    Study of Saiga Horn Using High-Performance Liquid Chromatography with Mass Spectrometry

    The saiga horns have been investigated the using of modern analytic methods. High-performance liquid chromatography (HPLC) with mass-spectrometric (MS and MS/MS) detection and polyacrylamide gel electrophoresis (PAGE) were used. It could be concluded that basic proteins of the saiga horns are keratins and collagen. The basic representation protein in all samples is keratin type I microfibrillar (from sheep), keratin type II microfibrillar (from sheep), collagen type I (α1) (from bovine) and collagen type I (α2) (from bovine).     Eggshell pigment composition covaries with phylogeny

    No single hypothesis is likely to explain the diversity in eggshell coloration and patterning across birds, suggesting that eggshell appearance is most likely to have evolved to fulfill many nonexclusive functions. By controlling for nonindependent phylogenetic associations between related species, we describe this diversity using museum eggshells of 71 British breeding passerine species to examine how eggshell pigment composition and concentrations vary with phylogeny and with life-history and nesting ecology traits. Across species, concentrations of biliverdin and protoporphyrin, the two main pigments found in eggshells, were strongly and positively correlated, and both pigments strongly covaried with phylogenetic relatedness. Controlling for phylogeny, cavity-nesting species laid eggs with lower protoporphyrin concentrations in the shell, while higher biliverdin concentrations were associated with thicker eggshells for species of all nest types. Overall, these relationships between eggshell pigment concentrations and the biology of passerines are similar to those previously found in nonpasserine eggs, and imply that phylogenetic dependence must be considered across the class in further explanations of the functional significance of avian eggshell coloration

    Modification of Nonlinear Mapping Technique for Quantitative Structure-Retention Relationship Studies

    The retention of 19 solutes (Ala, Gly, Lys, Phe and their homopeptides) has been determined on impregnated alumina layers using various mixtures of water-methanol as mobile phases. Lipophilicity and specific hydrophobic surface area of solutes were estimated from linear correlations between the retention of solutes and the concentration of methanol in the mobile phase. The relationship between the hydrophobic retention parameters and physicochemical parameters has been elucidated by the principal component (PC) analysis followed by nonlinear mapping carried out on both the original and absolute values of PC loadings and variables. Ala, Gly, Lys and their homopeptides showed anomalous behavior, the retention increasing with increasing the concentration of methanol in the mobile phase. The use of absolute values in the calculations enhanced the reliability of the mapping technique. It was found that other than hydrophobic forces are involved in the solute stationary phase interaction (mixed retention mechanism)

    A relationship has been suggested between eggshell colour and female body condition based on the opposing antioxidant properties of the two main eggshell pigments: the antioxidant biliverdin (blue–green) and the pro-oxidant protoporphyrin (brown). We hypothesized that experimentally food-restricted females with low antioxidant capacity would deposit more protoporphyrin and less biliverdin in their eggshells, resulting in eggshells of reduced brightness but increased colour intensity. Two eggs were collected at the beginning and two at the end of a 2 week period from each of 24 female Japanese quails that were either food restricted or receiving ad libitum food (i.e. controls) during that time. Reflectance spectra were recorded and analysed using spectral shape descriptors, chromatic and achromatic contrasts were computed accounting for avian visual sensitivities, and eggshell pigments were quantified. We examined both spot and background pigmentation and found no significant effect of food restriction on eggshell reflectance. However, food-restricted females in lower body condition increased the deposition of protoporphyrin and decreased the amount of biliverdin invested in their eggshells. We hypothesize that in species laying brown-spotted eggshells, females modulate eggshell pigment investment in response to their body condition. According to this hypothesis, we predict that females maintain eggshell colour to limit visible changes that could be detected by predators and thereby conceal their eggs, although this work has yet to be conducted. We suggest that further experimental work on egg camouflage under different environmental conditions will elaborate on the process of pigment deposition and the physiological costs to females of laying heavily pigmented eggshells

    Article first published online: 4 DEC 2013Eggshell maculation of most passerines is due to the deposition of the pigment protoporphyrin which is produced during biosynthesis of blood haem. Its functional significance has only received empirical attention in recent years. This interest has generated a number of hypotheses of which some remain untested partly because the quantification of protoporphyrin is analytically challenging and can be prohibitively expensive. Many studies have therefore used the extent of eggshell spotting as a proxy for total eggshell protoporphyrin concentration, although this has not been formally tested. Pigment scoring involves recording visible eggshell pigment attributes, such as spot intensity, distribution and size. Since even immaculate eggs can contain some protoporphyrin, there remains doubt over the degree to which visible pigment correlates with total pigment content of the shell. In this study, we test whether visible pigment scoring can be used as a proxy for protoporphyrin concentration of an eggshell. We use pigmented eggshells of two common British passerine species to compare eggshell spot intensity, distribution and spot size (as used by the visual pigment scoring method) with direct measures of eggshell protoporphyrin concentration. In addition, we compared an alternative method of pigment scoring, the pixel pigment scoring method, using a computer programme to quantify the number of pixels exceeding a specified colour threshold.     Reproduction is a critical period for birds as they have to cope with many stressful events. One consequence of an acute exposure to stress is the release of corticosterone, the avian stress hormone. Prolonged stress can have negative impacts on the immune system, resulting in, for example, increased oxidative stress. Through maternal effects, females are known to modulate their investment in eggs content according to their own physiological condition. Less is known about maternal investment in eggshells, especially in pigments. The two main eggshell pigments may possess opposite antioxidant properties: protoporphyrin (brown) is a pro-oxidant, whereas biliverdin (blue-green) is an antioxidant. In Japanese quail, we know that the deposition of both pigments is related to female body condition. Thus, a chronic stress response may be reflected in eggshell coloration. Using female Japanese quails that lay brown-spotted eggs, we explored whether physiological exposure to corticosterone induces a change in female basal stress and antioxidant factors, and eggshell pigment concentration, spectrophotometric reflectance, and maculation coverage. We supplemented adult females over a 2 week period with either peanut oil (control) or corticosterone (treatment). We collected pre- and post-supplementation eggs and analysed the effect of corticosterone treatment on female physiology and eggshell appearance parameters. Except for corticosterone-fed birds which laid eggs with brighter spots, supplementation had no significant effect on female physiology or eggshell pigment concentration, reflectance and maculation. The change in eggshell spot brightness was not detected by a photoreceptor noise-limited color opponent model of avian visual perception. Our data confirms that eggshell reflectance in spotted eggs varies over the laying sequence, and spot reflectance may be a key factor that is affected by females CORT exposure, even if the changes are not detected by an avian visual model.</p
