    FORMULATION AND CHARACTERIZATION OF POLOXAMER 407 (R): THERMOREVERSIBLE GEL CONTAINING POLYMERIC MICROPARTICLES AND HYALURONIC ACID

    The influence of the composition and preparation method on the sol-gel transition temperature (Tsol-gel) and rheological response of poloxamer-based formulations was determined. Manual and more complex mechanical stirring were found to provide similar results. In addition, a linear dependence of Tsol-gel on the poloxamer content was observed in the range of concentrations analyzed, and a Poloxamer 407® concentration of 18% was selected. The addition of hyaluronic acid did not lead to significant changes in the Tsol-gel values. In contrast, the addition of microparticles caused a reduction in Tsol-gel without a significant reduction in gel strength, and pseudoplastic characteristics were observed, indicating that a thermoreversible gel was obtained with a rheology suitable for application in the treatment of burn wounds

    A mosca-branca Bemisia tabaci Biótipo B (Hemiptera: Aleyrodidae), é um herbívoro de difícil controle devido à alta plasticidade genotípica da espécie. No tomateiro pode causar danos severos principalmente pela transmissão de diversas viroses. O manejo do sistema de produção e o consórcio de culturas podem ter um efeito direto nas populações desse herbívoro, sem que seja necessária a aplicação de inseticidas. Avaliou-se a influência dos sistemas de produção orgânico e convencional e o consórcio tomate-coentro na dinâmica populacional da mosca-branca no campo experimental da Embrapa Hortaliças, de maio a setembro/06. O monitoramento dos adultos da mosca-branca e de seus inimigos naturais foi realizado utilizando-se armadilhas adesivas amarelas fixadas nas bordas e no interior das parcelas experimentais e a amostragem de ninfas foi realizada por observação direta das folhas de tomate no campo. Embora as populações ao redor dos diferentes tratamentos fossem equivalentes, a abundância de adultos de mosca-branca foi significativamente menor nas parcelas de tomate consorciado com coentro, tanto no sistema convencional como orgânico. Apenas o consórcio tomatecoentro em sistema orgânico apresentou redução significativa na quantidade de ninfas por planta em relação aos demais tratamentos. Os inimigos naturais foram significativamente mais abundantes em sistema orgânico e foi verificada uma correlação negativa da abundância dos inimigos naturais e a quantidade de ninfas por planta.     Cryptosporidium Priming Is More Effective than Vaccine for Protection against Cryptosporidiosis in a Murine Protein Malnutrition Model

    Cryptosporidium is a major cause of severe diarrhea, especially in malnourished children. Using a murine model of C. parvum oocyst challenge that recapitulates clinical features of severe cryptosporidiosis during malnutrition, we interrogated the effect of protein malnutrition (PM) on primary and secondary responses to C. parvum challenge, and tested the differential ability of mucosal priming strategies to overcome the PM-induced susceptibility. We determined that while PM fundamentally alters systemic and mucosal primary immune responses to Cryptosporidium, priming with C. parvum (106 oocysts) provides robust protective immunity against re-challenge despite ongoing PM. C. parvum priming restores mucosal Th1-type effectors (CD3+CD8+CD103+ T-cells) and cytokines (IFNγ, and IL12p40) that otherwise decrease with ongoing PM. Vaccination strategies with Cryptosporidium antigens expressed in the S. Typhi vector 908htr, however, do not enhance Th1-type responses to C. parvum challenge during PM, even though vaccination strongly boosts immunity in challenged fully nourished hosts. Remote non-specific exposures to the attenuated S. Typhi vector alone or the TLR9 agonist CpG ODN-1668 can partially attenuate C. parvum severity during PM, but neither as effectively as viable C. parvum priming. We conclude that although PM interferes with basal and vaccine-boosted immune responses to C. parvum, sustained reductions in disease severity are possible through mucosal activators of host defenses, and specifically C. parvum priming can elicit impressively robust Th1-type protective immunity despite ongoing protein malnutrition. These findings add insight into potential correlates of Cryptosporidium immunity and future vaccine strategies in malnourished children

    Nf2/Merlin controls spinal cord neural progenitor function in a Rac1/ErbB2-dependent manner

    Objective: Individuals with the neurofibromatosis type 2 (NF2) cancer predisposition syndrome develop spinal cord glial tumors (ependymomas) that likely originate from neural progenitor cells. Whereas many spinal ependymomas exhibit indolent behavior, the only treatment option for clinically symptomatic tumors is surgery. In this regard, medical therapies are unfortunately lacking due to an incomplete understanding of the critical growth control pathways that govern the function of spinal cord (SC) neural progenitor cells (NPCs). Methods: To identify potential therapeutic targets for these tumors, we leveraged primary mouse Nf2-deficient spinal cord neural progenitor cells. Results: We demonstrate that the Nf2 protein, merlin, negatively regulates spinal neural progenitor cell survival and glial differentiation in an ErbB2-dependent manner, and that NF2-associated spinal ependymomas exhibit increased ErbB2 activation. Moreover, we show that Nf2-deficient SC NPC ErbB2 activation results from Rac1-mediated ErbB2 retention at the plasma membrane. Significance: Collectively, these findings establish ErbB2 as a potential rational therapeutic target for NF2-associated spinal ependymoma
