    Heterogeneity and evolution of the Miocene Iceland mantle plume

    The North Atlantic Igneous Province (NAIP) is unique in that it presents a complete magmatic history from ~62 Ma to today. This provides an unrivalled opportunity to study the geochemical and thermal evolution of a mantle plume through time. Despite the wealth of geochemical data available for Iceland, our knowledge of mantle heterogeneity beneath Iceland during the mid-Miocene remains sketchy. The oldest basalts (15-16 Ma) on Iceland are preserved in several ~1 km thick sequences in Vestfirdir in Northwest Iceland. Each sequence contains a hiatus in volcanism, marked by a laterite-lignite horizon, that is likely the result of the relocation of a spreading axis. The basalt geochemistry from above and below this horizon provide an insight into the heterogeneity of the Iceland mantle plume during the mid-Miocene. The Vestfirdir basalts are also characterised by the highest 3He/4He (~40 Ra) on Iceland requiring the involvement of deep mantle. Incompatible trace element and Nd-Sr-Pb isotope data indicate that there are two enriched components (NWE1 and NWE2) and one depleted component (NWD1) within the mantle beneath Iceland during the Miocene. Both enriched components have low 143Nd/144Nd and high 87Sr/86Sr, that are comparable with the most enriched modern Iceland basalts. The two components are differentiated by the relative enrichment in the more incompatible trace elements and their Pb-isotope compositions. Component NWE1 is similar to the modern Iceland mantle plume composition, while NWE2 is closer in composition to modern depleted Iceland rift-zone basalts. The depleted component identified in the Vestfirdir basalts (NWD1) is distinct from modern North Atlantic N-MORB and the depleted component in the neovolcanic rift zones by, for instance, higher incompatible trace element concentrations and a more negative ∆207Pb composition. These observations allow the identification of a depleted mantle component present at 15-16 Ma that is no longer sampled in the modern Iceland plume setting and argue for a distinct compositional change in the mantle beneath Iceland over the last 15-16 Myr. Existing and new 3He/4He and magmatic temperature measurements help understand how primordial helium and heat in mantle plumes vary with time and provide insight into the nature of the deep mantle and the source of primordial terrestrial volatiles. The highest 3He/4He (42 RA) recorded in Vestfirdir is midway between the starting plume (50 RA) and modern Iceland values (34 RA), demonstrating that 3He/4He appears to have steadily decreased with time. The high-3He/4He Vestfirdir basalts have a large range of incompatible trace element and Sr-Nd-Pb isotope ratios, as also seen in the PIP picrites, implying that there is no compositionally-unique mantle source for the high 3He/4He component in the Iceland plume. Additionally, the uncontaminated Vestfirdir basalts that haven’t been erupted through continental crust also have Pb-isotope values that rule out the need for a primordial, isolated mantle reservoir formed ~4.55-4.45 Ga. The new Al-in-olivine data from high-3He/4He Vestfirdir basalts yield crystallisation temperatures significantly less than that of proto-Iceland plume and modern Iceland picrites. The apparent ~40°C increase of the Iceland plume temperature in the last 15 million years contrasts strongly with the decrease of 3He/4He. This suggests that the heat and primordial 3He are decoupled in the Iceland plume. This may reflect the different sources (i.e. heat from the core, 3He from the deep mantle) or alternatively different diffusion rates of heat and helium across the core-mantle boundary

    The impact of a single round of community mass treatment with azithromycin on disease severity and ocular Chlamydia trachomatis load in treatment-naĂŻve trachoma-endemic island communities in West Africa.

    BACKGROUND: Trachoma, a neglected tropical disease, is caused by ocular infection with Chlamydia trachomatis (Ct). The World Health Organization (WHO) recommends three annual rounds of community mass drug treatment with azithromycin (MDA) if the prevalence of follicular trachoma in 1-9 year olds (TF1-9) exceeds 10% at district level to achieve an elimination target of district-level TF1-9 below 5% after. To evaluate this strategy in treatment-naĂŻve trachoma-endemic island communities in Guinea Bissau, we conducted a cross-sectional population-based trachoma survey on four islands. The upper tarsal conjunctivae of each participant were clinically assessed for trachoma and conjunctival swabs were obtained (n = 1507). We used a droplet digital PCR assay to detect Ct infection and estimate bacterial load. We visited the same households during a second cross-sectional survey and repeated the ocular examination and obtained conjunctival swabs from these households one year after MDA (n = 1029). RESULTS: Pre-MDA TF1-9 was 22.0% (136/618). Overall Ct infection prevalence (CtI) was 18.6% (25.4% in 1-9 year olds). Post-MDA (estimated coverage 70%), TF1-9 and CtI were significantly reduced (7.4% (29/394, P < 0.001) and 3.3% (34/1029, P < 0.001) (6.6% in 1-9 year olds, P < 0.001), respectively. Median ocular Ct load was reduced from 2038 to 384 copies/swab (P < 0.001). Following MDA cases of Ct infection were highly clustered (Moran's I 0.27, P < 0.001), with fewer clusters of Ct infection overall, fewer clusters of cases with high load infections and less severe disease. CONCLUSIONS: Despite a significant reduction in the number of clusters of Ct infection, mean Ct load, disease severity and presence of clusters of cases of high load Ct infection suggesting the beginning of trachoma control in isolated island communities, following a single round of MDA we demonstrate that transmission is still ongoing. These detailed data are useful in understanding the epidemiology of ocular Ct infection in the context of MDA and the tools employed may have utility in determining trachoma elimination and surveillance activities in similar settings

    Carbon capture and storage at the end of a lost decade

    Following the landmark 2015 United Nations Paris Agreement, a growing number of countries are committing to the transition to net-zero emissions. Carbon capture and storage (CCS) has been consistently heralded to directly address emissions from the energy and industrial sectors and forms a significant component of plans to reach net-zero. However, despite the critical importance of the technology and substantial research and development to date, CCS deployment has been slow. This review examines deployment efforts over the last decade. We reveal that facility deployment must increase dramatically from current levels, and much work remains to maximize storage of CO2 in vast subsurface reserves. Using current rates of deployment, CO2 storage capacity by 2050 is projected to be around 700 million tons per year, just 10% of what is required. Meeting the net-zero targets via CCS ambitions seems unlikely unless worldwide coordinated efforts and rapid changes in policy take place

    The liminality of training spaces: Places of private/public transitions

    This paper draws upon research, conducted for the London West Learning and Skills Council, on the training experiences of women with dependent children. One of the striking revelations of the research, we suggest, is the way in which training spaces are used and perceived by women, which are often at odds with government intentions. To help make sense of women’s use of and motivation for training we utilise the concept of ‘liminality’ and the private/public imbrication to explain the ways in which women use, or are discouraged from using, training spaces. Further, how the varied and multiple uses women in our research have put training to in their own lives has encouraged us to rethink the relationship between the private and the public more generally. In the light of this, we suggest that training and the places in which training take place, have been neglected processes and spaces within feminist geography and might usefully be explored further to add to an extensive literature on women’s caring and domestic roles and their role in the paid workplace

    On-orbit servicing commercial opportunities with security implications

    The On-Orbit Servicing (OOS) working group discussed legal and political implications of developing a commercial OOS industry. The group considered the benefits that OOS and Active Debris Removal (ADR) can offer the satellite industry, as well as potential disadvantages for international relations between space faring nations. To gain an accurate perspective of stakeholders involved in such a process, the OOS working group held a mock hearing for OOS licensing, with members of the working group assigned to represent stakeholders. Working group members presented their cases at a simulated domestic regulatory panel, constructed of members representing various government ministers, to fully explore stakeholder views. The mock hearings explored the challenges faced by OOS and ADR entrepreneurs as well as the benefit of regulation. The groups highlighted recommendations to ensure the practicality of OOS and determine how best to encourage licensing and regulation of such activities, as summarised below. 1. The United Nations (UN) should provide regulatory guidelines for OOS and ADR. 2. Government agencies should license OOS. The Federal Aviation Administration (FAA) has taken responsibility for licensing commercial space transportation in the United States and this should be extended to OOS/ADR missions to enable short-term advancement prior to further UN regulation. 3. Government should support OOS and ADR development to ensure continued demand. This includes leading by example on government satellites and potential launch levies to enable on-going ADR funding. 4. All stakeholders should prevent weaponisation of space through transparency of operations. 5. Nations should initiate international cooperation on ADR. OOS and ADR will ensure sustainable use of satellites, particularly in LEO and GEO, for the coming decades. It is through transparency, economic stimulation and close monitoring that such endeavours will be successful

    Orbital controls on eastern African hydroclimate in the Pleistocene

    Understanding eastern African paleoclimate is critical for contextualizing early human evolution, adaptation, and dispersal, yet Pleistocene climate of this region and its governing mechanisms remain poorly understood due to the lack of long, orbitally-resolved, terrestrial paleoclimate records. Here we present leaf wax hydrogen isotope records of rainfall from paleolake sediment cores from key time windows that resolve long-term trends, variations, and high-latitude effects on tropical African precipitation. Eastern African rainfall was dominantly controlled by variations in low-latitude summer insolation during most of the early and middle Pleistocene, with little evidence that glacial–interglacial cycles impacted rainfall until the late Pleistocene. We observe the influence of high-latitude-driven climate processes emerging from the last interglacial (Marine Isotope Stage 5) to the present, an interval when glacial–interglacial cycles were strong and insolation forcing was weak. Our results demonstrate a variable response of eastern African rainfall to low-latitude insolation forcing and high-latitude-driven climate change, likely related to the relative strengths of these forcings through time and a threshold in monsoon sensitivity. We observe little difference in mean rainfall between the early, middle, and late Pleistocene, which suggests that orbitally-driven climate variations likely played a more significant role than gradual change in the relationship between early humans and their environment

    Daratumumab, Cyclophosphamide, Bortezomib, Lenalidomide, and Dexamethasone as Induction and Extended Consolidation Improves Outcome in Ultra-High-Risk Multiple Myeloma

    Purpose: The multicenter OPTIMUM (MUKnine) phase II trial investigated daratumumab, low-dose cyclophosphamide, lenalidomide, bortezomib, and dexamethasone (Dara-CVRd) before and after autologous stem-cell transplant (ASCT) in newly diagnosed patients with molecularly defined ultra–high-risk (UHiR) multiple myeloma (NDMM) or plasma cell leukemia (PCL). To provide clinical context, progression-free survival (PFS) and overall survival (OS) were referenced to contemporaneous outcomes seen in patients with UHiR NDMM treated in the recent Myeloma XI (MyeXI) trial.Methods: Transplant-eligible all-comers NDMM patients were profiled for UHiR disease, defined by presence of ≄2 genetic risk markers t(4;14)/t(14;16)/t(14;20), del(1p), gain(1q), and del(17p), and/or SKY92 gene expression risk signature. Patients with UHiR MM/PCL were offered treatment with Dara-CVRd induction, V-augmented ASCT, extended Dara-VR(d) consolidation, and Dara-R maintenance. UHiR patients treated in MyeXI with carfilzomib, lenalidomide, dexamethasone, and cyclophosphamide, or lenalidomide, dexamethasone, and cyclophosphamide, ASCT, and R maintenance or observation were identified by mirrored molecular screening. OPTIMUM PFS at 18 months (PFS18m) was compared against MyeXI using a Bayesian framework, and patients were followed up to the end of consolidation for PFS and OS.Results: Of 412 screened NDMM OPTIMUM patients, 103 were identified as UHiR or PCL and subsequently treated on trial with Dara-CVRd; 117 MyeXI patients identified as UHiR formed the external comparator arm, with comparable clinical and molecular characteristics to OPTIMUM. Comparison of PFS18m per Bayesian framework resulted in a 99.5% chance of OPTIMUM being superior to MyeXI. At 30 months' follow-up, PFS was 77% for OPTIMUM versus 39.8% for MyeXI, and OS 83.5% versus 73.5%, respectively. Extended post-ASCT Dara-VRd consolidation therapy was highly deliverable, with limited toxicity.Conclusion: Our results suggest that Dara-CVRd induction and extended post-ASCT Dara-VRd consolidation markedly improve PFS for UHiR NDMM patients over conventional management, supporting further evaluation of this strategy.</p
