 15 pages, 4 figure

    Mechanically Robust and Spectrally Selective Convection Shield for Daytime Subambient Radiative Cooling

    As a passive cooling strategy, radiative cooling is becoming anappealing approach to dissipate heat from terrestrial emitters to the outer space. However, the currently achieved cooling performance is still underperforming due to considerable solar radiation absorbed by the emitter and nonradiative heat transferred from the surroundings. Here, we proposed a mechanically robust and spectrally selective convection shield composed of nanoporous composite fabric (NCF) to achieve daytime subambient radiative cooling. By selectively reflecting ∼95% solar radiation, transmitting ∼84% thermal radiation, and suppressing the nonradiative heat transferred from warmer surroundings, the NCF-based radiative cooler demonstrated an average daytime temperature reduction of ∼4.9 °C below the ambient temperature, resulting in an average net radiative cooling power of ∼48 W/m2 over a 24 h measurement. In addition, we also modeled the potential cooling capacity of the NCF-based radiative cooler and demonstrated that it can cover the cooling demands of energy-efficient residential buildings in most regions of China. Excellent spectral selectivity, mechanical strength, and weatherability of the NCF cover enable a much broader selection for the emitters, which is promising in the real-world deployment of direct daytime subambient radiative cooling

    Suppressing Effect of Na+/Ca2+ Exchanger (NCX) Inhibitors on the Growth of Melanoma Cells

    The role of calcium ion (Ca2+) signaling in tumorigenicity has received increasing attention in melanoma research. Previous Ca2+ signaling studies focused on Ca2+ entry routes, but rarely explored the role of Ca2+ extrusion. Functioning of the Na+/Ca2+ exchanger (NCX) on the plasma membrane is the major way of Ca2+ extrusion, but very few associations between NCX and melanoma have been reported. Here, we explored whether pharmacological modulation of the NCX could suppress melanoma and promise new therapeutic strategies. Methods included cell viability assay, Ca2+ imaging, immunoblotting, and cell death analysis. The NCX inhibitors SN-6 and YM-244769 were used to selectively block reverse operation of the NCX. Bepridil, KB-R7943, and CB-DMB blocked either reverse or forward NCX operation. We found that blocking the reverse NCX with SN-6 or YM-244769 (5–100 μM) did not affect melanoma cells or increase cytosolic Ca2+. Bepridil, KB-R7943, and CB-DMB all significantly suppressed melanoma cells with IC50 values of 3–20 μM. Bepridil and KB-R7943 elevated intracellular Ca2+ level of melanoma. Bepridil-induced melanoma cell death came from cell cycle arrest and enhanced apoptosis, which were all attenuated by the Ca2+ chelator BAPTA-AM. As compared with melanoma, normal melanocytes had lower NCX1 expression and were less sensitive to the cytotoxicity of bepridil. In conclusion, blockade of the forward but not the reverse NCX leads to Ca2+-related cell death in melanoma and the NCX is a potential drug target for cancer therapy
