Background and objectives: The development of mobile and on the edge appli-cations that embed deep convolutional neural models has the potential to revolutionisebiomedicine. However, most deep learning models require computational resourcesthat are not available in smartphones or edge devices; an issue that can be faced bymeans of compact models. The problem with such models is that they are, at leastusually, less accurate than bigger models. In this work, we study how this limitationcan be addressed with the application of semi-supervised learning techniques. Methods: We conduct several statistical analyses to compare performance of deepcompact architectures when trained using semi-supervised learning methods for tack-ling image classification tasks in the biomedical context. In particular, we explore threefamilies of compact networks, and two families of semi-supervised learning techniquesfor 10 biomedical tasks. Results: By combining semi-supervised learning methods with compact net-works, it is possible to obtain a similar performance to standard size networks. Ingeneral, the best results are obtained when combining data distillation with MixNet,and plain distillation with ResNet-18. Also, in general, NAS networks obtain betterresults than manually designed networks and quantized networks. Conclusions: The work presented in this paper shows the benefits of apply semi-supervised methods to compact networks; this allow us to create compact models thatare not only as accurate as standard size models, but also faster and lighter. Finally,we have developed a library that simplifies the construction of compact models usingsemi-supervised learning method

A PARTICIPAÇÃO CIDADÃ NOS PROCESSOS DE INTEGRAÇÃO REGIONAL: PELA EFETIVA CONSTRUÇÃO DE ESPAÇOS DEMOCRÁTICOS NAS RELAÇÕES INTERESTATAIS

    Get PDF

Dexamethasone-Loaded Lipomers: Development, Characterization, and Skin Biodistribution Studies

Follicular targeting has gained more attention in recent decades, due to the possibility of obtaining a depot effect in topical administration and its potential as a tool to treat hair follicle-related diseases. Lipid core ethyl cellulose lipomers were developed and optimized, following which characterization of their physicochemical properties was carried out. Dexamethasone was encapsulated in the lipomers (size, 115 nm; polydispersity, 0.24; zeta-potential (Z-potential), +30 mV) and their in vitro release profiles against dexamethasone in solution were investigated by vertical diffusion Franz cells. The skin biodistribution of the fluorescent-loaded lipomers was observed using confocal microscopy, demonstrating the accumulation of both lipomers and fluorochromes in the hair follicles of pig skin. To confirm this fact, immunofluorescence of the dexamethasone-loaded lipomers was carried out in pig hair follicles. The anti-inflammatory (via TNFα) efficacy of the dexamethasone-loaded lipomers was demonstrated in vitro in an HEK001 human keratinocytes cell culture and the in vitro cytotoxicity of the nanoformulation was investigated

Dexamethasone-loaded polymer hybrid nanoparticles were developed as a potential tool to treat alopecia areata due to their follicular targeting ability. Freeze drying (FD) is a common technique used to improve nanoparticle stability; however, there are few studies focused on its effect on ethyl cellulose lipid-core nanoparticles. Nanoparticles were lyophilized with different cryoprotectants. Sucrose was selected because it allowed for a good resuspension and provided acceptable physicochemical parameters (374.33 nm, +34.7 mV, polydispersion 0.229%, and 98.87% encapsulation efficiency). The nanoparticles obtained were loaded into a pleasant xanthan gum hydrogel, and the rheological, release, and skin permeation profiles of different formulations were studied. The FD formulation significantly modified the particle size, and the drug release and permeation properties were also altered. In addition, analyses of the cytotoxicity and anti-inflammatory efficacy of FD and non-FD particles on human keratinocytes indicated no differences

Secondary coenzyme Q10 deficiency triggers mitochondria degradation by mitophagy in MELAS fibroblasts

9 páginas.Mitochondrial encephalomyopathy, lactic acidosis, and stroke-like episodes (MELAS) is a mitochondrial disease most usually caused by point mutations in tRNA genes encoded by mtDNA. Here, we report on how this mutation affects mitochondrial function in primary fibroblast cultures established from 2 patients with MELAS who harbored the A3243G mutation. Both mitochondrial respiratory chain enzyme activities and coenzyme Q(10) (CoQ) levels were significantly decreased in MELAS fibroblasts. A similar decrease in mitochondrial membrane potential was found in intact MELAS fibroblasts. Mitochondrial dysfunction was associated with increased oxidative stress and the activation of mitochondrial permeability transition (MPT), which triggered the degradation of impaired mitochondria. Furthermore, we found defective autophagosome elimination in MELAS fibroblasts. Electron and fluorescence microscopy studies confirmed a massive degradation of mitochondria and accumulation of autophagosomes, suggesting mitophagy activation and deficient autophagic flux. Transmitochondrial cybrids harboring the A3243G mutation also showed CoQ deficiency and increased autophagy activity. All these abnormalities were partially restored by CoQ supplementation. Autophagy in MELAS fibroblasts was also abolished by treatment with antioxidants or cyclosporine, suggesting that both reactive oxygen species and MPT participate in this process. Furthermore, prevention of autophagy in MELAS fibroblasts resulted in apoptotic cell death, suggesting a protective role of autophagy in MELAS fibroblasts. This work was supported by Ministerio de Sanidad, Spain, grants FIS PI080500 and FIS EC08/00076, and by Asociación de Enfermos de Patología Mitocondrial (AEPMI) and Fundación Española de Enfermedades Lisosomales (FEEL).
