    New WS9326A Congeners from <i>Streptomyces</i> sp. 9078 Inhibiting <i>Brugia malayi</i> Asparaginyl-tRNA Synthetase

    Lymphatic filariasis is caused by the <i>Brugia malayi</i> parasite. Three new congeners of the depsipeptide WS9326A (<b>1</b>), WS9326C (<b>2</b>), WS9326D (<b>3</b>), and WS9326E (<b>4</b>), were isolated from <i>Streptomyces</i> sp. 9078 by using a <i>B. malayi</i> asparaginyl-tRNA synthetase (<i>Bm</i>AsnRS) inhibition assay. WS9326D specifically inhibits the <i>Bm</i>AsnRS, kills the adult <i>B. malayi</i> parasite, and does not exhibit significant general cytotoxicity to human hepatic cells, representing a new lead scaffold for antifilarial drug discovery

    <p>Data are presented as means ± SEM. The number of cells migrated in response to 1 ng/ml rBmAsnRS was 14, and 10 ng/ml rBmAsnRS caused the greatest migration (72.75) similar to 10 ng/ml VEGF. Asterisk (*) denotes significant differences (p<0.0001) for negative control vs 10 ng/ml rBmAsnRS and VEGF.</p

    Hexa-histidine tagged rBmAsnRS was purified from the sonicated supernatant of IPTG induced <i>E</i>. <i>coli</i> cultures expressing the full length wild type BmAsnRS cDNA.
