Motif Dynamics in Signed Directional Complex Networks
Complex networks evolve and vary their structure as time goes by. In
particular, the links in those networks have both a sign and a directionality.
To understand their structural principles, we measure the network motifs, which
are patterns that appear much more than one would expect in randomized
networks, considering both link properties. We propose motif dynamics, which is
a study to investigate the change in the number of motifs, and applied the
motif dynamics to an open evolving network model and empirical data. We confirm
that a non-cyclic motif has a greater correlation with the system size than a
cyclic structural motif. Furthermore, the motif dynamics can give us insight
into the friendship between freshmen in a university
Large-scale preparation of active caspase-3 in E. coli by designing its thrombin-activatable precursors
<p>Abstract</p> <p>Background</p> <p>Caspase-3, a principal apoptotic effector that cleaves the majority of cellular substrates, is an important medicinal target for the treatment of cancers and neurodegenerative diseases. Large amounts of the protein are required for drug discovery research. However, previous efforts to express the full-length caspase-3 gene in <it>E. coli </it>have been unsuccessful.</p> <p>Results</p> <p>Overproducers of thrombin-activatable full-length caspase-3 precursors were prepared by engineering the auto-activation sites of caspase-3 precursor into a sequence susceptible to thrombin hydrolysis. The engineered precursors were highly expressed as soluble proteins in <it>E. coli </it>and easily purified by affinity chromatography, to levels of 10–15 mg from 1 L of <it>E. coli </it>culture, and readily activated by thrombin digestion. Kinetic evaluation disclosed that thrombin digestion enhanced catalytic activity (<it>k</it>cat/<it>K</it><it>M</it>) of the precursor proteins by two orders of magnitude.</p> <p>Conclusion</p> <p>A novel method for a large-scale preparation of active caspase-3 was developed by a strategic engineering to lack auto-activation during expression with amino acid sequences susceptible to thrombin, facilitating high-level expression in <it>E. coli</it>. The precursor protein was easily purified and activated through specific cleavage at the engineered sites by thrombin, generating active caspase-3 in high yields.</p
Susceptibility to Oxidative Stress is Greater in Korean Patients with Coronary Artery Disease than Healthy Subjects
There are some evidences that the increased oxidative stress and thus increased oxidizability of lipoproteins and DNA can contribute to the development of certain human diseases, such as cardiovascular disease. To confirm the association of DNA damage with cardiovascular disease, we investigated susceptibility of DNA to oxidation in lymphocytes and oxidative stress related parameters in blood of patients with coronary artery disease (CAD). Subjects were consisted of 42 patients (27 men, 15 women) with documented CAD and 49 apparently healthy subjects (33 men, 16 women) as controls. Cellular DNA damage induced by 100 µM H2O2 was measured using Comet assay and quantified by TL. There were no differences in age (61.4 ± 1.7 years vs 62.0 ± 2.2 years) between the two groups. All the findings were shown to be independent of either sex or smoking habit. The patients showed significantly higher TL (87.3 ± 1.6 µm) compared to the control (79.3 ± 1.7 µm, p<0.01). Plasma TRAP, vitamin C, γ-tocopherol, and α-carotene levels in patients group were lower than those of control groups, while erythrocytic catalase activity increased in patients group. In conclusion, we observed that reduced overall antioxidant status was closely connected to higher susceptibility of DNA damage in CAD patients
Mixed-State Entanglement and Quantum Teleportation through Noisy Channels
The quantum teleportation with noisy EPR state is discussed. Using an optimal
decomposition technique, we compute the concurrence, entanglement of formation
and Groverian measure for various noisy EPR resources. It is shown analytically
that all entanglement measures reduce to zero when , where
is an average fidelity between Alice and Bob. This fact indicates
that the entanglement is a genuine physical resource for the teleportation
process. This fact gives valuable clues on the optimal decomposition for
higher-qubit mixed states. As an example, the optimal decompositions for the
three-qubit mixed states are discussed by adopting a teleportation with W-stateComment: 18 pages, 4 figure
Potassium chloride elicits enhancement of bilobalide and Ginkgolides production by Ginkgo biloba cell cultures
This study investigated the ability of potassium chloride (KCl) to elicit the production of bilobalide (BB), ginkgolide A (GA) and ginkgolide B (GB) by Ginkgo biloba cell suspension cultures. The salt stress by KCl treatments increased production of BB, GA and GB in both suspended cells and cultured medium. Especially, treatment of KCl 800 mM of highest concentration was stimulated emission into cultured medium BB, GA and GB compounds accumulated in cells. Although KCl 800 mM severely inhibited cells growth, the maximum content of GA and GB in cells was obtained in the treatment of KCl 800 mM, which was 1.9 and 4.0 times higher than the control. These results thus suggest that salt stress can afford enhanced production of secondary metabolites by plant cell cultures
