Neuronen-Mikrosonde. Teilprojekt: Nervenzellkultur Abschlussbericht

Neuron microprobes should be developed as biohybrid systems (biological host cells integrated into technical microsystems): (1) to a medical objective, neuroprostheses for contacting of an injured peripheral nerve ('probe implant'), and (2) to a technical pharmacological objective, an in vitro testsystem for drug developmental screening using neuronal cells ('neurosubstrate'). (1) Flexible and ultra-light probe implants were micromechanically fabricated from polyimide in a three-dimensional shape which enables an atraumatic microsurgical adaption to the nerve stump. Biocompatibility was improved by physical surface modification. The implants were sterilized and delivered ready for implantation in animals and for the culture of host cells. (2) With the input from drug companies, a demonstrator system was designed for the incorporation of isolated neuronal cells. Their reactions to compounds can be monitored by electrical measurements using extracellular electrodes. This neurosubstrate was fabricated on the basis of silicon microtechnology. It contains micro-ringelectrodes around micro-holes onto which neuroblastoma cells can be positioned by controlled suction. This cell handling procedure did not affect cell viability. Mitosis of positioned cells was observed. Electrical activity of cells was recorded.
