    Abundance and diversity of predominant sulfate-reducing bacteria in the gut of pufferfish

    Sulfate-reducing bacteria (SRB) exist in anaerobic environments, such as marine sediments, and produce hydrogen sulfide, which is toxic to marine animals. However, little is known about the ecology of SRB in the gut of fish. In the present study, we used molecular techniques to analyze the predominant SRB community in the gut of pufferfish inhabiting coastal areas of Japan. The density of the dissimilatory sulfite reductase alpha gene, dsrA, derived from SRB and total count of bacteria in guts of pufferfish was 4.4í—106 - 1.8í—107 copies/g and 3.5í—108 - 6.3í—109 cells/g, respectively, in all specimens. Clones of dsrA associated with Desulfobulbus oligotrophicus, a dominant SRB species, were detected in all 12 libraries, accounting for 57.7-94.3% of clones in each library. These results strongly suggest that SRB are indigenous bacteria in the gut of pufferfish and that hydrogen sulfide produced by SRB may be a risk factor for fish health

    CD153/CD30 signaling promotes age-dependent tertiary lymphoid tissue expansion and kidney injury

However, the cells and molecular pathways that govern TLT formation are poorly defined. Here we identify TNF superfamily CD153-CD30 signaling between two unique age-dependent lymphocyte subpopulations, CD153⁺PD-1⁺CD4⁺ senescence-associated T (SAT) cells and CD30+T-bet+ age-associated B cells (ABCs), as a driver for TLT expansion. SAT cells, which produced ABC-inducing factors IL21 and IFNγ, and ABCs progressively accumulated within TLTs in aged kidneys after injury. Notably, in kidney injury models, CD153 or CD30 deficiency impaired functional SAT cell induction, which resulted in reduced ABC numbers and attenuated TLT formation with improved inflammation, fibrosis and renal function. Attenuated TLT formation after transplantation of CD153-deficient bone marrow further supported the importance of CD153 in immune cells. Clonal analysis revealed that SAT cells and ABCs in the kidneys arose from both local differentiation and recruitment from the spleen. In the synovium of aged rheumatoid arthritis patients, T peripheral helper/T follicular helper cells and ABCs also expressed CD153 and CD30, respectively. Together, our data reveal a previously unappreciated function of CD153-CD30 signaling in TLT formation and propose targeting CD153-CD30 signaling pathway as a therapeutic target for slowing kidney disease progression

    Heterogeneous fibroblasts underlie age-dependent tertiary lymphoid tissues in the kidney

    Acute kidney injury (AKI) is a common clinical condition defined as a rapid decline in kidney function. AKI is a global health burden, estimated to cause 2 million deaths annually worldwide. Unlike AKI in the young, which is reversible, AKI in the elderly often leads to end-stage renal disease, and the mechanism that prevents kidney repair in the elderly is unclear. Here we demonstrate that aged but not young mice developed multiple tertiary lymphoid tissues (TLTs) in the kidney after AKI. TLT size was associated with impaired renal function and increased expression of proinflammatory cytokines and homeostatic chemokines, indicating a possible contribution of TLTs to sustained inflammation after injury. Notably, resident fibroblasts from a single lineage diversified into p75 neurotrophin receptor(+) (p75NTR(+)) fibroblasts and homeostatic chemokine-producing fibroblasts inside TLTs, and retinoic acid-producing fibroblasts around TLTs. Deletion of CD4(+) cells as well as late administration of dexamethasone abolished TLTs and improved renal outcomes. Importantly, aged but not young human kidneys also formed TLTs that had cellular and molecular components similar to those of mouse TLTs. Therefore, the inhibition of TLT formation may offer a novel therapeutic strategy for AKI in the elderly.</p

    Impact of Chronic Kidney Disease on the Presence and Severity of Aortic Stenosis in Patients at High Risk for Coronary Artery Disease

    <p>Abstract</p> <p>Objective</p> <p>We evaluated the impact of chronic kidney disease (CKD) on the presence and severity of aortic stenosis (AS) in patients at high risk for coronary artery disease (CAD).</p> <p>Methods</p> <p>One hundred and twenty consecutive patients who underwent invasive coronary angiography were enrolled. Aortic valve area (AVA) was calculated by the continuity equation using transthoracic echocardiography, and was normalized by body surface area (AVA index).</p> <p>Results</p> <p>Among all 120 patients, 78% had CAD, 55% had CKD (stage 3: 81%; stage 4: 19%), and 34% had AS (AVA < 2.0cm<sup>2</sup>). Patients with AS were older, more often female, and had a higher frequency of CKD than those without AS, but the prevalence of CAD and most other coexisting conventional risk factors was similar between patients with and without AS. Multivariate linear regression analysis indicated that only CKD and CAD were independent determinants of AVA index with standardized coefficients of -0.37 and -0.28, respectively. When patients were divided into 3 groups (group 1: absence of CKD and CAD, n = 16; group 2: presence of either CKD or CAD, n = 51; and group 3: presence of both CKD and CAD, n = 53), group 3 had the smallest AVA index (1.19 ± 0.30*# cm<sup>2</sup>/m<sup>2</sup>, *p < 0.05 vs. group 1: 1.65 ± 0.32 cm<sup>2</sup>/m<sup>2</sup>, and #p < 0.05 vs. group 2: 1.43 ± 0.29* cm<sup>2</sup>/m<sup>2</sup>) and the highest peak velocity across the aortic valve (1.53 ± 0.41*# m/sec; *p < 0.05 vs. group 1: 1.28 ± 0.29 m/sec, and #p < 0.05 vs. group 2: 1.35 ± 0.27 m/sec).</p> <p>Conclusion</p> <p>CKD, even pre-stage 5 CKD, has a more powerful impact on the presence and severity of AS than other conventional risk factors for atherosclerosis in patients at high risk for CAD.</p

    This report describes a case of hyperplasia of the mandibular coronoid process treated by coronoidotomy. A 26-year-old man visited our clinic complaining of restricted mouth opening. Panoramic radiography revealed marked hyperplasia of the right mandibular coronoid process. Three-dimensional CT showed that the coronoid process touched the temporal surface of the zygomatic bone at the maximal mouth opening position. His maximum mouth opening range was 27mm. The coronoid process was amputated about 8mm in width, and mouth opening training was done for a year after the surgery. Although the coronoid process and mandibular ramus reunited, the maximum mouth opening range was maintained at 40mm 13 months after the surgery
