    Water Relations in Berseem Cultivars Under Drought Stress

    Berseem clover (Trifolium alexandrinum L.) is a crop of particular interest in semi-arid regions, used for herbage and hay productions. However, its seed yield is reduced by drought stress that occurs during seed filling. In this study five cultivars (Axi, Big Bee, Lilibeo, Sacromonte and Saniros) subjected to four irrigation frequencies in controlled environment, were compared for their physiological responses to subsequent water stress period (3 days). Some parameters (leaf water potential (), osmotic potential (ĂŸ), relative water content (RWC) and gravimetric soil water content (GSWC)) related to plant water status were recorded. Plants subjected to previous periods of water stress were less sensitive to final drought stress than plants never stressed (control). Among cultivars examined, Lilibeo showed significantly higher values of , ĂŸ and RWC. Therefore, it seems to be the most tolerant cultivar for its ability to mantain high water status under water stress conditions

    Luciferase-based reporting of suicide gene activity in murine mesenchymal stem cells

Due to their ease of isolation, gene modification and tumor-homing properties, mesenchymal stem cells (MSCs) are an attractive cellular vehicle for the delivery of toxic suicide genes to a variety of cancers in pre-clinical models. In addition, the incorporation of suicide genes in stem cell-derived cell replacement therapies improves their safety profile by permitting graft destruction in the event of unexpected tumorigeneses or unwanted differentiation. Due to the functional requirement of ATP for the Firefly luciferase gene Luc2 to produce light, luciferase-based reporting of cytotoxicity can be engineered into potential cell therapies. Consequently, we nucleofected mammalian expression plasmids containing both the Luc2 and the yeast fusion cytosine deaminase uracil phosphoribosyltransferase (CDUPRT) genes for expression in murine MSCs to assess luciferase as a reporter of suicide gene cytotoxicity, and MSC as vehicles of suicide gene therapy. In vitro bioluminescence imaging (BLI) showed that following the addition of the non-toxic prodrug fluorocytosine (5-FC), CDUPRT-expressing MSCs displayed enhanced cytotoxicity in comparison to Luc2 reporter MSC controls.     Combined flow-based imaging assessment of optimal cardiac resynchronization therapy pacing vector: A case report

    Background: There are still many pendent issues about the effective evaluation of cardiac resynchronization therapy impact on functional mitral regurgitation. In order to reduce the intrinsic difficulties of quantification of functional mitral regurgitation itself, an automatic quantification of real-time three-dimensional full-volume color Doppler transthoracic echocardiography was proposed as a new, rapid, and accurate method for the assessment of functional mitral regurgitation severity. Recent studies suggested that images of left ventricle flow by echo-particle imaging velocimetry could be a useful marker of synchrony. Echo-particle imaging velocimetry has shown that regional anomalies of synchrony/synergy of the left ventricle are related to the alteration, reduction, or suppression of the physiological intracavitary pressure gradients. Case summary: We describe a case in which the two technologies are used in combination during acute echocardiographic optimization of left pacing vector in a 63-year-old man, Caucasian, who showed worsening heart failure symptoms a few days after an implant, and the effect of the device\u2019s optimization at 6-month follow-up. Discussion: The degree of realignment of hemodynamic forces, with quantitative analysis of the orientation of blood flow momentum (\u3c6), can represent improvement of fluid dynamics synchrony of the left ventricle, and explain, with a new deterministic parameter, the effects of cardiac resynchronization therapy on functional mitral regurgitation. Realtime three-dimensional color flow Doppler quantification is feasible and accurate for measurement of mitral inflow, left ventricular outflow stroke volumes, and functional mitral regurgitation severity. Conclusion: This clinical case offers an innovative and accurate approach for acute echocardiographic optimization of left pacing vector. It shows clinical utility of combined three-dimensional full-volume color Doppler transthoracic echocardiography/echo-particle imaging velocimetry assessment to increase response to cardiac resynchronization therapy, in terms of reduction of functional mitral regurgitation, improving fluid dynamics synchrony of the left ventricle

    Restrictions on Access to Social Protection by New Southern European Migrants in Belgium

    peer reviewedDespite the fact that the global financial crisis has affected Belgium’s economic performance, the country has continued to be an attractive destination for migrants. Recently, however, there has been a dramatic change in migrant flows to Belgium, as a result of two phenomena. First and foremost, the country has experienced large inflows of post-accession migration from Central and Eastern European citizens. Second, 40 years after the end of the guest worker programmes, the economic crisis reactivated migration from Southern Europe. One important consequence of the economic crisis and the arrival of Central and Eastern European migrants has been the growing scepticism that has developed in Belgium towards the freedom of circulation. This has led the authorities to implement specific policies aimed at discouraging further migration of low-skilled EU migrants, who are deemed undesirable. In this chapter, we analyse a specific policy consisting of the removal of residence permits from EU jobseekers who claim social security benefits in Belgium. This policy has affected both Central and Eastern European as well as new Southern European EU migrants.Examining the mobilization of different organizations, we then show that—while they are numerically fewer—new Southern European immigrants are in a better position than other new immigrants in Belgium to challenge receiving country policies that target them

    Eurocity London: a qualitative comparison of graduate migration from Germany, Italy and Latvia

    This paper compares the motivations and characteristics of the recent migration to London of young-adult graduates from Germany, Italy and Latvia. Conceptually the paper links three domains: the theory of core–periphery structures within Europe; the notion of London as both a global city and a ‘Eurocity’; and the trope of ‘crisis’. The dataset analysed consists of 95 in-depth biographical interviews and the paper’s main objective is to tease out the narrative similarities and differences between the three groups interviewed. Each of the three nationalities represents a different geo-economic positioning within Europe. German graduates move from one economically prosperous country to another; they traverse shallow economic and cultural boundaries. Italian graduates migrate from a relatively peripheral Southern European country where, especially in Southern Italy, employment and career prospects have long been difficult, and have become more so in the wake of the financial crisis. They find employment opportunities in London which are unavailable to them in Italy. Latvian graduates are from a different European periphery, the Eastern one, post-socialist and post-Soviet. Like the Italians, their moves are economically driven whereas, for the Germans, migration is more related to lifestyle and life-stage. For all three groups, the chance to live in a large, multicultural, cosmopolitan city is a great attraction. And for all groups, thoughts about the future are marked by uncertainty and ambiguity

    Prodrug converting enzyme gene delivery by L. monocytogenes

    <p>Abstract</p> <p>Background</p> <p><it>Listeria monocytogenes </it>is a highly versatile bacterial carrier system for introducing protein, DNA and RNA into mammalian cells. The delivery of tumor antigens with the help of this carrier into tumor-bearing animals has been successfully carried out previously and it was recently reported that <it>L. monocytogenes </it>is able to colonize and replicate within solid tumors after local or even systemic injection.</p> <p>Methods</p> <p>Here we report on the delivery of two prodrug converting enzymes, purine-deoxynucleoside phosphorylase (PNP) and a fusion protein consisting of yeast cytosine deaminase and uracil phosphoribosyl transferase (FCU1) into cancer cells in culture by <it>L. monocytogenes</it>. Transfer of the prodrug converting enzymes was achieved by bacterium mediated transfer of eukaryotic expression plasmids or by secretion of the proteins directly into the host cell cytosol by the infecting bacteria.</p> <p>Results</p> <p>The results indicate that conversion of appropriate prodrugs to toxic drugs in the cancer cells occured after both procedures although <it>L. monocytogenes</it>-mediated bactofection proved to be more efficient than enzyme secretion 4T1, B16 and COS-1 tumor cells. Exchanging the constitutively P<sub>CMV</sub>-promoter with the melanoma specific P<sub>4xTETP</sub>-promoter resulted in melanoma cell-specific expression of the prodrug converting enzymes but reduced the efficiencies.</p> <p>Conclusion</p> <p>These experiments open the way for bacterium mediated tumor specific activation of prodrugs in live animals with tumors.</p
