AKTIVITA OCT A GLYKAÄŚNĂŤ POĹ KOZENĂŤ V MITOCHONDRIĂŤCH JATER KRYSY Lucie Merhutová Diplomová práce. Univerzita Karlova v Praze, Farmaceutická fakulta v Hradci KrálovĂ©. 2007 Tato práce byla vytvoĹ™ena na základÄ› pokusĹŻ provedenĂ˝ch v LBBCV (LaboratoĹ™i buněčnĂ© biologie a biochemie stárnutĂ) v rámci Univerzity Paris Diderot - Paris 7. TĂ˝m tĂ©to laboratoĹ™e studuje mechanismy posttranslaÄŤnĂch neenzymatickĂ˝ch modifikacĂ proteinĹŻ, zahrnutĂ˝ch v patofyziologii stárnutĂ. Glykace je jednĂm z mechanismĹŻ odpovÄ›dnĂ˝ch za modifikaci nitrobuněčnĂ˝ch makromolekul, kterĂ© vedou ke ztrátÄ› jejich struktury a funkce. Ornitin karbamoyltransferáza (OCT), druhĂ˝ enzym moÄŤovinovĂ©ho cyklu, byl v pĹ™edchozĂm vĂ˝zkumu identifikován jako jeden z enzymĹŻ, kterĂ© jsou vĂ˝znamnÄ› glykovány uvnitĹ™ mitochondriĂ jater stárnoucĂch krys. CĂlem tĂ©to práce bylo sledovat glykaci OCT in vitro a ex vivo a jejĂ vliv na funkci tohoto enzymu. Vzorky purifikovanĂ© OCT a extraktĹŻ mitochondriálnĂ matrix byly inkubovány s methylglyoxalem, jednĂm z nejreaktivnÄ›jšĂch fyziologickĂ˝ch glykaÄŤnĂch ÄŤinidel. U vzorkĹŻ byla změřena enzymatická aktivita a byly podrobeny imunochemickĂ© analĂ˝ze. PodaĹ™ilo se nám prokázat, Ĺľe methylglyoxal modifikuje OCT jak in vitro, tak ex vivo, a zpĹŻsobuje velice rychlĂ˝ a rozsáhlĂ˝ pokles jeho enzymatickĂ© aktivity. Charles University in Prague, Faculty of Pharmacy in Hradec KrálovĂ©. 2007 This work was created following investigations carried out in LBBCV (Laboratoire de Biologie et Biochimie Cellulaire du Vieillissement) at University Paris Diderot - Paris 7. The team of this laboratory studies the mechanisms of posttranslational non-enzymatic modifications of proteins, involved in the pathophysiology of aging. Glycation is one of the mechanisms responsible for the modification of intracellular macromolecules leading to the loss of their structure and function. In previous investigations, ornithine carbamoyltransferase (OCT), the second enzyme of urea cycle, has been identified as one of the markedly glycated proteins in liver mitochondria of senescent rats. The goal of this work was to investigate in vitro and ex vivo glycation of OCT and the effect of glycation on its function. Purified OCT and mitochondrial matrix extracts were incubated with methylglyoxal, one of the most reactive physiological glycating agents, and submitted to the enzymatic assay and immunochemical analysis. Creative techniques in supervision
The main aim of my diploma work called "Creative Techniques in Supervision", which is divided into two parts, is creating of a practical manual containing art-therapeutic techniques that can be used in supervision. This work is dedicated to all supervisors who are originally not art-therapist but despite this fact want use this way of work. The first part of my work is an introduction of the art-therapeutic method that (with the use of technical literature) deals with definitions of special terms. This part is dedicated to the theoretical knowledge and is also filled up with my own knowledge and experience from the field of the art-therapeutic praxes. It is actually a foundation for the second part of my diploma work that contains concrete techniques that can be used in supervision. The second part of my work contains a list of twenty creative techniques - methods of work - that are suitable for the individual, group or team supervision. All the techniques are proceeded according to one schema. It means that in all techniques are mentioned theirs goals, forms, technical severity, supposed time, procedure and also possibly risks. Le bateau ivre in Czech translation
This thesis is focused on the Drunken Boat poem by J. A. Rimbaud and its Czech translation. In particular, it focuses on the content of the poem and the meaning of specific images. It seeks an answer to a question to what extent does the preservation of poem's formal elements adversely affect its content and the vividness of its images. For this purpose, an original Czech translation of the poem is introduced, which is in turn analysed together with the other pre- existing translations. The study finds a parallel between Rimbaud's own path to freedom, leading from formal aspects to content, and his work, reflecting the same principles. For this reason, the formal aspects of the original poem and its Czech translations (e.g. ÄŚapek, HrubĂn, Neznal, Kadlec) do not represent the primary topic of this work. Instead, the study focuses on preserving the content of the poem and its new interpretations. Finally, the thesis also provides a review of the Czech translations of the poem
Simulation teaching of telephonic instruction of first aid provision
Bakalářská práce se zabĂ˝vá simulaÄŤnĂ vĂ˝ukou a jejĂm vyuĹľitĂm ve vzdÄ›lávánĂ nelĂ©kaĹ™skĂ˝ch zdravotnickĂ˝ch pracovnĂkĹŻ, konkrĂ©tnÄ› zdravotnickĂ˝ch záchranářů. SimulaÄŤnĂ vĂ˝uka je nedĂlnou součástĂ pĹ™Ăpravy studentĹŻ na jejich budoucĂ povolánĂ, jelikoĹľ mohou detekovat vlastnĂ problematickĂ© oblasti, kterĂ˝m se v praxi lze vyvarovat. V teoretickĂ© části je popsána historie simulaÄŤnĂ vĂ˝uky, jejĂ modernĂ pojetĂ a fáze. Dále jsou pĹ™iblĂĹľeny aspekty poskytovánĂ telefonicky asistovanĂ© prvnĂ pomoci a telefonicky asistovanĂ© neodkladnĂ© resuscitace. VĂ˝zkumná část se zabĂ˝vá vytvoĹ™enĂm scĂ©náře simulace a jeho ověřenĂm v praxi. Simulovaná situace se tĂ˝ká telefonicky asistovanĂ© prvnĂ pomoci pĹ™i aspiraci a následnĂ© telefonicky asistovanĂ© neodkladnĂ© resuscitace.This bachleor thesis deals with simulation teaching and its use in the education of non-medical health professionals, especially paramedics. Simulation teaching is an integral part of preparing students for their future careers, as they can detect their own problĂ©m areas, which can be avoided in practice. The teoretical part describes the history of simulation teaching, its modern concept and phases. Aspects of the provision of thelephone-assisted first aid and telephone assisted emergency resuscitation are also described. The research part deals with the creation of simulation scenario and its verification in practice. The simulated situation concerns telephone-assisted first aid for aspiration and subsequent telephone-assisted emergency resuscitation
The space in Horacio Quiroga's short stories
( in English) The aim of this Bachelor's thesis is to describe the category of space in Horacio Quiroga's short novel. Above all, it focuses on the creation of so-called jungle tales and its particular environment. The first part is theoretical and tries to sum up the most important facts of the author's life and creation and it is followed by the part summarizing various information about the theory of the concept of space in literature. A special chapter is dedicated to (the) a term "topos", which is appllied to the hispanoamerican literature and to the generation of regionalism, especially to the novel of the jungle. The work continues with the a practical analysis of the short stories. It describes the comparison of expressions of Quiroga's different styles and the image of the province of Misiones, where the author lived for a long time and which remains captured in his works. Furthermore, the thesis focuses on the influence of the environment of the jungle on the characters and what kind of characters it creates. It finishes with the image of the jungle as a sacred place
ÄŚeská literárnĂ moderna v ÄŤasopise Aus fremden Zungen — ÄŤtyĹ™i pĹ™Ăklady prostĹ™edkovánĂ // Czech literary modernism in the journal Aus fremden Zungen — four examples of mediation
The study focuses on Bronislav Wellek, Adolph Donath, Camill Hoffmann, Oskar Wiener and Otto
Hauser and their translations and articles on modern Czech literature published in the German
journal Aus fremden Zungen (1891–1910). This fortnightly established by Joseph Kürschner dealt
exclusively with foreign literature. Its openness to all kinds of literary currents and little-known
minor literatures was unique within the system of German literary journals. A comparison indicates
the key role played by mediators. The primary requirement for their activity was multilingualism,
and it turns out that in the case of all the figures under consideration, language did not necessarily
have the nationally symbolic significance ascribed by politics, and their identity basically did not
depend on their use of one language or another. The progress and success of this mediation was not
just decided by the literary standard of the translation, but all five translators were concerned to
improve the standard of translation, as well as to change the approach to translation in general. The
choice of text and its succeeded publication was determined by the involvement of the mediators in
the Czech and German literary context. Hence Bronislav Wellek focused primarily on translations
and interpretations of poetry by Jaroslav VrchlickĂ˝ and J. S. Machar, whom he knew personally and
who also introduced him to their own literary and intellectual circles. It was thanks to VrchlickĂ˝
that he printed his translations in Eduard Albert’s anthologies Poesie aus Böhmen (1893–1895).
Machar was responsible for other translations, e.g. Hilbert’s drama Vina, which Hermann Bahr was
to help promote in the German-language sphere. Between 1896 and 1898, Adolph Donath attempted
to build up his position as mediator between the ModernĂ revue circle and some representatives of
Viennese modernism (e.g. Karl Kraus, Peter Altenberg and Felix Rappaport). For Aus fremden Zungen
he translated an impressionistic short story by Luisa Ziková BĂlĂ˝ kvÄ›t (White Flower). In a fivepart
series and an accompanying study modern Czech prose (J. Zeyer, V. MrštĂk, F. X. Svoboda,
J. K. Šlejhar and R. Svobodová) was presented by Camill Hoffmann and Oskar Wiener. Their
mediation activities were based on direct relations with Prague Czech authors of the youngest
generation, on whom Hoffmann also focused in his articles for other German journals, though in
Aus fremden Zungen he was concerned with surveying the modernist work of the 1890s. Samples
of Czech prose were followed on by translations from modern Czech lyric poetry (O. Březina,
A. Sova, K. Hlaváček and V. Dyk) written by Otto Hauser. This polyglot and translator from numerous
languages was a different kind of Viennese mediator. In contrast to Wellek and Hoffmann, Hauser
was only in sporadic correspondence with Sova and Březina. His literary assessment of their
poetry was based on his broad knowledge of contemporary European poetry, including minor
literatures (such as Danish, Dutch and Croatian) and as such is of importance. These examples show
that literary mediation is a complex rapprochement of variously defined and construed contexts,
which have their reflected and unrealized limits
