    Evaluation of roughness, hardness, and strength of AA 6061 molds for manufacturing polymeric microdevices

    In the manufacturing of polymeric microfluidic devices, micro-molds play a key role because they determine not only the manufacturing cost but also the quality of the molded parts. Recently, a high-quality aluminum alloy 6061 (AA6061) mold with fine features less than its grain size has been fabricated economically by a hot embossing technique. However, temperature cycling during hot embossing process in mold manufacturing reduces significantly the original tensile strength and hardness of the AA6061-T6 alloy substrate, which is not desirable. In this study, a tempering process is carried out to recover the tensile strength and hardness of the embossed mold. To evaluate the changes of these properties, surface roughness, tensile strength, and hardness values were measured in each stage: (1) before hot embossing, (2) after hot embossing, and (3) tempering to T4 and tempering to T6. The results obtained demonstrate that the original strengths and hardness can be fully recovered by a post-tempering process after hot embossing, but with an increase in surface roughness. Moreover, accelerated testing was carried out to evaluate the changes in hardness and roughness of AA6061-T4 and T6 molds under the typical hot embossing temperature cycles of manufacturing polymeric devices. The results obtained indicate that these temperature cycles have only a minor effect on the roughness of both T4 and T6 molds and will increase the hardness of T4 molds to T6 temper, and have negligible effect on the hardness of a T6 temper mold

    Dual Effects of Chinese Herbal Medicines on Angiogenesis in Cancer and Ischemic Stroke Treatments: Role of HIF-1 Network

    This work is licensed under a Creative Commons Attribution 4.0 International License.Hypoxia-inducible factor-1 (HIF-1)–induced angiogenesis has been involved in numerous pathological conditions, and it may be harmful or beneficial depending on the types of diseases. Exploration on angiogenesis has sparked hopes in providing novel therapeutic approaches on multiple diseases with high mortality rates, such as cancer and ischemic stroke. The HIF-1 pathway is considered to be a major regulator of angiogenesis. HIF-1 seems to be involved in the vascular formation process by synergistic correlations with other proangiogenic factors in cancer and cerebrovascular disease. The regulation of HIF-1–dependent angiogenesis is related to the modulation of HIF-1 bioactivity by regulating HIF-1α transcription or protein translation, HIF-1α DNA binding, HIF-1α and HIF-1α dimerization, and HIF-1 degradation. Traditional Chinese herbal medicines have a long history of clinical use in both cancer and stroke treatments in Asia. Growing evidence has demonstrated potential proangiogenic benefits of Chinese herbal medicines in ischemic stroke, whereas tumor angiogenesis could be inhibited by the active components in Chinese herbal medicines.     Polarization-based cyclic weak value metrology for angular velocity measurement

    Weak value has been proved to amplify the detecting changes of the meters at the cost of power due to post-selection. Previous power-recycling schemes enable the failed post-selection photons to be reselected repeatedly, thus surpassing the upper noise limit and improving the precision of interferometric systems. Here we introduce three cyclic methods to improve the sensitivity of polarization-based weak-value-based angular velocity measurement: power-, signal- and dual-recycling schemes. By inserting one or two partially transmitting mirrors inside the system, both the power and precision of detected signals are greatly enhanced, and the dual-recycling scheme has wider optimal region than that of power- or signal-recycling schemes. Compared to non-polarization schemes, polarization-based schemes enjoy lower optical loss and unique cyclic directions. These reduce the crosstalk among different paths of light and, theoretically, eliminate the walk-off effect, thus towering in both theoretical performance and application.Comment: 7 pages, 3 figure

    Screening A Trinucleotide Repeat Expansion: How precise PCR can be?

    ABSTRACT Background. Trinucleotide Repeat Expansion (TRE) in human DNA could lead to various diseases. An expanded CAG repeat (>31 or 37 repeats, depends on the ethnicity) in Androgen Receptor gene is suggested to be associated with the occurrence of isolated hypospadias. In an effort to identify the exact numbers of repeats, sequencing has been the most favored method to be conducted despite its cost. Objective. This study wished to investigate the possibilities of using Polymerase Chain Reaction (PCR) method to screen expanded repeats in isolated hypospadias, as one of the TRE diseases. Materials and Methods. Numbers of CAG repeat in twelve hypospadias patients and one normal male was first predicted from the visualization of PCR products in 3% agarose gel electrophoreses with 20 bp ladder marker before it was finally sequenced. Results. Two samples gave the same exact result, while the rest showed a range of 1-11 bp differences. Statistically, there was a significant difference between the mean of CAG repeats from PCR method (M=26.1667, SD=6.71272) and the mean of CAG repeats from sequencing (M=23.75, SD=5.70685); t(11)= 4.570, p=0.001. Furthermore, the sensitivity of PCR was 100% and the specificity was 83.33%. Conclusion. It can be concluded that PCR method could be used as a screening method in identifying TRE with large numbers of repeats. However, PCR in TRE disease with small numbers of expanded repeats needs to be followed by sequencing in order to obtain the exact numbers of repeats.   Keywords: Trinucleotide Repeat Expansion, Polymerase Chain Reaction, Sequencing, Isolated Hypospadia

    Preclinical Models for Investigation of Herbal Medicines in Liver Diseases: Update and Perspective

    Liver disease results from a dynamic pathological process associated with cellular and genetic alterations, which may progress stepwise to liver dysfunction. Commonly, liver disease begins with hepatocyte injury, followed by persistent episodes of cellular regeneration, inflammation, and hepatocyte death that may ultimately lead to nonreversible liver failure. For centuries, herbal remedies have been used for a variety of liver diseases and recent studies have identified the active compounds that may interact with liver disease-associated targets. Further study on the herbal remedies may lead to the formulation of next generation medicines with hepatoprotective, antifibrotic, and anticancer properties. Still, the pharmacological actions of vast majority of herbal remedies remain unknown; thus, extensive preclinical studies are important. In this review, we summarize progress made over the last five years of the most commonly used preclinical models of liver diseases that are used to screen for curative herbal medicines for nonalcoholic fatty liver disease, liver fibrosis/cirrhosis, and liver. We also summarize the proposed mechanisms associated with the observed liver-protective, antifibrotic, and anticancer actions of several promising herbal medicines and discuss the challenges faced in this research field

    The Potential and Action Mechanism of Polyphenols in the Treatment of Liver Diseases

    Liver disease, involving a wide range of liver pathologies from fatty liver, hepatitis, and fibrosis to cirrhosis and hepatocellular carcinoma, is a serious health problem worldwide. In recent years, many natural foods and herbs with abundant phytochemicals have been proposed as health supplementation for patients with hepatic disorders. As an important category of phytochemicals, natural polyphenols have attracted increasing attention as potential agents for the prevention and treatment of liver diseases. The striking capacities in remitting oxidative stress, lipid metabolism, insulin resistance, and inflammation put polyphenols in the spotlight for the therapies of liver diseases. It has been reported that many polyphenols from a wide range of foods and herbs exert therapeutic effects on liver injuries via complicated mechanisms. Therefore, it is necessary to have a systematical review to sort out current researches to help better understand the potentials of polyphenols in liver diseases. In this review, we aim to summarize and update the existing evidence of natural polyphenols in the treatment of various liver diseases by in vitro, in vivo, and clinical studies, while special attention is paid to the action mechanisms

    Multiscale Characterized Methods for the Stability of Whole Grain Drink

    With the popularization of the concepts of whole grain health cognition and scientific diet, consumers are paying more and more attention to different forms of whole grain food. Whole grain drink is one of the important products made from whole grain, and the physical stability of whole grain drink is a major challenge in the process of research and development. Since whole grain drink is a thermodynamically unstable system of multiphase turbidite liquid with water as the continuous phase and starch, dietary fiber, protein as the dispersed phase, the separation speed of whole grain drink is fast, which seriously affected the acceptability of consumers. Based on literature review, the multiscale characterization methods of macroscopic, mesoscopic, microscopic and nano-scale for the stability of whole grain drink and characterization methods of the characteristic chemical components were summarized. This paper summarized the stability of whole grain drink characterized with multiscale methods, which could provide methodological guidance for the development of whole grain drink and facilitate the rapid development of whole grain foods
