    Best Practices among Certain Classes of Pennsylvania Destination Marketing Organizations (DMOs)

    The researchers originally became aware of the existence of DMOs (previously known as tourism promotion agencies or TPAs) when reading materials in the Gettysburg Times. The two student-authors plan to work in local government following graduation, and they were curious about perspectives shared in the Gettysburg Times about the local DMO. Thereafter, a representative from the Gettysburg Borough Council approached the faculty-author about conducting research on DMOs’ policies, procedures, and practices, and, relatedly, the “pillow tax,” a hotel room occupancy tax that is administered by counties and disbursed to DMOs, among others. Specifically, the Borough Council representative was curious about best practices with respect to allocation of the pillow tax; how DMOs use pillow tax revenue; and how DMOs account for such use. The Borough representative also was curious about general business practices among DMOs. The faculty-author applied for and obtained approval from the Gettysburg College Institutional Review Board for research among Pennsylvania DMOs and their representatives on these issues. This white paper briefly summarizes the history of the relationship between Pennsylvania DMOs and the pillow tax then describes the researchers’ methodology. Next, the white paper describes what Study DMOs appear to believe are best practices, or most commonly accepted practices, in six categories: Boards of Directors; Revenue; Grant Making; Spending; Assessment; and Future Planning. All data are reported in aggregate to minimize the risk of revealing individually identifiable data. These findings may inform DMO, community, and county decision-making

    A Gene-Based Association Method for Mapping Traits Using Reference Transcriptome Data

    Genome-wide association studies (GWAS) have identified thousands of variants robustly associated with complex traits. However, the biological mechanisms underlying these associations are, in general, not well understood. We propose a gene-based association method called PrediXcan that directly tests the molecular mechanisms through which genetic variation affects phenotype. The approach estimates the component of gene expression determined by an individual’s genetic profile and correlates ‘imputed’ gene expression with the phenotype under investigation to identify genes involved in the etiology of the phenotype. Genetically regulated gene expression is estimated using whole-genome tissue-dependent prediction models trained with reference transcriptome data sets. PrediXcan enjoys the benefits of gene-based approaches such as reduced multiple-testing burden and a principled approach to the design of follow-up experiments. Our results demonstrate that PrediXcan can detect known and new genes associated with disease traits and provide insights into the mechanism of these associations

    Evaluation of the Physicochemical, Structural, Thermal, and Behavioral Properties of the Energy of Consciousness Healing Treated Zinc Chloride

    Zinc chloride is a source of zinc used in various pharmaceutical/nutraceutical formulations. The objective of the current study was to investigate the impact of The Trivedi Effect® - Energy of Consciousness Healing Treatment (Biofield Energy Treatment) on physical, structural, thermal, and behavioral properties of zinc chloride using PXRD, PSD, FT-IR, UV-vis, and DSC analysis. Zinc chloride was divided into two parts – one part was control, while another part was treated with The Trivedi Effect® remotely by twenty renowned Biofield Energy Healers and defined as The Trivedi Effect® Treated sample. A significant alteration of the crystallite size and relative intensities of the PXRD peaks was observed in The Trivedi Effect® treated sample compared with the control sample. The average crystallite size of the treated sample was significantly increased by 23.18% compared with the control sample. The particle size values at d10, d50, and d90 values were significantly decreased by 3.70%, 4.13%, and 6.13%, respectively in the treated sample compared with the control sample. Therefore, the surface area of the treated sample was increased by 4.21% compared with the control sample. The FT-IR spectroscopic analysis revealed that Zn-Cl stretching in the control sample was found at 512 cm-1, whereas it was significantly shifted upward to 520 cm-1 in the treated sample. The UV-vis analysis exhibited that wavelength of the maximum absorbance (λmax) of the control and treated samples were at 197.6 nm and 197.1 nm, respectively. The DSC analysis exhibited that the melting temperature was decreased by 0.22%, while decomposition temperature was increased by 2.56% in the treated sample compared to the control sample. The latent heat of fusion of the treated sample (320.44 J/g) was significantly decreased by 16.70% compared with the control sample (284.67 J/g). Similarly, the enthalpy of decomposition of the treated sample (952.53 J/g) was significantly increased by 122.61% compared with the control sample (427.90 J/g). Thus, the results indicated that the thermal stability of the treated zinc chloride was improved compared with the control sample. The current study anticipated that The Trivedi Effect® - Energy of Consciousness Healing Treatment might lead to produce a thermally stable new polymorphic form of zinc chloride, which would be more soluble and bioavailable compared with the untreated compound.     Evaluation of the Impact of the Trivedi Effect® -Energy of Consciousness on the Structure and Isotopic Abundance Ratio of Magnesium Gluconate Using LC-MS and NMR Spectroscopy

    Magnesium gluconate is a classical pharmaceutical/nutraceutical compound used as a magnesium ion source for the prevention and treatment of hypomagnesemia. The present study was aimed to investigate the effect of The Trivedi Effect® - Energy of Consciousness Healing Treatment (Biofield Energy Healing Treatment) on magnesium gluconate for the change in the structural properties and isotopic abundance ratio (PM+1/PM and PM+2/PM) using LC-MS and NMR spectroscopy. Magnesium gluconate was divided into two parts – one part was control, and another part was treated with The Trivedi Effect® - Biofield Energy Healing Treatment remotely by twenty renowned Biofield Energy Healers and defined as The Trivedi Effect® Treated sample. The LC-MS analysis of both the control and treated samples indicated the presence of mass of the protonated magnesium gluconate at m/z 415 at the retention time of 1.52 min and fragmentation pattern of the both sample were almost similar. The relative peak intensities of the fragment ions were significantly changed in the treated sample compared with the control sample. The proton and carbon signals for CH, CH2 and CO groups in the proton and carbon NMR spectra were observed almost similar for the control and the treated samples. The percentage change in the isotopic abundance ratio of PM+1/PM (2H/1H or 13C/12C or 17O/16O or 25Mg/24Mg) was significantly decreased in the treated sample by 17.51% compared with the control sample. Consequently, the isotopic abundance ratio of PM+2/PM (18O/16O or 26Mg/24Mg) in the treated sample was significantly increased by 79.44% compared to the control sample. Briefly, 13C, 2H, 17O, and 25Mg contributions from (C12H23MgO14)+ to m/z 416; 18O and 26Mg contributions from (C12H23MgO14)+ to m/z 417 in treated sample were significantly altered compared with the control sample. Thus, The Trivedi Effect® Treated magnesium gluconate might be supportive to design the novel potent enzyme inhibitors using its kinetic isotope effects. Consequently, The Trivedi Effect® Treated magnesium gluconate would be valuable for designing better pharmaceutical and/or nutraceutical formulations through its changed physicochemical and thermal properties, which might be providing better therapeutic response against various diseases such as diabetes mellitus, allergy, aging, inflammatory diseases, immunological disorders, and other chronic infections.     Identification of alleles of carotenoid pathway genes important for zeaxanthin accumulation in potato tubers

    We have investigated the genetics and molecular biology of orange flesh colour in potato (Solanum tuberosum L.). To this end the natural diversity in three genes of the carotenoid pathway was assessed by SNP analyses. Association analysis was performed between SNP haplotypes and flesh colour phenotypes in diploid and tetraploid potato genotypes. We observed that among eleven beta-carotene hydroxylase 2 (Chy2) alleles only one dominant allele has a major effect, changing white into yellow flesh colour. In contrast, none of the lycopene epsilon cyclase (Lcye) alleles seemed to have a large effect on flesh colour. Analysis of zeaxanthin epoxidase (Zep) alleles showed that all (diploid) genotypes with orange tuber flesh were homozygous for one specific Zep allele. This Zep allele showed a reduced level of expression. The complete genomic sequence of the recessive Zep allele, including the promoter, was determined, and compared with the sequence of other Zep alleles. The most striking difference was the presence of a non-LTR retrotransposon sequence in intron 1 of the recessive Zep allele, which was absent in all other Zep alleles investigated. We hypothesise that the presence of this large sequence in intron 1 caused the lower expression level, resulting in reduced Zep activity and accumulation of zeaxanthin. Only genotypes combining presence of the dominant Chy2 allele with homozygosity for the recessive Zep allele produced orange-fleshed tubers that accumulated large amounts of zeaxanthin

    "To Tell the Story": Cultural Trauma and Holocaust Metanarrative

    This article explores the aporia between the alleged inexplicability of the Holocaust and the wealth of narrative that has proceeded from the event in the years since 1945, proposing the existence of a generic Holocaust metanarrative that has been adopted and inscribed into Western cultural memory as the accepted framework for interpretation. Taking as a starting point the idea that culture itself has been somehow ‘ruptured’ in the wake of the Holocaust, this article explores the ways in which this rupture manifests itself, viewing the shattering impact of the Holocaust on the Western cultural imagination as macrocosmically comparable to the impact of psychic trauma on the individual survivor of the Holocaust. Just as an individual act of narration (the act of testimony) is believed to provide a cure for trauma, so a collective act of narration may hold the key to repairing the post-Holocaust cultural rupture. During the exploration of this process, it becomes apparent that cultural memory of the Holocaust is in fact informed by a metanarrative account that appears to offer the possibility of an engagement with the Holocaust, but which in fact acts as a screen between the event itself and the culture that would seek to memorialize it. Finally, this article explores the notion that the most appropriate narrative response is one that accepts the impossibility of its own position, rejecting the easy redemption offered by the assimilation of Holocaust metanarrative and instead inhabiting the dialectic between knowing and understanding that the Holocaust presents

    Navigating New Landscapes: The Contribution of Socio-Legal Scholarship in Mapping the Plurality of International Economic Law and Locating Power in International Economic Relations

    The evolution of international economic law in the past two decades has been characterised by the growth and diversification of international economic actors, the expansion in the substantive areas governed by international law, and, crucially, the proliferation of multiple sites of international economic governance. This web of multi-layered international economic governance is, in turn, underpinned by complex dynamics of power which structure the legal and economic relations between the subjects of international economic law and other actors impacted by international legal rules and regulation. The challenge for international legal scholarship lay not only in mapping the multiple sites of international economic governance but also in unmasking the power dynamics inherent in international economic relations. Locating and analysing power relations underlying international economic law is to crucial to understanding the cause and effect of international economic rules and institutions for rulemaking. Conventional legal scholarship with its doctrinal focus, while useful in providing the foundational basis for analysis, cannot adequately capture the complexity of contemporary international economic law. Socio-legal approaches may be able to overcome these epistemological limitations by supplying: a) the methodologies to study international economic law beyond a focus on rules and institutions; and b) the critical theoretical lens to understand the power dynamics inherent in international legal relations. The objective of this paper is twofold: firstly, it will seek to identify the contributions of socio-legal approaches to the study of international economic law; and secondly, it will explore how socio-legal scholarship can provide a methodological and theoretical framework to construct an understanding of the pluralistic nature of international economic regulatory regimes and their underlying dynamics of power. In doing so, the paper will also consider the value of juxtaposing an empirical methodology for mapping legal regimes with a critical normative approach for analysing power relations in international economic law
